[The clinical analysis of corticosteroid ocular hypertension and corticosteroid glaucoma after photorefractive keratectomy].

OBJECTIVE: To inquire into the incidence, clinical characteristics and therapeutic effects of corticosteroid hypertension and corticosteroid glaucoma after photorefractive keratectomy (PRK). METHODS: The authors completed a follow up study on 1 590 eyes on which this procedure was performed and corticosteroid (0.1% fluoromethalone, FML) eye drops were applied 4 to 6 months post-operatively to observe intraocular pressure at 1, 3, 6, 12 and 24 months, and treated the eyes with increase of intraocular pressure. RESULTS: Corticosteroid ocular hypertension or corticosteroid glaucoma occurred in 2.70% of 1 590 eyes. The intraocular pressure of the eyes was recovered to normal in all patients after treatments of stopping FML eye drops, using topical 0.5% timolol (or 0.5% levobunolol hydrochloride), taking diamox orally or receiving trabeculectomy. CONCLUSIONS: Topical application of corticosteroid may cause corticosteroid hypertension and corticosteroid glaucoma after PRK, and visual function damage may occur in a few eyes. Under this situation, the corticosteroid eye drops should be stopped and glaucomatous treatment should be given.

[Studies on changes in levels of hormone and expression of ob gene during induction of obesity in rats].

OBJECTIVE: To study the rules of changes in ob mRNA and its relation to hormone level in vivo in rats during their dynamic process to obesity induced by high-caloric fodder. METHODS: Level of ob mRNA in adipose tissues of rats was determined with dot-hybridization techniques, and levels of serum insulin and growth hormone were measured with radioimmunoassay. RESULTS: Level of ob mRNA in the fatty tissues and serum insulin level increased, but serum level of growth hormone decreased, with the increase in body weight of rats. CONCLUSION: Increase in expression of ob mRNA could be the result, but not the cause, of obesity. Changes in the levels of ob mRNA, serum insulin and growth hormone indicated that they could affect each other and could be regulated by common metabolic signals.

[The effect of defibrase in prevention of experimental atherosclerosis in rabbits].

OBJECTIVE: To study the anti-atherosclerosis effect of difibrase in rabbits. METHODS: Use of computer, light microscope, electron microscope, biochemistry and histochemical methods to identify the total cholesterol (TC) content, mean plaque area in aorta and its cholesterol content, degree of stenosis of coronary arteries of the experimental animals and control animals. RESULTS: Defibrase could decrease the mean plaque area of aorta and the TC content in the experimental animal group was less than that in the control group (P < 0.01). The findings in aortic arches and thoracic aorta were similar to those in the aorta (P < 0.05, P < 0.01). The number of plaques at the small artery openings in the thoracic aorta and abdominal aorta of the experimental group were less than that of the control group (P < 0.01). The cross sections made at the upper third of all hearts showed that the degree of stenosis of coronary artery branches in the heart wall of the experimental group was much less than that of the control group. CONCLUSION: Defibrase can significantly inhibit atherosclerosis.

[A clinico-pathological study of granulomatous amoebic encephalitis].

OBJECTIVE: To study the clinico-pathological characteristics and differential diagnosis of granulomatous meningioencephalitis caused by acanthamoeba. METHODS: The clinical features, CT scan findings and post mortem pathological changes of 3 patients afflicted with granulomatous amoebic encephalitis (GAE) were analysed. RESULTS: GAE is a chronic clinical entity. Clinical manifestations include headache, low fever, seizures, hemiparesis and coma leading to death. CSF: pressure slightly elevated with presence of lymphocytes and plasma cells, glucose usually low with moderately high protein levels. CT scans were non specific. Clinical course insidious and may mimic bacterial meningitis or tuberculous meningitis. Post mortem pathological examinations showed that acanthamoeba produced a chronic or subacute granulomatous meningioencephalitis. Acanthamoebic trophoziotes and cysts were found within the hemorrhagic necrotic cerebral tissues. In some cases there is severe angitis and fibrinoid necrosis of the vascular wall with perivascular cuffing by lymphocytes and plasma cells. Trophozoites and cysts were found within the perivascular space. CONCLUSIONS: Acanthamoeba is an opportunistic pathogenic free living amoeba which can produce fatal GAE.

