Studies on the cytophilic properties of human beta2 microglobulin.

Human beta2 microglobulin is cytophilic for mouse, rat, and to a lesser extent guinea pig lymphocytes and polymorphonuclear leukocytes but not for human, rabbit, chicken, frog, and turtle cells, nor for any mature erythrocytes tested. Mouse splenic lymphocytes bind 5- 7-fold more beta2 microblobulin than thymocytes although hydrocortisone-resistant thymocytes resemble spleen cells in this regard. The beta2 microglobulin binding by B and T cells in mouse spleen is similar. The structure responsible for beta2 microglobulin binding to the surface of cells is distinct from the Fc receptor specific for aggregated IgG.

Receptors for aggregated IgG on mouse lymphocytes: their presence on thymocytes, thymus-derived, and bone marrow-derived lymphocytes.

An autoradiographic binding assay employing (125)I-labeled heat-aggregated mouse IgG2b myeloma protein (MOPC 141) was used to demonstrate receptors for IgG on 20-45% of Balb/c thymocytes and on 70-80% of splenocytes. Binding could also be shown with heat or BDB aggregates of another IgG2b (MOPC 195), with IgG1 and with human gamma-globulin, but not with aggregated chicken gamma-globulin, IgA, BSA, nor with aggregated Fab fragments of IgG2b. Optimum binding was obtained at 37 degrees C. Detection of binding was dependent upon aggregate size with complexes of more than 100 IgG molecules being optimal, aggregates of 6-25 detecting splenocytes but not thymocytes, and aggregates of less than 6 binding to a negligible extent. Comparison of grain counts on various cell types showed mastocytoma cells (P815) and macrophages averaging 40-50 grains/cell/day, allogeneically activated thymocytes 20-30, splenocytes 2-3, L5178 lymphoma cells 1, and positive thymocytes 0.6 grains/cell/day. Double labeling experiments for surface Ig, theta-antigen, and agg IgG receptor on mouse spleen cells indicated that a relatively high density of receptor was present on about 80% of B cells, 30% of T cells, and 60% of SIg(-), theta(-), null cells.

Effects of prevention of afferentation of the development of the chick optic lobe.

The effects of unilateral extirpation of the right optic cup of the three-day incubated chick embryo upon the rate of synthesis and the stability of DNA in the non-innervated optic lobe, have been studied. This surgical procedure prevents innervation of the optic lobe contralateral to the removed eye, while the other optic lobe is normally innervated by retinal ganglion cells of the remaining eye. At the 20th day of incubation, the DNA content of the non-innervated lobe was below that of the paired lobe receiving normal innervation. This deficiency of cell number was caused by two events; death of an excess number of neurons formed early in embryogenesis and a reduced rate of glial proliferation in the later stages of incubation.

Neonatal burst suppression: its developmental significance.

Burst suppression was recorded on electroencephalograms of 15 of 274 term infants (5.4%) in our hospital within a 5 year period. These 15 infants were examined, their perinatal histories reviewed, and detailed neurodevelopmental testing performed to examine the prognostic significance of burst suppression. Fourteen children (93%) had poor outcomes. Four (26%) died in infancy. Ten survivors were followed for a mean of 30 months (range: 18-61 months). Nine have severe motor deficits, 6 require total custodial care, 5 have intractable epilepsy, and 3 are blind. Their mean Gesell Developmental Quotient was 28; a score of less than 68 suggests severe retardation. A single patient, the only 1 with a perinatal history complicated by an easily-treated bacterial meningitis, achieved normal growth and development. This study documents the dire prognosis of neonatal burst suppression. The appearance of burst suppression, though transient, portended death or severe neurodevelopmental disability in 93% of our patients.

Differences between pickup truck and automobile driver-owners.

This study compares pickup truck driver-owners and drivers who owned only automobiles with respect to demographic factors, conditions of use, risk-taking driving behavior, prior driving history and attitudes towards motor vehicle laws. A telephone survey conducted in Riverside County, CA determined that 36% of the households had a pickup truck. Pickup truck owners were primarily male, aged 30-39 years, married, reported lower restraint use and more risky driving behaviors, and had more traffic citations. Differences in behaviors and attitudes were largely a function of age and gender. There is a need to design appropriate occupant safety interventions for those most likely to own pickup trucks.

