Neoplastic transformation and cytogenetic aberrations of rat tracheal epithelial cells induced by alpha-particle irradiation.

Primary rat tracheal epithelium (RTE) cells can be transformed in vitro by alpha-particles irradiation. One out of 3 isolated morphologically transformed colonies gave rise to an immortal cell line and ultimately became neoplastic by successive passaging. Cytogenetic analysis was performed on the cell line in order to understand how the specific chromosome alterations participate in the process of neoplastic transformation of RTE cells. The 5th, 18th, 39th passage cells and the colony cells isolated from soft agar (SA) culturing of the 40th passage cells were analyzed. The results showed that the 20th passage cells were nontumorigenic, but the 40th passage cells were tumorigenic. The increase of the length of the long arm of chromosome 15 (15q+) was frequent at the 5th passage (67.8%), especially in SA cells in which it occurred in a clonal fashion (100%). Therefore, 15q+ might be the critical gene lesion after alpha-particles irradiation. Monosomy of chromosome 8 that occurred frequently at passage 5 (54%) and 40 (53%) might be correlated with earlier transformation of RTE cells. In addition, three marker chromosomes (M1, M2 and M3) showed a passage-dependent increase. In particular M1 and M2 that were highly frequent at passage 39 (90%) suggested that they might play an important role in the process of cell transformation.

[Pharmacological studies of luohanguo yanhou tablet].

Luohanguo Yanhou Tablet possesses the action of cotton ball granuloma of mice and the auricular edema induced by xylol, and also inhibiting the swelling of rat hind paw induced by carrageenin. The tests of mice writhing induced by acetic acid showed that the preparation had obvious analgesic action. It also possesses the obvious bacteriostatic action in vitro on five kinds of strains.

[Clinical study on the antithrombotic effects of Holothuria ldeucospilota acidic mucopolysaccharide].

OBJECTIVE: To study the antithrombotic effects of Holothuria Ldeucospilota Acidic Mucopolysaccharide (HLAM). METHODS: A series of variables were observed before and after treatment with HLAM in normal middle-aged subjects and patients recovering from cerebral embolism or suffering from ischemic heart disease, each of which were divided into two groups according to the dosage administered (60 mg/d and 120 mg/d). RESULTS: The fat metabolism was enhanced in addition to the anticoagulant and the blood viscosity reducing effects. No side-effect was found. CONCLUSION: HLAM is a promising drug for antithrombotic therapy.

Field emission SEM, conventional TEM and HVTEM study of submandibular gland in prenatal and postnatal aging mouse.

The development of acinar and ductal cells of the mouse submandibular gland was studied using field emission SEM, conventional TEM and HVTEM methods. The specimens, at 15 and 18 days of gestation and 1, 3, 7, 14, 21, 30, 90 and 180 postnatal days were fixed in 2.5% glutaraldehyde solution in 0.1 M sodium phosphate buffer (pH 7.3). At 15 and 18 days of gestation, the structure of mouse submandibular gland contains acinar and ductal cells in proliferation. The cytoplasmic organelles such as mitochondria, granular endoplasmic reticulum and Golgi apparatuses are scattered in the cytoplasm. At 18 prenatal days only several acinar cells present immature secretory granules in the apical portion. In this stage the acinar and ductal cells are enveloped by bundles of fine collagen fibrils disposed in several directions. There are also numerous capillaries located closely to the acinar cell membranes. In the aging stages of 1, 3, 7, 14, 21, and 30 postnatal days, the histo-differentiation of acinar, intercalated and ductal cell components are observed. At newborn day one the cytoplasmic organelles start to place themselves around the nucleus. Several immature secretory granules are observed at day one, however, they increase in the aging days. At postnatal day 30, the cytoplasms of acinar and ductal cells are filled with a large number of secretory granules of different sizes. The stacks of granular endoplasmic reticulum and Golgi apparatus and some vesicles and free ribosomes are noted. The intercellular membranes are attached by desmosomes and cytoplasmic interdigitations. The luminal surface shows several small projections of microvilli. An electron-dense line of basement membranes followed by fine collagen fibrils are recognized. Delicate capillaries are found in the outer surface of acinar cells. At postnatal day 90 and 180 the acinar, intercalated and striated ductal cells reveal numerous secretory granules in the apical portion. The acinar cells showed basal nuclei and the parallel arrangement of granular endoplasmic reticulum. The mitochondria are located at the base of ductal cells showing a typical pattern of cristae. In these stages the intercellular digitations of cytoplasmic protrusions and desmosomes are also noted. The cytoplasm of myoepithelial cells are seen along the cell membranes. The spongy-like structures constituting the basement membrane are followed by bundles of fine collagen fibers.

Clonal chromosome abnormalities in two chemodectomas.

