Computer programs for handling nucleic acid sequences.

Programs have been developed that will accurately display restriction enzyme maps, open translational reading frames and the results of specific searches on a high resolution graphics terminal (Retrographics VT640). Many of our earlier programs have been upgraded to allow online access to GENBANK, the NIH data bank of DNA sequences and also to deal with sequences of any length. Progress has also been made in the automation of the DNA assembly programs.

The role of fluid hydrostatic pressure in bone-implant interface load transfer.

Permeability of the soft tissue-bone system surrounding artificial joints fixed in cancellous bone was measured in four adult dogs after implants had been in place 2 months. Fluid was forced through a cavity formed by removal of the implant, the cavity was capped with a stopper to allow for pressure generation. Surface permeability of the 2-month-old implant cavity was 45 times less than the permeability of freshly drilled holes in cancellous bone. A mathematical model of a rigid implant resting on a biphasic solid-fluid layer showed the fluid carried 90% of the load when the implant cavity permeability was assumed, but only 27% when the freshly drilled permeability was used. The results suggest caution in interpreting finite-element models with bonded interfaces and suggest a possible role of the fluid in biological response at the interface.

[Extracardiac atherosclerosis in patients with familial hypercholesterolemia].

Among 64 patients with familial hypercholesterolemia (FHC) confirmed by tissue culture of skin fibroblasts (3 homozygotes, 9 heterozygotes) 12 (3 homozygotes, 9 heterozygotes) had signs of atherosclerosis of the carotid arteries and of the peripheral arteries (extracardiac vasculature). 8 heterozygotes as well as the three homozygotes had coronary atherosclerosis. Three of the 12 patients with carotid artery disease suffered from clinical symptoms of cerebrovascular insufficiency. Only five patients with heterozygous FHC had symptomatic peripheral artery disease though 10 including the three homozygotes had signs of atherosclerosis of the abdominal aorta and its peripheral branches. It is concluded that patients with FHC develop generalized atherosclerosis but not always symptoms of peripheral artery disease. Risk factors other than hypercholesterolemia do not seem to contribute essentially to the development of atherosclerosis. It seems important to evaluate patients with FHC regularly for atherosclerosis of the carotid arteries which might be recognized earlier than coronary atherosclerosis.

Xeroradiographic determination of Achilles tendon thickness in familial hypercholesterolemia confirmed by tissue cultures.

Twenty-three patients with familial hypercholesterolemia (FHC) confirmed by tissue cultures of skin fibroblasts (2 homozygotes, 21 heterozygotes) and 3 patients with sporadic hypercholesterolemia were evaluated for Achilles tendon (AT) thickness by xeroradiography. Both homozygotes had thick ATs and coronary heart disease (CHD), 20 heterozygotes had thick ATs, but only 8 of them had CHD. One heterozygote had a small AT value, but CHD for a long time. The 3 patients with sporadic hypercholesterolemia, one with CHD, had small AT. There was no correlation between serum cholesterol concentration and AT thickness, between age and AT thickness, nor between AT thickness and CHD.

Plasmids of human strains of Yersinia enterocolitica: molecular relatedness and possible importance for pathogenesis.

One-hundred fifteen strain of Yersinia enterocolitica were examined for plasmids and plasmid-mediated pathogenic properties. Human strains of serotypes O:3 and O:9 harbored plasmids of 46 and 44 megadaltons, respectively, with 90% homology of DNA sequences. The plasmid-mediated properties were calcium dependence, survival in human serum, conjunctivitis provocation in guinea pigs, and O agglutinogens. One strains of serotype O:8 harbored a 42-megadalton plasmid with 75% sequence homology with plasmids of serotypes O:3 and O:9. An additional plasmid-mediated property was lethality for white mice. Filter hybridization of restriction endonuclease-digested plasmid DNA indicated that a 5.6-megadalton fragment of the plasmid of serotype O:8 had virtually no sequence homology with plasmid DNA of serotypes O:3 and O:9 and therefore may be associated with the lethal factor for mice.

Chylothorax secondary to superior vena caval obstruction.

Bilateral lethal chylothoraces developed in a premature infant secondary to superior vena caval obstruction related to central venous catheterization for total parenteral nutrition. The accumulation of chyle in the lungs was preceded radiographically by the typical lymphatic pattern in the lungs, usually associated with lymphangiectasia. A nuclear medicine lymphangiogram showed abnormal accumulation of the isotope in the lungs secondary to the lymphatic obstruction.

Serum cholesterol levels in patients with familial hypercholesterolemia confirmed by tissue culture.

Sixty-two subjects from 23 families were evaluated by serum lipid analyses and tissue culture biochemistry in skin fibroblasts. In 53 cases from 19 families with proven familial hypercholesterolemia (FHC), fibroblast cultures were successful. In 45 of these cases (85%) the clinical diagnosis of hyper- or normocholesterolemia was in accordance with the tissue culture findings. Four patients 2-38 years old, had hypercholesterolemia but normal tissue culture results. Four patients, 18-44 years old, had normal serum cholesterol levels for their age and sex, but were heterozygotes according to tissue culture results. In the remaining four families only the propositus had hypercholesterolemia. All members of the families including the propositus had normal tissue culture determinations indicating that not all cases of idiopathic hypercholesterolemia are due to the Goldstein-Brown mechanism of defective LDL receptor function.

A 300 kcal (1.2 MJ) diet using conventional food.

A 300 kcal (1.25 MJ) diet of conventional food is described, which has been studied under in-patient conditions for four to six weeks. It contained 22.6 g protein, 34 g CHO and 6.9 g fat but not the full RDA of vitamins and minerals since this is impossible without supplementation. Hunger disappeared after the third day. Patients developed ketonuria and hyperuricemia; serum lipids were normalised and hypertension disappeared. The diet offers advantages in that it induces better nutritional knowledge and habits.

[Homozygous familial hypercholesterolemia. Cardiovascular findings in two patients (author's transl)].

In patients dying in adolescence with familial hypercholesterolemia a combination of xanthomata of the skin and tendons, atheromatous plaques on the aortic and mitral valves, the coronary artery outlets and in the proximal aorta is frequently found. However, little is known of the sequence of these changes or their clinical equivalents. For this reason findings are described in two patients whose homozygous familial hypercholesterolemia is confirmed by studies of the cell biochemistry of the dermal fibroblasts after an observation period of eight and twelve years.
