In a recent nosocomial outbreak of Pseudomonas pickettii infection at the general hospital of Corbeil-Essonnes, France, two patients were moderately affected, but two others, who were diabetic, died of septicemia. The infection was transmitted by 0.05% aqueous solutions of chlorhexidine, the treatment of water for these solutions being the cause of contamination. The ion-exchange resin in the deionization cartridges allowed the surviving bacteria in the city water supply to grow while the bacteriological filters downstream only lowered the contamination level. The 0.05% chlorhexidine aqueous solutions failed to prevent growth up to 4 X 10(6) bacteria/ml. P. pickettii is resistant to aminoglycosides and colistine.
A model incorporating the metal chelating moiety of bleomycin and an anilinoacridine ring able to intercalate in DNA has been synthesized. The copper(II) complex of that molecule has been studied using circular dichroism and electron spin resonance by comparison with bleomycin. The introduction of the anilinoacridine ring involves a modification in the geometry of the complex. A distortion of the square-pyramidal form (type II complex) gives rise to a type I complex in which the metallic atom is drawn out of the plane of the four square-planar ligands and displaced slightly towards the fifth ligand.
The present case report is on a patient treated for tubercle in 1949 by an artificial pneumothorax. Thirty three years after the initial illness some radiological changes occurred suggesting a recurrence, especially as a bacteriological examination showed the presence of acid-alcohol fast bacilli on direct smear. Triple therapy (Isoniazid, Rifampicin, Streptomycin) was then prescribed, and was followed by a radiological improvement, even though the sputum remained positive. After multiple seedings, it was possible to isolate from 11 tubes and 2 expectorations a rare strain of mycobacterium: mycobacterium malmoense. The present observation seems to be the first indisputable French case of infection by this bacterium. From the opportunity presented by this observation the bacteriological characteristic of this mycobacterium, as well as the clinic aspects of the infection are discussed.
Vasoactive intestinal peptide (VIP) was found to cause a dose-dependent decrease in fructose 2,6-bisphosphatase concomitant with an increase in cyclic AMP in cultured HT29 cancer cells from human colon. The maximum effect was a 41% decrease obtained with 10 nM-VIP, and half-maximum effect was obtained with 0.75 nM-VIP. The effect of 2.5 nM-VIP was almost totally counteracted (i.e. fructose 2,6-bisphosphate concentration was restored) by either adrenaline (1 microM) or the alpha 2-adrenergic agonist UK-14304 (1 microM); the alpha 2-agonist clonidine (1 microM) was less efficient, since the VIP effect was decreased by 72% only. The adrenaline effect was totally antagonized by 1 microM-yohimbine. It is concluded that, in the HT29 cancer cells, the fructose 2,6-bisphosphate-producing system is sensitive to variations of cyclic AMP concentration and is under the dual control of VIP and alpha 2-adrenergic receptors.
Under the standard conditions of culture, Fru-2,6-P2 level in HT29 cells is transitorily increased as a consequence of medium change; the peak value occurs after 2 hr, followed by a gradual return to a basal value (40 pmol/mg protein) which is maintained as long as medium glucose concentration stands above 2 mM. A 20 hr glucose deprivation lowers Fru-2,6-P2 level to trace value, but, when glucose is reintroduced, the peak value is much higher; large Fru-2,6-P2 accumulation is correlated with higher rates of glucose uptake and lactate release, which suggests an activation of glycolysis at the level of phosphofructokinase-1. Fru-2,6-P2 level depends on the glucose concentration within the range of 0 to 5 mM. At this concentration and above, maximal effect is reached. Previous glucose deprivation renders the Fru-2,6-P2 forming system more sensitive to glucose. When given instead of glucose, fructose enters the glycolytic pathway and produces same effect as glucose on the Fru-2,6-P2 level. Galactose turns it to almost zero which coincides with low glycolytic rate. Acidity of the culture medium favorishes the Fru-2,6-P2 formation; however, change in pH cannot explain the variations of Fru-2,6-P2 level observed under the standard culture conditions. Lactate concentrations over 10 mM in the medium are found to significantly inhibit the Fru-2,6-P2 producing system. Therefore, lactate accumulation in the medium could be an important factor controlling Fru-2,6-P2 level during standard cell culture.