[A clinico-pathological and immunohistochemical study on anaplastic large cell lymphoma].

OBJECTIVE: To study the clinicopathological characteristics, immunophenotype and biologic behavior of anaplastic large cell lymphoma (ALCL). METHODS: Clinicopathological and immunohistochemical methods (ABC method) were used to analyse 8 cases of ALCL and followed for 6 to 25 months. RESULTS: Eight ALCL patients (4 male and 4 female) accounted for 4.1% of the 195 lymphomas seen from 1994 to 1997. Average age was 40 years. Two tumors were located in the skin, the remainder were systemic. Clinical stage: stage I, 2 cases; stage II, 1 case; stage III, 5 cases. Histopathology: Tumor cells exhibited high pleomorphism and a large amount of mitotic figures. Reactive components (cells) often observed in the background. Immunohistochemical studies found that all tumors reacted positively with CD30 and negatively with CD15. Five cases expressed T-cell's markers (CD3, UCHL-1, CD43) and three expressed B-cell markers (CD79a, L26). After chemotherapy, two patients with cutaneous type ALCL had complete remission and survived 14 and 25 months respectively. Of the six systemic cases, three survived for 6-8 months, the remaining three died at 2 days, 20 days and 18 months after diagnosis. CONCLUSIONS: This study found that ALCL was a group of lymphomas showing similarity in morphology but diversity in clinical presentation and immunophenotype. Even if the tumor is very responsive to chemotherapy, ALCL remains a high grade malignant lymphoma.

[PCR for the detection of 16S rRNA gene bacteria].

According to the high conservative region of 16S rRNA gene in bacteria, PCR primers of the broad-range bacteria, gram-positive bacteria and gram-negative bacteria were synthesized to detect 13 bacterium species and (40) clinical specimens. All the tested bacterium species were positive. The lowest concentration of Escherichia Coli detected by serial dilution was 4 CFU.ml-1. The positive rate of PCR (27/40) was higher than that of bacterium culture(18/40). The results indicate that these PCR primers possess high specificity and sensitivity in identifying 16S rRNA gene of bacteria.

[Specific cellular and humoral immune responses induced by intramuscular injection of DNA vaccine containing HBV HBsAg gene in mice].

To know the influences on the function of cellular and humoral immunity in mice by DNA vaccine containing HBV HBsAg gene, the DNA vaccine, pcDNA3-HBsAg, in which the HBsAg gene sequences of HBV were inserted into downstream of CMV promoter in pcDNA3, were injected intramuscularly into Balb/C mice. The results were that the serum anti-HBsIgG was detected in all of the pcDNA3-HBsAg immunized mice, but none in control mice; serum levels of Th1 cytokines, interleukine 2, and interferon gamma, were significantly higher in pcDNA3-HBsAg immunized mice than these in controls. It is indicated that the good responses of cellular and humoral immunity can be induced by injection of pcDNA3-HBsAg in Balb/C mice.

[Application of intracoronary ultrasound in directional coronary atherectomy].

Intracoronary ultrasound(ICUS) was successfully applied in 32 patients who accepted directional coronary atherectomy(DCA). After the procedure, the mean internal diameter of coronary artery increased from 1.06 mm to 3.59 mm, and the residual stenosis was < 25%. No severe complications, such as rupture of perforation, occurred. The clinical application of ICUS in DCA may improve the therapeutic effects as well as success rate of DCA, and may minimize the incidence of complications.

[Skeletal effect of xianzhen gubao on preventing prednisone-induced osteoporosis in male rats].

