The effects of substance P and related peptides on alpha-amylase release from rat parotid gland slices.
1 The effects of substance P and related peptides on amylase release from rat parotid gland slices have been investigated. 2 Supramaximal concentrations (1 microM) of substance P caused enhancement of amylase release over the basal level within 1 min; this lasted for at least 40 min at 30 degrees C. 3 Substance P-stimulated amylase release was partially dependent on extracellular calcium and could be inhibited by 50% upon removal of extracellular calcium. 4 Substance P stimulated amylase release in a dose-dependent manner with an ED50 of 18 nM. 5 All C-terminal fragments of substance P were less potent than substance P in stimulating amylase release. The C-terminal hexapeptide of substance P was the minimum structure for potent activity in this system, having 1/3 to 1/8 the potency of substance P. There was a dramatic drop in potency for the C-terminal pentapeptide of substance P or substance P free acid. Physalaemin was more potent than substance P (ED50 = 7 nM), eledoisin was about equipotent with substance P (ED50 = 17 nM), and kassinin less potent that substance P (ED50 = 150 nM). 6 The structure-activity profile observed is very similar to that for stimulation of salivation in vivo, indicating that the same receptors are involved in mediating these responses. 7 All the fragments of substance P tested were capable of eliciting a full amylase release response. This indicates that the apparent partial agonist action of the C-terminal nonapeptide fragment on in vivo salivation is not explicable at the receptor level.

Rapid cytological diagnosis of basal cell carcinoma of the skin.
Cytological diagnosis in a series of 153 skin lesions is presented. The diagnosis in each case has been compared with the clinical assessment and, where available, the biopsy result (85 lesions). Of 131 basal cell carcinomas, a cytological diagnosis was possible in 124 while biopsyl failed to identify two of the 73 lesions studied. A correct cytological diagnosis was made in five squamous cell carcinomas and in one malignant melanoma. Basal cell papillomas and solar keratoses were not accurately identified. We thus conclude that with this very simple, rapid, and inexpensive method a clinical diagnosis of basal cell carcinoma is reliably confirmed with advantages for both the clinic and the laboratory.

Gas phase carboxylate anions fron p-nitrobenzyl esters of fatty acids.
Gas phase carboxylate anions have been efficiently generated from the p-nitrobenzyl esters of nine fatty acids ranging in size from C6 to C22. The process involves the dissociative resonance capture of 2.9 e V electrons by the derivatives. The procedure is specific and gives the carboxylate anions as the base peak in all but two of the derivative. The technique gives extremely simple spectra, with little additional fragmentation and no rearrangement ions.
Identification of oleoresin in epoxy-embedded slash pine tissue.
Sudan black B stains oleoresin blue-black in epoxy-embedded material as well as in living tissue. The Sudan black B staining properties of oleoresin are similar to those of lipid, but it can be distinguished from tannin, which stains brown. Pratically all oleoresin present in resin ducts and intercellular spaces, and much of that contained in epithelial and ray cells, is extracted in preparatory procedure for electron microscopy. A fixation procedure is proposed which preserves significantly more oleoresin in situ. The use of Sudan black B enables one to localize oleoresin by light microscopy, and permits direct comparison of adjacent sections of epoxy-embedded material at the ultrastructure level. Ultrastructurally oleoresin and lipid possess similar electron densities and can be distinguished from the highly electron-opaque tannin deposits.

Primary malignant tumours of the thyroid: the relationship between histological classification and clinical behaviour.
One hundred and seventy-nine primary malignant tumours of the thyroid seen at The London Hospital between 1945 and 1972 were classified by the system of Woolner et al. (1961) and Hazard (1964). The distinct pathological and clinical features of the differentiated primary carcinomas and the similarities and differences between malignant lymphoma and anaplastic carcinoma were confirmed. This study showed the 'benign' behaviour of more than half the 'differentiated' papillary and follicular carcinomas when treated by thyroid lobectomy and the very malignant nature of anaplastic carcinomas and lymphomas whatever their treatment. The behaviour of medullary carcinoma was closer to that of the other differentiated tumours than to the undifferentiated varieties. Our patients were not thought to have been exposed to known goitrogens or previous thyroid irradiation.