Effect of glutamine deprivation (GLN- medium) and of its replacement by 4mM ammonium chloride (GLN-/NH4+ medium) or by 4mM glutamate (GLN-/Gt+ medium) was studied on growth rate, morphology and metabolism of HT29 human colon cancer cells. Growth rates were modified as follows: at the first passage, growth of GLN- cells was strongly decreased (doubling time: 192 hr vs 32 hr in control cells grown in GLN+ medium); GLN-/NH4+ cells and GLN-/Gt+ cells were found to have doubling times of 72 and 70 hr, respectively. At the 8th passage, doubling times were decreased in all cases, being: 144 hr for GLN- cells, 60 hr for GLN-/NH4+ cells and 24 hr for GLN-/Gt+ cells, which indicates a capacity of adaptation of the cell-line to new culture conditions. GLN- cells and GLN-/NH4+ cells were found to exhibit an enterocytic type of differentiation (polarization of the cell layer with apical and cystic brush border and tight junctions); GLN-/Gt+ cells remained undifferentiated and comparable to control GLN+ cells. Glycogen level varied according to the phases of the culture, with a trend to lower level in glutamine deprived cells; glucose uptake and lactate production varied as a function of the medium composition and of the phases of the culture. At the 8th passage, all the glutamine deprived cells produced less lactate than control; GLN-/Gt+ cells were found to utilize less glucose than others.
Two groups of human subjects were submitted to a 20-week endurance training program (1 h a day, 4 days a week, 70-80% max VO2). The first group (G20) consisted of eight 22 +/- 3 years male students, the second group (G60) was composed of seven still very physically active elderly male subjects (62 +/- 4 years). Training significantly increased max VO2 by 15% in G20 and 7% in G60. Muscle samples taken from the vastus lateralis muscle before and after training were histochemically stained for fibre-typing (myofibrillar ATPase), capillary supply and fibre area measurements (amylase PAS and NADH-TR). Fibre-type distribution was unchanged with training. Capillary density (cap X mm-2) increased significantly in both groups from 316 +/- 42 to 396 +/- 73 in G20 and from 308 +/- 48 to 409 +/- 55 in G60. This enhancement of capillary supply was linked to the proliferation of capillaries in G20 where the number of capillaries in contact with ST and FTa fibres (CC) significantly increased from 4.6 to 5.9 and from 4.8 to 6.1 respectively. No significant changes in fibre areas were found in G20. On the contrary, G60 did not show any significant sign of capillary growth (CC unchanged) whereas fibre areas significantly decreased in ST (6,410 to 5,520 micron 2) and FTa fibres (5,830 to 5,090 micron 2). A methodological evaluation of fibre-area measurement was described, with confirmation of the data. It was concluded that this study may illustrate the trainability of skeletal muscle of elderly men in a possibly different way to that seen in a younger age group.
Some aspects of carbohydrate metabolism were investigated in three non-malignant, glycogen storing, cell lines derived from a primary culture of rat hepatocytes, and in the Morris hepatoma 3924 cells. The three cell lines show biochemical alterations which are, to a large extent, similar to those found in the hepatoma cells: increased activity of glycolytic enzymes and decreased activity of gluconeogenetic enzymes. An increase of glucose-6-phosphate dehydrogenase activity is also found. The three cell lines, as the Morris hepatoma cells, actively convert glucose into lactate under the in vitro conditions of culture. Fructose is not taken up as quickly as glucose and galactose is not metabolized. As compared with normal hepatocytes, the three cell lines have altered metabolism and growth behaviour. They largely resemble the preneoplastic cells appearing in rat liver at the early stages of experimental carcinogenesis.
