Conventional and rapid methods for yeast identification.

Identification of yeasts depends on data obtained from morphological features and physiological characteristics. This article reviews the commonly used conventional methods for the identification of yeast to set the stage for a critical review of rapid systems for yeast identification. Comparative analyses of percentage agreement of API 20C, Uni-Yeast Tek, Minitek, Mycotube, Micro-Drop, Auxodisk, Iatron, Autobac 1, AMS, Abbott MS-2, and Abbott Quantum 11 against the conventional method and, in some cases, against each other are tabulated. Gas liquid chromatography method, enzyme method, MUG, and the Fung's mini-system are also mentioned as possible rapid methods for yeast identification. The purpose of this review is to update the rapidly expanding information concerning diagnostic kits and systems for yeast identification and analyze some of the advantages, disadvantages and potential uses of rapid systems of yeast identification.

Impaired natural killer cytotoxicity during recrudescence of recurrent herpes simplex virus type 1 infection.

We investigated natural killer (NK) cytotoxicity in patients with recurrent herpes infection by using K562 cells infected with Herpes simplex virus type 1 (HSV-1) as target. The NK cytotoxicity was lower than during convalescence and in controls during recrudescence (0 to 3 days postonset of clinical symptoms; P less than 0.001). In the convalescent phase (4 to 14 days postonset of symptoms) the NK cytotoxicity was significantly higher than in controls (P less than 0.05). These results suggest that low NK cytotoxicity is related to the recurrence of herpes.

Vascular ectasia of the duodenum detected by duodenoscopy.

A 65-yr-old Chinese woman suffered from repeated episodes of upper gastrointestinal bleeding for 5 yr. Despite repeated panendoscopic and angiographic studies, the bleeder was not found until a duodenoscopic examination was performed. A small bleeder was found near the papilla vater area. The lesion was excised and proved to be a vascular ectasia of duodenum.

[Study of the natural killer cell activity in lung cancer patients].

A total of 50 lung CA patients and 20 normal persons as control were included in our study. We obtained lymphocytes from heparinized blood by Ficoll-Hypaque gradient method and after incubation with Cr51 labelled K562 cells at concentrations of 100: 1, 50:1, 12.5:1 for four hours, we calculated the cytotoxicity activity of NK cells. Significant impairment was noted in the comparison of NK cytotoxicity between stage III-M1 lung CA patients (n = 28) and the control group (n = 20) (E/T; 50:1, 29.55 +/- 24.20% vs 49.83 +/- 14.46%, p less than 0.001; E/T: 25:1, 10.23 +/- 7.19% vs 27.66 +/- 6.06%, p less than 0.001). 4 cases in stage I underwent surgery, the NK cytotoxicity before the operation was noted to be low, E/T: 25:1, 9%, 11%, 14% and 19% respectively. After the operation, the NK cytotoxicity was increased to 33%, 35%, 42% and 44% respectively. 3 stage I lung CA patients has continuously followed up, 2 relapsed cases had decreased NK cytotoxicity to 22% and 25%. These results indicated that the NK cytotoxicity assay was impaired in stage III-M1 lung CA and also suggested that it may act as a therapeutic guide for the results of the operation.

Studies on growth and distribution of Angiostrongylus cantonensis larvae in Ampullarium canaliculatus.

The intramolluscan development of Angiostrongylus cantonensis larvae in Ampullarium canaliculatus, a fresh-water migrant snail which multiplies in tremendous rate in Taiwan, was followed for 28 consecutive days in this study. The snails were infected with first stage larvae of A. cantonensis and groups of three snails were sacrificed daily to study various larval stages in different organs. A. cantonensis larvae were found in all snails examined. During the first four days, they were obtained only from the digestive tract. Mouth is thus suggested as the primary route of natural infection. The first molt was observed on the fifth day of infection, and the second molt, the 12th day. Molting occurred in almost all kinds of organs examined. Studies on tissue tropism showed that the predilection site of A. cantonensis in this freshwater snail was headfoot region. Relatively few larvae were found in visceral organs including liver, heart, and ovary. Most parasites, 62.1%, recovered on the 28th day of infection were third-stage larvae.

4;11 translocation-associated acute leukemia: a comprehensive analysis.

A girl with common acute lymphoblastic leukemia associated with t(4;11)(q21;q23) is presented. The physical examination revealed hepatosplenomegaly, and the hematologic analysis showed a high count of white cells. Chemotherapy induced a complete remission (CR), and the patient is still in CR (8 months). At the present time, 43 cases (including ours) of acute leukemia with t(4;11) have been published. This disease more often affects children and women than adult men and is characterized by a high blood white cell count and a short survival. The t(4;11)-associated acute leukemia is a heterogeneous group, including acute undifferentiated leukemia, acute undifferentiated lymphoblastic leukemia, and "common" acute lymphoblastic leukemia.

The occurrence of the 15;17 translocation in acute promyelocytic leukemia.

For many years, acute promyelocytic leukemia (APL) has been associated with the 15;17 translocation. However, questions concerning an eventual geographic distribution have been raised by the Second and Fourth International Workshops on Chromosomes in Leukemia. Using a statistical approach, we showed that, even if the techniques of processing play an important role in detecting the 15;17 translocation, a geographic distribution exists, which is difficult to explain. Finally, we discuss the possible role of c-erb A1 in acute promyelocytic leukemia.

An outbreak of N-hexane induced polyneuropathy among press proofing workers in Taipei.

The objective of this study was to determine the prevalence and the etiology of polyneuropathy observed among press proofing workers in Taipei. Neurological examinations of 59 workers, from 16 press proofing factories, were conducted. Fifty-four of those workers subsequently underwent studies of nerve conduction velocities. Samples of bulk solvent from the involved factories were analyzed for their contents using gas chromatography. Fifteen (25%) of the study group were found to have polyneuropathy. All 15 patients with polyneuropathy were from factories in which solvents containing n-hexane were regularly used, and there was a significant association between n-hexane concentration in the bulk samples and prevalence of polyneuropathy. The air concentration of n-hexane in one factory in which all six employees developed polyneuropathy was 190 ppm. Workers who were exposed to n-hexane at air levels of less than 100 ppm but who frequently worked overtime showed a significant slowing of motor nerve conduction velocities on median, ulnar, and peroneal nerves. Of 13 workers who regularly slept in the factory, 12 (92%) had polyneuropathy compared to three (7%) of 46 workers who did not sleep in the factory. The outbreak of polyneuropathy was attributed to a combination of the use of solvents with high contents of n-hexane, poor ventilation, and the practice of sleeping in the factories between shifts.