Short-term cultures from two histologically benign chemodectomas, one from the carotid body and one from the vagal nerve, were analyzed cytogenetically. The former had a small abnormal clone with the karyotype 46,XX,t(3;19)(q21;q13),t(12;15) (p13;q12-14), whereas the majority of the cells from the latter tumor displayed two related abnormal clones: 46,XY,i(I)(q10)/ 46,iderm,add(2)(q37). The findings add to the evidence that chemodectomas are heterogeneous neoplasms and suggest that the heterogeneity may possibly be associated with the site of origin.

[Photocoagulation and scleral cryotherapy for Coat's disease].

PURPOSE: To evaluate the results and significance of Coat's disease with argon laser photocoagulation and cryotherapy. METHODS: 22 patients (23 eyes) were treated by argon laser (German Opton Meditek 40 E, 0.2w power, 100-500 um spot, 0.1-0.2 second exposure and grade II reaction) and or scleral cryotherapy (-60 to approximately -80 degrees C, 5 second duration per time, repeating treatment if necessary until pale edema was seen in retina). RESULTS: 18 eyes in all (78.2%) had good results with visual acuity improvement and the lesion of fundus had been seen decreased in varient degrees. The follow-up ranged from 1 to 3 years. CONCLUSION: Photocoagulation and scleral cryotherapy are effective for those patients with Coat's disease. It is very important to treat them promptly so as to prevent the damage of visual function progressively.

Study on DNA synthesis of aging mouse colon by light and electron microscopic radioautography.

Aging changes of DNA synthesis and of ultrastructure in mouse colon were analyzed by light and electron microscopic radioautography. The sites of 3H-thymidine uptake were identified in each layer of the colon. The highest labeling index (LI) of the epithelial cells was found at embryonic day 19; it decreased from neonatal day to 1 week and then remained on a constant value until 1 year. The LI's of the other layer cells were high at prenatal day 19, followed by a decrease with the development of the colon disappearing between 2 and 6 months after birth. The LI of the inner circular muscle layer was in every experimental age group higher than that of the outer longitudinal layer. The LI of vacuolated cells, high in the late embryonic and neonatal periods, decreased significantly between 3rd and 8th day and then remained unchanged until the old stage. The LI of the goblet cells showed a peak at prenatal day 19, decreased afterwards with time and disappeared 2 months after birth. The ultrastructural aspects showed that many silver grains located over the nuclei of the labelled cells which contained less cell organelles, in their cytoplasm as compared to unlabelled cells.

[The detection of laryngeal carcinoma-associated antigens in tissue and serum using three mixed monoclonal antibodies against laryngeal cancer].

Laryngeal carcinoma-associated antigens in tissue and serum of patients with laryngeal carcinoma and normal adults were detected by using ABC and ELISA methods. The results showed that the positive rate rised from 80-86.6% to 97.7% by applying mixed monoclonal antibodies compared with single monoclonal antibody against laryngeal cancer. It was also found that the level of laryngeal carcinoma-associated antigens was much higher in serum of laryngeal carcinoma group than that of control group. The statistic difference was very significant (P < 0.01). The level was different with clinical types, stages and increased in line with the tumor growth. So it was suggested that the mixed monoclonal antibodies were sensitive and specific, which was considered as a useful tumor marker for diagnosis, monitoring clinial course and judging prognosis of patient with laryngeal carcinoma.

[Applied anatomy of the medial superior genicular artery and the retrograde island flap].

Through anatomical dissection of fifty freshly amputated adult lower extremities, the origin, length, diameter and extent of perfusion of the superior genicular artery and its relation with the anterior tibial recurrent artery were investigated. Results revealed that the medial superior genicular artery originated primarily from the femoral artery (95.5%). It descends between the sartorius and majormedial muscles and then separates into branches that nourish the upper one-third of the lower leg and the medial skin of the knee, and then, by anastomosis, joins the anterior tibia recurrent artery on the level of the medial malleolus of the tibia. The formation rate of the anastomotic arch is 100% with the average outer diameter of 1.3 +/- 0.39 mm (x +/- s). Based on these observations, we conclude that the medial area of the knee is the best donor site of an island flap, which can be used to repair the medial superior one-third of the lower leg and the region around the knee.

[Preparation of galactosyl-cytochrome C and studies on its hepatic targeting property].

This paper reports the preparation of the coupling compound between thiogalactose and cytochrome C(Cyt-C). The binding tests of this compound to asialoglycoprotein receptor have also been carried out in vivo. The results show that the pharmacokinetics of this compound is in accordance with the basic characteristic of receptor induced binding. The highest uptake of liver reaches to 32.9% and at the same time the conjugate remains 78.6% the biological activity of Cyt-C. It is possibly a hepatic targeting drug or a hepatic targeting carrier.