Six healthy subjects, aged 35.8 +/- 4.4 years, volunteered to participate in a 40-week training program on a bicycle ergometer [three 60-min sessions per week at 80%-85% of maximal oxygen uptake (VO2 max)]. Before training and at the 10th, 20th, 30th, and 40th weeks of the training program, plasma testosterone, cortisol, and androstenedione concentrations were measured at rest (t0) and at the end (t60) of a 1-h endurance exercise requiring 85%-90% of VO2 max. Training resulted in significant increases of anaerobic threshold (12.6%) and VO2 max (7.3%). The training program did not significantly alter the resting values of plasma testosterone, androstenedione, and cortisol; in contrast, the exercise responses (delta = t60-t0) of testosterone, androstenedione, and cortisol were increased. The highest amplitude of these responses was reached at the 30th week for cortisol and androstenedione and at the 40th for testosterone. These results suggest that long-term training enhances both testicular adrenal and responses to endurance exercise. The possible role of hormonal changes in the mobilization of energy substrates during exercise is discussed.
A simple technique was elaborated for the measurement of muscle lactate concentration. It was tested on samples of muscle vastus lateralis taken by the Bergström (1962) needle biopsy technique at the end of 20 min exercise bouts corresponding to 60-70% of Vo2 max. The biopsies were freshly frozen in liquid nitrogen, powdered and weighed in a cryostat at -20 degrees C. The extraction were made by saponine and the lactate measured in the saponine solution by an electrochemical-enzymatic method (LA 640). The results concern: the time of taking the biopsies and the freezing time (27 +/- 11 s and 34 +/- 9 s respectively); the accuracy of weighing (inducing a 1% uncertainty in the final result); a comparison of the saponine extraction with the perchloric acid extraction and a checking of the extraction capacity of the former; the accuracy of the whole measurement (the mean relative confidence limits are +/- 8-10%; the reproducibility of the technique through measurement of the variation coefficient (18%) calculated on measurements performed at a 15 days interval in 6 trained subjects. The discussion of the results and their comparison with those of the literature lead to the conclusion that the described method is suitable for muscle lactate measurements.
Activity of several enzymes of the glycogen and carbohydrate metabolism is studied in HT 29 colon adenocarcinoma cell line and in HT 29 tumors developed in nude mice, by reference to the normal human colon mucosa. Activity of glycogen synthase, glycogen phosphorylase, pyruvate kinase, fructose-1,6-diphosphatase, glucose-6-phosphate dehydrogenase and lactate dehydrogenase is found to be increased in both the cultured cells and the tumors. It indicates that the biochemical strategy of malignant cells, due to the neoplastic transformation process, involves specific changes in the carbohydrate metabolism of tumor as well as in vitro growing correspondent cell line.
This study examined the effect of a 20-week training program of two groups: six middle-aged men (37 +/- 4 yrs) (GIT) and six young male subjects (20 +/- 1 yrs) (GIIT). The training program consisted of bicycle ergometer exercise, 1 h/day, 3.5 days/week at a work load corresponding to 80%-85% of HR max. Before (S0) and at the end of the training program (S20), measurements of VO2 max, maximal work load (MWL), net efficiency, onset of blood lactate accumulation, absolute (OBLAW), and relative to MWL (OBLA %) were made on GIT and GIIT groups and on a third group (21 +/- 2 yrs) (GIIC), used as a control. Muscle fiber composition of m. vastus lateralis was studied after training for GIT and before and after the training period for GIIT and GIIC. VO2 max (ml X kg-1 X min-1), which was initially similar in GIT and GIIT (49 ml X kg-1 X min-1), increased significantly by 8% in GIT and by 19% in GIIT. OBLAW increased significantly to the same level in the two groups (38% and 42%, respectively). OBLA % increased significantly (20%) in GIT only. In the groups studied (GIIT), no change was observed for muscle fiber composition. % ST fiber type did not correlate to OBLAW or OBLA % S0 values nor to OBLAW and OBLA % changes during training. This leads to the conclusion that age and the initial physical fitness were the two major factors affecting the outcome of this endurance training program upon the two groups. Further research is needed to establish which of these two factors is the most influential.