OBJECTIVE: To study the effect of Chinese medicine Xianzhengubao (XZGB) capsules of compound prescription on corticosteroid-induced osteoporosis in rats. METHOD: 3-month-old SD male rats were randomly divided into basal control and aging control, prednisone (4.5 mg.kg-1, ig, twice a week) and XZGB capsules were used in three different doses (0.2, 0.4, 0.8 g.kg-1.d-1 ig)6 times a week. All the rats were killed. On the 90th day afteradministration. The proximal tibiae were processed in undecalcified sections at a thickness of 4-10 microns for histomorphometric analysis. RESULT: Compared with the aging controls corticosteroid reduced markedly the trabecular bone mass due to increase of bone resorption and decrease of bone formation (%Tb. Ar 52.9%). Treatment with XZGB capsules in high doses increased significantly the trabecular area as compared with the hormone control(% Tb. Ar + 91.2%). The protective mechanism of XZGB capsules mainly consists in suppressing bone resorption and mildly increasing bone formation in rats. CONCLUSION: Xianzhen gubao capsules have an effect on preventing bone loss in rats induced by corticosteroid, and higher doses produce greater effect.

[Expression of heat shock protein (HSP70) in normal endometrium and endometrial carcinomas].

Expression of heat shock protein 70 (HSP70) in normal endometrium tissue and endometrial carcinoma was analyzed by the present authors to determine the relationship between HSP70 expression and the prognosis of endometrial carcinomas. The immunohistochemical reactivity of monoclonal antibodies against HSP70 was examined in 44 endometrial carcinomas and 40 normal endometria HSP70 respectively. HSP70 was expressed stronger in carcinomas than in normal. Expression of HSP70 was found in 56.82% of endometrial carcinomas and was well correlated with histological grade (P < 0.05). These results suggest that HSP70 may play a role in the prognosis of endometrial carcinomas.

[Cloning and sequencing analysis of rice stripe virus genome segment 4 of Chinese isolate Y].

The cDNA fragment covering full-length sequence of RStV RNA4 of Yunnan isolate in China was obtained by RT-PCR. The PCR-derived fragment was then cloned into vector pCRII. The cloned cDNA was sequenced. Comparison of the nucleotide and deduced amino acid sequences with those of the Japanese isolate T was made. The results showed that at the nucleotides level, vORF, vcORF and the intergenic region had 94.9%, 94.1% and 86.1% identity respectively, the 5'-untranslational region was exactly the same as that of Japanese isolate T, while the 3'-terminal sequence had 96.1% identity, differing by two nucleotides; at the amino acid level, vORF and vcORF had 99.4% and 98.3% identity respectively. Therefore, as well as being exactly the same size for the two isolates, the amino acid sequences of the coding regions and the 5'- and 3'-terminal sequences were well conserved. Our results indicated that the Chinese isolate is closely related to the Japanese isolate T.

[Observation on the effect of Sj32DNA vaccine of Schistosoma japonicum].

AIM: To explore the protective immunity against cercariae challenge in mice immunized with Sj32DNA vaccine of Schistosoma japonicum. METHODS: Primers were designed and the full length and part cDNA encoding 32 kDa(Sj32) amplified from pSj32 by PCR were cloned into pBK-CMV. Eukaryotic expression vector pBK-Sj32-1 containing full-length Sj32 cDNA and pBK-Sj32-2 containing fragment of Sj32 cDNA were constructed. After selection and identification, two plasmids were transfected into NIH 3T3 cells. The expression of Sj32 in transfected cells was confirmed by IFA. After identification, the two plasmids were prepared and purified on batches. Each BALB/c mouse was vaccinated at weeks 0, 3, and 5, by injection with 100 micrograms of purified Sj32DNA into quadriceps muscle. Mice immunized with constructs and the control mice were challenged four weeks after final DNA injection, and worms and the number of eggs in the liver tissue were counted at 8 weeks after challenge. RESULTS: Perfusion and egg counts showed significant differences in terms of worm reduction rate 38.6%-30.9% and egg reduction rate 55.7%-55.2% between the vaccinated and control mice. CONCLUSION: Sj32 DNA vaccine could result in protection against a subsequent challenge of S. japonicum in mice.