Rapidly progressive renal failure as the primary manifestation of Wegener's granulomatosis.

A 43-year-old woman was treated for a rapidly progressive glomerulonephritis with periodic renal dialysis for a period of over one year before the development of a pulmonary lesion which enabled a diagnosis of Wegener's granulomatosis to be made. The lung lesion cleared rapidly with azathioprine and steroid therapy; severe renal failure, however, persisted, necessitating continued hemodialysis. An early and predominant manifestation of a rapidly progressive glomerulonephritis in the presence of minimal, if any, upper respiratory disease, albeit rare, should be considered as a possible presentation of Wegener's granulomatosis.

Males as sexual assault victims: multiple levels of trauma.

The definite and persistent phenomenon of sexual assault upon males is virtually ignored in the literature, although incidence data reported here suggest the greater scope of the problem. The avoidance of the subject of sexual assault on males creates a negative environment for victims. While the motivation of assailants is briefly discussed, the article focuses on the psychological aftermath for sexual assault victims. A paradigm is offered, consisting of "Set-up," "Attack," and "Aftermath" phases. Male victims suffer "Rape Trauma Syndrome" as described for females, as well as various forms of stigmatization and secondary trauma. Differences and similarities between male and female victims are identified. Victim responses are discussed as they proceed through several stages, with implications for appropriate interventions on both the clinical and community levels. The article concludes with an extensive bibliography.

The adolescent ex-cultist: a nursing staff challenge.

Cult memberships involve between 2 and 3 million young adults. Ex-cultists present the health care facility with a complexity of both medical and psychological problems. Nursing staff with excellent communication skills and knowledge about adolescents and cults can make accurate assessments of needs and implement effective interventions.

Cord blood thyrotropin screening for congenital hypothyroidism. Three years' experience on the Island of Saint Lucia.

Cord blood thyrotropin (TSH) screening for congenital primary hypothyroidism has been in effect on the island of St. Lucia for the past three years. Umbilical cord blood samples are obtained on Guthrie filter paper and then transported 3,000 miles to Loyola University of Chicago and delivered to the Illinois State Metabolic Screening Laboratory. There TSH is measured by radioimmunoassay (RIA). After three years, 1,789 newborns have been screened, and the mean value is 6.23 +/- 0.13 microIU per ml. This mean value is less than previously reported by us in 1986 (10.23 +/- 0.29 microIU per ml).13 It is concluded that this screening service continues to be possible far removed from the population under observation. No case of primary hypothyroidism has been detected. Our decreased mean TSH value is due to the new method currently used by the Illinois State Metabolic Screening Laboratory. Congenital hypothyroidism will not be missed provided internal controls are established and rigidly observed.

BAY K 8644-induced enhancement of 45Ca uptake by rabbit aortic rings.

BAY K 8644, a Ca2+ channel activator, enhances uptake of 45Ca by rabbit aortic rings. This effect depends on the concentration of K+ in the medium: at 20 mM K+ the effect of BAY K 8644 was more pronounced than at 5 mM, whereas at 80 mM, no significant enhancement of 45Ca uptake by BAY K 8644 was found. In the medium containing 5 mM K+, BAY K 8644 was effective in experiments involving 10 or 30 (but not 3) min exposure of aortic rings to 45Ca. The dose-response curve for BAY K 8644 was established in 5 mM K+-containing medium and for 30 min exposure to 45Ca. BAY K 8644 was effective at 0.01 microM and higher concentrations. In the presence of norepinephrine (0.1 or 10 microM), BAY K 8644 had no greater effect on 45Ca uptake than in control 5 mM K+ medium. Our observation that the presence of norepinephrine in 5 mM K+ did not enhance BAY K 8644-induced 45Ca uptake suggests that activation of alpha-adrenergic receptor does not depolarize aortic membranes to the same extent as an increase in K+ concentration to 20 mM or that BAY K 8644 does not enhance Ca2+ entry through receptor-operated channels.
