A role for c-Abl in c-myc regulation.

c-Abl, a nonreceptor tyrosine kinase, appears to play a role in cell cycle progression, cell proliferation and differentiation. Mice homozygous for a mutation in c-abl (ablml), show pleiotropic abnormalities, including neonatal death, developmental defects, susceptibility to infection and dehydration (Schwartzberg et al., 1991). However, the exact substrates of c-Abl and the signal transduction pathways it might initiate are not known. We have examined how c-Abl affects c-myc expression by studying ablml mice. Quantitative riboprobe analyses demonstrated that in the heart, liver, thymus, brain, testes, intestines and lung, there were no differences in the steady-state level of c-myc RNA between the ablml mice and littermate controls. However, in adrenal glands, kidneys and splenic B cells, c-myc RNA levels were decreased approximately 50% compared to littermate controls. Induction of c-myc mRNA following activation of splenic B cells with LPS is also defective in ablml splenocytes. Finally, we show that c-Abl can directly transactivate c-myc transcription. These results suggest that c-Abl is involved in the normal transcription regulation of c-myc in selected tissues and that decreased c-myc RNA could be one cause of abnormalities in the ablml mice.

Work stress and health in a sample of U.S. police.

This study assessed responses to the Occupational Stress Indicator in the Naperville Police Department. This 40% sample of 49 police officers were compared against population norms for employed workers in a range of occupations. Although the police officers perceived less stress from "factors intrinsic to the job", they expressed greater job-related pressure from "organizational structure and climate". They also rated greater the competitiveness and assertiveness in their work behavior.

Limited expression of C/EBP family proteins during B lymphocyte development. Negative regulator Ig/EBP predominates early and activator NF-IL-6 is induced later.

The importance of C/EBP proteins in B cell biology is suggested by the occurrence of functionally important C/EBP binding sites in Ig gene enhancers and promoters, and the knowledge that family member NF-IL-6 is induced in other systems in response to regulators of B cell differentiation. We have studied the expression pattern and activity of C/EBP family transcriptional regulators in B cells at different developmental stages by using B cell lines and normal splenic B cells. Two family members, Ig/EBP and NF-IL-6, seem to be the major regulators of C/EBP site-dependent transcriptional activity in B cells. Negative regulator Ig/EBP is predominantly present in early B cells; activator NF-IL-6 increases in more mature B cells and is induced by LPS activation of splenic B cells. LIP, an N-terminally truncated form of NF-IL-6, was found in most B cell lines tested; LIP can act as a weak transcriptional activator in B cell lines. Partly as a result of the differential amounts of C/EBP family proteins, C/EBP sites do not function as activator sites in early B cells but are activator sites in terminally differentiated B cells.

Psychosocial factors and chronic fatigue syndrome.

This study investigated the number and severity of life events, Type A behaviour, coping strategies and social support differences between chronic fatigue and irritable bowel syndrome patients prior to illness and between these groups and healthy controls. Although few differences were found between the groups for life events, a number of interesting results emerged with regard to different aspects of Type A behaviour, various coping strategies and social support. These findings are discussed with respect to existing research in the field.

Detoxification of the plant toxin fluoroacetate by a genetically modified rumen bacterium.

We isolated the fluoroacetate dehalogenase gene (H1), from Moraxella species strain B, and placed it under the transcriptional control of a 154 bp fragment of the erm gene promoter. The promoter/gene construct was attached to the Butyrivibrio fibrisolvens shuttle vector pBHerm, and the resulting dehalogenase expression plasmid (pBHf) was transferred to B. fibrisolvens OB156 by electroporation. The erm gene promoter directed expression of dehalogenase activity in both E. coli and B. fibrisolvens OB156. Cell-free lysates of the genetically modified OB156 defluorinated 10.6 nmol fluoroacetate/min/mg protein. Growing cultures of OB156 were able to detoxify fluoroacetate in the culture medium, at the rate of 9.9 nmol/min/mg. Plasmid pBHf was retained by 100% of OB156 cells after 500 generations of non-selective culture. The restriction pattern of pBHf remained unchanged after extensive non-selective growth and host bacteria continued to produce active dehalogenase. The construction of rumen bacteria that are able to detoxify an important natural poison supports the feasibility of using genetically modified rumen bacteria to aid animal production.

The impact of trust status on corporate culture.

Outlines the results of a study designed to examine the effects of trust status on perceived organizational culture and over time. Through the adoption of a prospective longitudinal design and incorporating both pre- and post-trust measures it has been possible to ascertain a number of issues including the move from a support-based to a power-based culture with greater autocracy and centralization of power and control. Further research is planned to examine the impact of these changes on the health and performance of individuals moving through the transitional stages of major organizational change.

