A safe, effective and economical vaccine is required for the prevention of Japanese encephalitis (JE), a disease with high mortality and grave sequelae, which is prevalent in Japan and other countries in east, south-east and southern Asia. As the initial step to produce a second-generation vaccine, recombinant DNA technology was utilized to express the JE virus envelope glycoprotein V3 (E) gene in yeast cells.This report describes the construction of a yeast expression vector in which a cDNA clone covering the V3 gene was connected to the acid-phosphatase promoter of a yeast vector plasmid. Successful expression of the V3 gene was detected by ELISA and Western blotting using monoclonal antibodies against JE V3. Immunization of mice with the V3 antigen expressed in yeast produced limited but definite levels of anti-JE antibodies which could neutralize JE virus. The results are an encouraging step in the development of a practical second-generation JE vaccine.
The synthesis of some amidic derivatives, obtained by condensation of various 5-substituted indoleglyoxylchlorides with physiologically important amines as tyramine, tryptamine and 5-aminouracil, is described. The preparation of one glycolyl derivative is also reported. Study of analgesic and antiinflammatory properties has shown only a feeble activity of these compounds.
The 5' region of the Japanese encephalitis virus (JEV) RNA was cloned and 3000 nucleotides (nt) were determined by sequencing DNA complementary to viral RNA, and genomic RNA, using oligodeoxynucleotide primers and the dideoxy chain-termination reaction. Comparison of the nt sequence and the reduced amino-acid sequence of JEV with those of other flaviviruses showed significant homologies, which allowed locations to be assigned for three structural proteins.
Two cohorts of Japanese children were studied longitudinally at 3-year intervals, at ages 6 and 9 years in cohort 1 and 9 and 12 years in cohort 2. There were 252 children in cohort 1 and 235 in cohort 2. The examinations performed were blood pressure, height, weight, skin fold thickness, upper arm length and circumference, ECG, VCG, M-mode echocardiography, serum cholesterol, hemoglobin and urinalysis. The subjects were divided into 5 quintile groups by their systolic blood pressure (SBP) percentiles calculated at each examination. SBP was tracked in each cohort throughout the follow-up period. Percentages remained in the highest SBP quintile group at both examinations were 43.5% (boys) and 59.1% (girls) in cohort 1, and 25.0% (boys) and 56.5% (girls) in cohort 2. The tracking of SBP was defined by body mass (height and weight) in most children. In some children, however, it was not defined by body mass tracking. Left ventricular muscle volume and its index were large in boys in the highest SBP tracking group. This indicates that the cardiac muscle is hypertrophied already, before the development of hypertension.
Mass screening by ECG for heart diseases in pupils and students was performed in Shimane Prefecture; all the children had primary screening and were under the supervision of a pediatric cardiologist in all phases, from primary to precise (tertiary) examination. The precise examination was performed with 2D echo, exercise ECG and/or cardiac catheterization when indicated, and an important feature of this mass screening was the ability to carry out an accurate follow-up in all the phases from primary to precise examination. The number of subjects in the primary screening from 1980 to 1984 was 50758 primary school pupils, 44216 junior high school students and 33480 senior high school students; organic heart diseases not under the supervision of the pediatric cardiologist at the time of mass screening were found in 22 primary school pupils (0.04% of participants in the primary screening), 14 junior high school students (0.03%) and 5 senior high school students (0.01%). Clinically significant arrhythmias without underlying organic heart diseases were discovered by mass screening in 8 primary school pupils (0.02%), 13 junior high school students (0.02%) and 16 senior high school students (0.05%).
Changes in hemodynamic parameters following "shaking" in 4, 12, 24 and 48 week old spontaneously hypertensive rats (SHR) and Wistar rats of the Kyoto strain (WKY) were analyzed. Mean arterial pressure (MAP) and cardiac output (CO) were measured in the following two conditions: (A) at rest, and (B) after shaking (these parameters were measured following a 3-sec rest after 5-sec of shaking). Both sets of data were compared in each age group between SHR and WKY. Heart rates (HR) in all age groups of SHR and WKY were significantly increased after shaking compared with those at rest. MAP in 4 week old SHR, but not in 4 week old WKY, was increased compared with that at rest. In 48 week old SHR and WKY, MAP after shaking was not increased compared with that at rest. This increased MAP induced by stress (shaking) in young SHR may be related to a causative factor in hypertension. CO in SHR did not indicate changes in shaking reaction as a result of increasing aging. Heart work index (HWI) after shaking in all age groups of SHR, including the prehypertensive stage, was significantly increased compared with that at rest; this was not the case in WKY. The intermittently increased heart work seen in response to stress may contribute to the early development of left ventricular hypertrophy in young SHR.
Several 1,8-naphthyridine derivatives have been synthesized and pharmacologically investigated. Some of them exhibited a marked antihypertensive activity on spontaneously hypertensive rats.
Some 3-amino- and 3-hydroxy-8-halosubstituted 6H-indolo[2,3-b] [1,8]naphthyridines were synthesized and tested for their affinity for the benzodiazepine receptor in bovine cortical membranes. All prepared compounds were more active than the corresponding 8-unsubstituted derivatives. Moreover, among these compounds the 8-chloroindolonaphthyridines were clearly the most potent.

Several 4H-pyrido[1,2-a]pyrimidine derivatives with a basic substituent in the 2-position have been synthesized. All the compounds tested showed no appreciable antihypertensive activity.

Using the 4-Lead Set Japanese Pediatric ECG Screening Criteria Proposal, the effects of fatness on the electrocardiograms were analyzed in 1025 junior high school children, aged 12-15 years (of whom 114 were obese). As the results show, significant differences were detected by the 1% two-sided test between the averages of the nonfat group and the middle- and high-fat groups with regard to S amplitude in leads V1-V3 for boys and girls, R amplitude in leads V4 and V5, SV1 + RV6, SV1 + RV5, and SV3 + RV3 for boys, and S amplitude in leads V4-V6 for girls. High R amplitudes were observed in leads V3 and V4 for the middle- and high-fat group of girls. When applying conventional ECG criteria for left ventricular hypertrophy to the middle- and high-fat groups of boys, the value of SV1 + RV6 greater than or equal to 4.5 mV instead of SV1 + RV6 greater than or equal to 5.0 mV is recommended.
