Detection by electron microscope of rod-shaped organisms in synovial membrane from a patient with the arthritis of Whipple's disease.

Rod-shaped organisms identical to those present in the jejunal mucosa have been found in the synovial membrane of a patient with Whipple's disease. These probably caused inflammatory changes which sere reflected in an increase of the cellular content and high enzyme levels (acid phosphatase and 5-nucleotidase) of the synovial fluid. Tetracycline was effective in controlling the bowel lesion but only had a temporary effect upon the arthritis. Erythromycin controlled both the bowel lesion and the arthritis.

Acetyl coenzyme a-glutamate acetyltransferase and N-acetylornithine-glutamate acetyltransferase of chlorella.

The enzymic formation of acetylglutamate has been studied in Chlorella vulgaris extracts. Acetyl CoA and N(2)-acetyl-l-ornithine served as substrates for glutamate acetylation whereas acetylphosphate, N(5)-acetyl-l-ornithine, and N(2)-acetyl-2,4-diamino butyrate were ineffective. Acetyl CoA-glutamate transacetylase and acetylornithine-glutamate transacetylase activities have been purified over 180-fold with no indication of any separation of activities. The acetyl CoA activity was more labile than acetylornithine activity so that preparations having acetylornithine-glutamate transacetylase activity but no acetyl CoA-glutamate transacetylase activity were obtained. The two acetylating activities appear to be properties of one enzyme with one portion more easily denatured. Both acetylating activities had pH optima between 8 and 8.5. The Km value for glutamate was 3 mm for both activities. The Km values were 0.2 mm for acetylornithine and 3.2 mm for acetyl CoA. Arginine inhibited acetyl CoA-glutamate transacetylase (Ki = 0.94 mm) and acetylglutamate phosphokinase (Ki = 0.5 mm) but had no effect on acetylornithine-glutamate transacetylase. The lack of an inhibitory effect of proline on any of the three enzymic activities indicates that acetylglutamate is not a normal intermediate in proline biosynthesis. Growth of Chlorella with arginine as a nitrogen source had no effect on enzyme levels, showing that end-product repression is not a control factor in arginine biosynthesis in Chlorella. In Chlorella, arginine controls its own biosynthesis by inhibiting acetylglutamate phosphokinase and controls the level of acetylated intermediates by inhibiting acetyl CoA-glutamate transacetylase.

Molecular-sieve chromatography and electrophoresis in polyacrylamide gels.

1. The absolute electrophoretic mobilities of eight proteins have been measured at pH8.76, I 0.05, in polyacrylamide gels of 20 different compositions at 10 degrees C. 2. The partition coefficients of these proteins have been determined chromatographically under the same conditions by using columns of granulated polyacrylamide gel prepared simultaneously. 3. The electrophoretic mobilities are an exponential function of the gel concentrations when the latter are corrected for water uptake. The constants of this function have been determined by curvefitting methods. They have been shown to be related to the free solution mobility and to the mean molecular radius respectively. 4. The reduced mobilities have been shown to be a linear function of the partition coefficients by statistical analyses. 5. The physical significance of the relation between electrophoretic mobility and chromatographic phase distribution in gel media is discussed in the context of these results.

Histochemically demonstrable fibre abnormalities in normal skeletal muscle and in muscle from carriers of Duchenne muscular dystrophy.

Deltoid muscle was removed at motor point biopsy from 10 female relatives of patients with Duchenne muscular dystrophy and from seven others, with no evidence of neuromuscular disease. Transverse cryostat sections of the muscle from each case were stained for reduced diphosphopyridine nucleotide diaphorase and it was found that all contained varying numbers of degenerating type 1 fibres. The percentage of abnormal fibres in the type 1 fibre population was then calculated for each case and it was found that the muscles from the patients with dystrophic relatives contained considerably higher percentages of abnormal fibres, which also showed more severe degeneration, than did the muscles from the normal cases. There was no absolute correlation between serum creatine kinase levels and degree of pathological change, though patients with the most severe changes in their muscles had abnormally high serum creatine kinase levels. It is suggested that histochemical studies could be a useful addition to the present diagnostic tests for the carrier state in Duchenne muscular dystrophy.

In vitro staining of intramuscular nerve endings.

A method is described for vital staining of the human intramuscular nerve endings after the muscle biopsy has been removed from the patient. The muscle is transferred from the site of operation to the laboratory in oxygenated physiological saline. Methylene blue solution (0·015%) is injected into the muscle strip, and development of the stain can be observed under the microscope after exposure of the muscle to air. Permanent squash preparations can be made of the stained muscle and these are identical in appearance to muscles stained by injection of methylene blue in vivo.

Metabolism of Glutamic Acid and N-Acetylglutamic Acid in Leaf Discs and Cell-free Extracts of Higher Plants.

Radioactive glutamic acid and N-acetylglutamic acid have been incubated with normal and wilted leaf discs and radioactivity recovered in uncombined proline. The discs which had been placed under moisture stress incorporated considerably more label in uncombined proline than did normal discs. In extracts of swiss chard leaves, both (14)C-N-acetylglutamic acid and glutamic acid are metabolized to their corresponding semialdehyde as evidenced by a recovery of proline after appropriate treatment. There is also incorporation of label in N(a)-acetylornithine from N-acetylglutamic acid. These reactions require ATP, Mg(2+), and NADH or NADPH.