Job stress, ill health and job satisfaction among health service employees.

The study examined the effects of occupational stressors, perceived locus of control, Type A behaviour pattern and use of coping strategies on well-being and job satisfaction of 1,176 health service employees. Several stressors were identified as having a negative impact on health and job satisfaction although these differed between the various occupational groups included in the study. The interactionist model of stress used in the study, and the diagnostic tool used (the Occupational Stress Indicator) proved to be useful in suggesting means of intervening to reduce ill health and increase job satisfaction among health service employees.

Personality, stress, coping, and sense of coherence among nephrology nurses in dialysis settings.

The goal of this study was to examine the relationships among personality types, personal and work-related stress, coping resources, and sense of coherence (SOC) among nephrology nurses in dialysis settings. Nurses (n = 49) from 13 dialysis units in New Mexico completed a demographic data form, Perceived and Nursing Stress Scales, SOC Scale, Coping Resources Inventory, and the Myers-Briggs Type Indicator (MBTI). The results indicated that there was a positive correlation between perceived personal stress and work-related stress, especially work load. Conversely, there were negative correlations between (a) both personal and work-related stress with SOC, and (b) both coping resources and SOC with burnout. High levels of personal and work-related stress were related to inadequate coping resources. Regression analysis indicated that the main contributing factors to emotional exhaustion (a major component of burnout) were low SOC, lack of staff support, personal stress, and heavy work load. Increased utilization of coping resources may facilitate the nurses' management of personal and work-related stressors.

Neomycin and spermine have similar effects on secretion and phosphoinositide metabolism in ATP-depleted permeabilized adrenal chromaffin cells.

To evaluate the requirement of bovine adrenal chromaffin cells for inositol phospholipids in secretion, the effects of neomycin and spermine on secretion and phosphoinositide metabolism were compared. Spermine and neomycin had virtually identical effects on secretion and phosphoinositide levels. Both polyamines 1) partially maintained secretion when added to permeabilized cells in the absence of ATP, 2) had no effect when added to cells in the presence of ATP and 3) inhibited secretion when present with the Ca2+ stimulus. In the absence of ATP the enhancements to secretion due to incubation with either polyamine were associated with sustained levels of the polyphosphoinositides. The ability of spermine and neomycin to maintain secretion and the polyphosphoinositides in the absence of ATP supports the hypothesis that the maintenance of the polyphosphoinositides is necessary for secretion. Neomycin was found to inhibit Ca2+ stimulated production of both inositol bis- and tris-phosphates while spermine was found to selectively inhibit inositol bis-phosphate production and had no effect on Ca(2+)-stimulated inositol tris-phosphate production in permeabilized cells.

Determination of total polyamines in tumor cells by high-performance capillary zone electrophoresis with indirect photometric detection.

Tumor cells contain much higher levels of common polyamines such as putrescine, spermidine, and spermine than normal cells. Our previous work demonstrated that high-performance capillary zone electrophoresis with indirect photometric detection was suitable for the determination of underivatized free polyamines in tumor cells. We now report improved sensitivity (7-fold), greater quantitative reproducibility, and quantitation of both bound and free, underivatized polyamines in PC 12 tumor cells. 1,7-Diaminoheptane was used as an internal standard for quantification. Femtomoles of three polyamines and simple cations such as K+ and Na+ were easily separated in less than 10 min. The quantitations for total spermidine and spermine (bound and free) were found to be 2-fold higher than the quantitation for free, unbound polyamines using our previous method. The signal response was linear over 2 orders of magnitude. The method described here provides a fast and simple technique for quantitation of bound, unbound, and total polyamines for use in the study of the biological role of polyamines.

Identifying distress among general practitioners: predictors of psychological ill-health and job dissatisfaction.

This paper attempts to identify sources of job stress and personality factors as predictors of psychological ill-health and job dissatisfaction among a large sample of general practitioners in the United Kingdom. Compared to a normative sample, male doctors exhibit significantly higher levels of anxiety and depression, whereas female doctors compare favourably to the population norms. Job satisfaction levels were significantly lower than when they were measured in 1987, although women were less job dissatisfied than men. The main predictors of lack of mental well-being were the job stressors associated with the 'demands of the job and patients' expectations', 'practice administration and routine medical work', 'role stress' and the use of 'social support' as a coping strategy.

Psychosocial stress and breast cancer: the inter-relationship between stress events, coping strategies and personality.