[Clinical observation of shexiang baoxinwan for treatment of coronary heart disease with angina pectoris].

Two hundred and ten patients with angina pectoris in coronary heart disease were randomly assigned to receiving Shexiang Baoxinwan (SXBXW), the protecting heart musk pills (110 patients), or receiving isosorbide dinitrate (100 patients) for 2 weeks. They were well matched for age, anginal types, complications and cardiac functional grading. The results demonstrated that both agents not only could reduce significantly the frequencies of anginal episodes and the consumption amount of nitroglycerin per day, they also could result in significant improvement of the sigma ST segment depression in electrocardiogram. Furthermore, the SXBXW had better efficacy than that of isosorbide dinitrate in the total improvement rate of symptoms of angina pectoris and of ECG, and in the reduction of incidence of headache, i.e. 90.0% vs. 76.0%; 58.6% vs. 40.8%; 0.9% vs. 17%, respectively (P < 0.05 or P < 0.01). In addition, the onset of action of both agents was rapid, within five minutes (83.4% in patients using SXBXW vs. 86.3% in control group, P > 0.05).

Nonrandom chromosome abnormalities in short-term cultured primary squamous cell carcinomas of the head and neck.

We report the finding of clonal chromosome abnormalities in short-term cultures from 44 squamous cell carcinomas of the head and neck region. Eleven tumors had gain or loss of the Y chromosome, sometimes one clone with +Y and another with -Y, as the sole anomaly, whereas the remaining 33 all carried structural rearrangements and usually were cytogenetically complex with multiple aberrations. The chromosomal bands most frequently involved were, in decreasing order of frequency, 8p11-q11, 1p11-q11, 3p11-q11, 11q13, 13p11-q11, 1p13, 5p11-q11, 7p11-q11, 15p11-q11, and 14p11-q11. Almost one-half of the breakpoints were located in centromeric or juxtacentromeric bands. Recurrent aberrations included i(8q), i(5p), i(1q), del(3)(p11-12), del(5)(p11), t(1;1)(p13;q25), and der(14;15)(q10;q10). To see whether the karyotypic features of head and neck squamous cell carcinoma differ depending on exact tumor site, we added to the present series our previously published 23 karyotypically abnormal head and neck squamous cell carcinomas that had been cultured in the same way as the tumors of the present series. In the ensuing correlation analysis, tumors of the oral cavity and oropharynx and hypopharynx were found to share many features: highly complex karyotypes were frequent, often containing isochromosomes such as i(8q) and i(5p), and also rearrangements of 11q13 (often as homogeneously staining regions) and loss of genetic material from the short arms of chromosomes 3, 13, 14, and 15 were repeatedly seen. Laryngeal carcinomas, on the other hand, often had simple karyotypic changes.

Light microscopic autoradiographic study of DNA synthesis in cecal epithelial cells of aging mice.

The localization and activity of DNA synthesis of mouse cecal epithelia from prenatal Day 19 embryos to 12-month adults were investigated by light microscopic autoradiography after [3H]-thymidine injection in vivo. The cecal epithelial labeled cells with [3H]-thymidine were located at the lower half of the crypts, and the labeling index (LI) changed with the development of the cecum due to aging. A peak in the LI of absorptive cells was found at embryonic Day 19, but it decreased strikingly at the first postnatal day and then kept an almost constant value until 12 months. The LI of the goblet cells showed the highest value at fetal Day 19, decreased gradually with aging from the first postnatal day, and completely disappeared from 1 month onwards. The basal granulated cells were located only in the base of crypts, and the significant difference of LI was not found from embryonic Day 19 through 12-month adult. The results obtained in this study suggest that there are some differences of DNA synthesis among different kinds of cecal epithelial cells from embryonic stages to postnatal stages as a result of aging.

Urinary Phenylglyoxylic Acid Excretion after Exposure to Ethylbenzene among Solvent-exposed Chinese Workers.

Factory surveys were conducted in the second half of work weeks on 360 solvent workers (202 men and 158 women) and 281 controls in China. Monitoring personal exposures showed that ethylbenzene exposure was low (geometric mean 1.8 ppm) and was accompanied by coexposure to toluene (1.5 ppm) and three xylene isomers (6.7 ppm). Urine samples collected at the end of the eight-hour shift were analyzed for phenylglyoxylic and mandelic acids by high-pressure liquid chromatography at 257 nm. Despite the low level of the exposures, a significant correlation was observed between ethylbenzene exposure and urinary phenylglyoxylic acid, with high (0.6-0.7) correlation coefficients, suggesting that urinary phenylglyoxylic acid is a good marker of occupational exposure to ethylbenzene. Mandelic acid also correlated with ethylbenzene exposure, but with much smaller coefficients (0.2), possibly because the method employed was more sensitive to phenylglyoxylic acid than to mandelic acid.