Actinomyces israelii was isolated from cervico-facial subacute or chronic infections (9 cases), and abdominal wall abscess (1 case). Two conditions were necessary for successful isolation of A. israelii: properly collected clinical specimen (pus aspirated from abscess or biopsy) and prolongated incubation of anaerobic cultures (at minimum 5 days). Two commercially available micro-method multitest systems (Api 20 anaerobes, Api Zym test) were compared with standard biochemical procedures. The three methods were evaluated as to the reproducibility and help in identifying anaerobic non spore-forming Gram-positive bacilli in a routine clinical laboratory.
In vitro activities of four cephalosporins, i.e. cefoperazone, cefsulodin, ceftazidime and CM 40874, on 31 ticarcillin-susceptible or resistant Pseudomonas aeruginosa strains were compared. Minimal inhibitory and bactericidal concentrations were determined by the conventional broth technique and by a filter membrane transfer method on agar. MICs determined by both methods were comparable for each strain. Some discrepancies were found for minimal bactericidal concentrations. Among the cephalosporins studied on ticarcillin-susceptible Pseudomonas aeruginosa strains, the most active was ceftazidime, followed by cefsulodin, CM 40874 and cefoperazone. Resistance of Pseudomonas aeruginosa to ticarcillin was associated with an overall increase in resistance to the four cephalosporins studied. These cephalosporins can be classified according to bacteriostatic and bactericidal effect in the following decreasing order: ceftazidime, CM 40874, cefsulodin and cefoperazone.
This research concerns the bacteriostatic and the bactericidal activity of two beta-lactams (cefsulodin and ceftazidime) on ten strains of Pseudomonas aeruginosa affected by carbenicillin. Additionally to classical approaches in the broth and agar technics, a method of culture on filter membrane is used. The results obtained after a 2 hours and 24 hours contact are reported. The resulting values of minimum inhibitory concentrations are comparable. As far as minimum bactericidal concentrations values are concerned, there is no significant difference between the two methods or between the two antibiotics. The analysis of the bactericidal effect of a 2 hours contact allows no inference relative to the comparative effect of the two antibiotics. Each strain of P. aeruginosa has its specific behaviour, and there seems to be no way to extrapolate from their minimum inhibitory concentrations the bactericidal effect of cefsulodin and ceftazidime.

The effect of a 40-week training program on the anaerobic threshold (AT) was studied in five subjects (35 +/- 5 yrs). The training program consisted of a bicycle ergometer exercise 1 h per day 3 days a week at a work load corresponding to 80%-85% of VO2 max. Before training (S0) and at the 10th, 20th, 30th, 40th weeks (S10, S20, S30, S40) of the training program, ventilatory AT (AT vent), lactate AT (AT lact), and 4 mmol AT were estimated using a graded exercise test. In the same period, another test (1) to determine VO2 max, maximal work load (MWL), and net efficiency (2) and a 1-h endurance exercise requiring 85%-90% of VO2 max were performed. After training, AT increased significantly by 10%, 11%, and 18% in AT vent, AT lact, and AT 4 mmol, respectively. MWL and net efficiency increased by 22% and 14% respectively, and there was no significant increase in VO2 max. The AT increase at S20 was delayed as compared to the change in MWL (S10). It is hypothesized that an AT increase provides good evidence for modifications of the muscular oxidative capacities during an endurance training program. A part of this paper is devoted to (1) the study of the reproducibility of AT estimation, (2) a comparison to other methods for determining the definition of AT, and (3) the correlations between the three methods utilized for AT estimation.