This quasi-prospective study of 2163 women attending a breast-screening clinic for a routine medical check-up indicates that certain types of coping strategies and personality dispositions predispose some women to an increased risk of developing breast cancer following the occurrence of a major life-event such as bereavement or other loss-related event. Regular exposure to stress situations appears to reduce the risk of a malignancy; experiencing a single, major life event was found to be potentially much more damaging, particularly if the individual was unable to externalize her emotions and obtain appropriate help and counselling.

Occupational stress in head teachers: a national UK study.

This study assessed occupational stress amongst 2,638 head teachers of primary and secondary schools, together with principals/directors of further and higher education establishments, throughout the United Kingdom. Data were collected on personal/job demographics, sources of job stress, mental health, job satisfaction and coping strategies. These data were analysed by SPSS-X, producing univariate, bivariate and multivariate techniques. It was found that as we moved from the further/higher education level to secondary to primary sectors, the levels of job dissatisfaction and mental ill health rose. In addition, it was found that, with the exception of primary schools, female head teachers in secondary and FHE seem to be suffering significantly greater job dissatisfaction than their male counterparts, although this does not translate itself into mental ill health. Male head teachers, on the other hand, seem to suffer more mental ill health than their female counterparts. And finally, the two main sources of occupational stress that appear in many of the multivariate analyses as predictors of job dissatisfaction and mental ill health are 'work overload' and 'handling relationships with staff'. The implications of all these findings are discussed in detail.

Research note: reliability of self-report sickness absence data in the health service.

Rees and Cooper (1990) used self-reports of sickness absence during the previous six months as a dependent measure in a diagnostic survey of stress in one health district. Comparison of self-reports with actual sickness absence showed that it achieved a high level of association (0.95) and can be recommended as an alternative to actual sickness data in studies in which, for whatever reason, actual sickness absence is difficult to obtain.

Characteristics of peripheral and peritoneal lymphocytes from continuous ambulatory peritoneal dialysis patients.

The role of peritoneal lymphocytes in host immunity for continuous ambulatory peritoneal dialysis (CAPD) patients is just beginning to be understood. CAPD therapy increases the proportion of peritoneal lymphocytes, most of which demonstrate signs of activation. There are decreased peritoneal T cells and increased peritoneal B cells as compared to the patients' peripheral blood. When studies examine immunophenotypes of peripheral and peritoneal lymphocytes over time, no significant changes are found. Although changes in peritoneal lymphocyte subsets occur during peritonitis episodes, there are no changes in peripheral blood lymphocytes. The purpose of this article is to provide a brief review of research that has studied lymphocytes in CAPD patients.

Indirect photometric detection of polyamines in biological samples separated by high-performance capillary electrophoresis.

A rapid separation of polyamines and some related amino acids in cultured tumor cells by high-performance capillary zone electrophoresis with indirect photometric detection is demonstrated. 60 cm x 75 microns I.D. fused-silica capillary was used for the separation and quinine sulfate was used as a background electrolyte (BGE). Several polyamines (putrescine, spermidine and spermine), amino acids (lysine, arginine, histidine) and simple cations (K+, Na+) were easily separated in less than 10 min. Using the indirect photometric detection method, femtomole amounts of polyamines extracted from the tumor cells were detected from nanoliter injection volumes, and the signal response was linear over two orders of magnitude.

Job stress, satisfaction, and mental health among general practitioners before and after introduction of new contract.

OBJECTIVE: To compare measures of job stress, job satisfaction, and mental health among general practitioners before and after the introduction of the new contract in April 1990. DESIGN: Cross sectional postal questionnaire survey in July 1990. Comparison of results with those obtained in previous survey in November 1987. SETTING: General practice in United Kingdom. SUBJECTS: 1500 general practitioners randomly selected from general medical services lists, 917 of whom (61%) returned questionnaires usable for statistical analysis. MAIN OUTCOME MEASURES: Aspects of job causing stress, job satisfaction (Warr, Cook, and Wall scale), and mental health (Crown-Crisp experiential index). RESULTS: Compared with 1987, in 1990 doctors experienced more stress from night calls (mean score 3.83 in 1990 v 3.45 in 1987), emergencies during surgery hours (3.72 v 3.48), and interruption of family life by telephone (3.58 v 2.73; p less than 0.001 for all three variables). Scores for somatic anxiety and depression were higher in both men and women in 1990 (men: somatic anxiety 3.12 v 2.36; depression 3.80 v 2.94; women: somatic anxiety 3.56 v 2.65; depression 4.02 v 3.37; p less than 0.001). Job satisfaction had also decreased in 1990 (5.23 v 4.26; p less than 0.001). CONCLUSIONS: Doctors experienced more stress, less job satisfaction, and poorer mental health in 1990 than in 1987. These changes may have resulted from the introduction of the new contract.
