[Expression of cytidine deaminase mRNA in bone marrow cells from patients with acute leukemia].

The aim of the study was to determine the expression of cytidine deaminase (CDD) gene in bone marrow cells from patients with acute leukemia (AL) and evaluate the relationship between CDD expression and clinical feature. Semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) was used for detection of expression level of CDD mRNA in bone marrow cells from 83 patients with acute leukemia and from 15 healthy peoples as control. CDD/beta-actin ratio >or= 0.5 was considered to be positive. The results showed that expression levels of CDD of 31 previously untreated patients were higher than those of 23 cases of AL in complete remission and of normal controls. Expression levels of CDD of 29 relapse/refractory patients were also higher than those of 23 AL patients in complete remission and of normal subjects. The expression levels of CDD in relapse/refractory ALL were higher than those in AML while expression levels of CDD were not correlated with the outcome of therapy. It is concluded that the level of CDD mRNA expression varies at the different stage of acute leukemia. The expression level of CDD seems not to be a prognostic factor.

[Treatment of 54 chronic myelogenous leukemia with Gleevec].

OBJECTIVE: To evaluate the efficacy and safety of Gleevec (Imatinib) in the treatment of patients with Ph positive chronic myeloid leukemia in chronic phase (CML-CP). METHODS: A total of 54 CML-CP patients in whom previous therapy with interferon-alpha had been failed or untolerated, or relapsed after allogeneic stem cell transplantation (allo-SCT) were treated with 400 mg/d of oral Gleevec for 6 to 11 months. RESULT: Fifty-three patients being able to evaluate achieved complete hematological response within 7 to 28 days. Fifty-two (98%) of them remained in this situation at last follow-up. One patient relapsed after 7 months' treatment, and progressed to accelerated phase. Gleevec induced major cytogenetic response in 37 patients (70%) and complete cytogenetic response in 27 (51%). Twenty-nine of 37 patients (78%) achieved major cytogenetic response within 3 months. Grade 3 neutropenia or thrombocytopenia occurred in about 10% of patients, which were manageable or tolerated. Grade 3 or 4 nonhematologic adverse effects were infrequent. Only 1 patient (2%) discontinued treatment because of drug-related adverse events. CONCLUSIONS: Gleevec induced high rate of cytogenetic and hematologic responses in patients with CML-CP who failed in previous interferon therapy. The adverse effects were mild and manageable, or no need for treatment.

[Influence of external electric current on the spatial distributions of porosity and fractional dimension of biofilms].

The spatial distributions of porosity and fractional dimension of autotrophic and heterotrophic/autotrophic biofilms applied by direct current in the range of 0-25 A/m2 were investigated by using the micro-slicing technique and image analysis. The results showed that the porosities of the autotrophic and heterotrophic/autotrophic biofilms respectively decreased from ca. 92% and 96% in the top layer to ca. 54% in the bottom layer while the fractional dimensions increased from 1.1 and 1.05 in the top layer to 1.4 in the bottom layer in the absence of current. The increase in the current density decreased the porosities and increased the fractional dimensions in the top layers. In the deeper layers, the current had insignificant influence on the porosity and fractional dimension.

[Evaluation on the effects of community-based anti-smoking intervention].

To explore effective antismoking intervention measures, measures including flyers and booklets as major mass media, school-based health education and follow-up to smokers were carried out in Beijing, Shanghai and Changsha urban communities. The changes of smoking behavior in population were evaluated by quasi-trial designation. Results showed that smoking rates in adult male, female and both genders decreased by 5.54%, 2.61% and 2.94% respectively and were statistically significant in intervention community. Compared with control community, the net decrease of smoking rates in adult male, female and both were 4.90%, 3.37% and 3.51% respectively. The net increase of quitting rates in adult male, female and both were 4.35%, 17.19% and 6.30% respectively. The net changes in female and in both genders of smoking and quitting were all statistically significant. It was suggested that comprehensive intervention measure with health education as the core might be the priority for the effective community-based antismoking activity. The policy-making and behavior encouraging activities should be focused.

[Preparation of bovine serum albumin nanoparticles surface-modified with glycyrrhizin].

AIM: To study the preparation of bovine serum albumin nanoparticles surface-modified with glycyrrhizin(BSA-NP-GL) targeting to hepatocytes. METHODS: The bovine serum albumin nanoparticles (BSA-NP) were prepared by desolvation process. Glycyrrhizin (GL) was oxidized by sodium periodate to be conjugated to surface reactive amino groups (SRAG) of the BSA-NP. The SRAG were quantified by spectrophotometric method using 2, 4, 6-trinitrobenzenesulfonic acid(TNBS). Glycyrrhetinic acid(GA) hydrolyzed from GL, which was on the surface of BSA-NP-GL was assayed by HPLC after isolation by sephadex G-50. Both methods were used to verify the conjugation achieved. HPLC was used to determine surface density of GL on BSA-NP-GL. RESULTS: The amount of SRAG of the BSA-NP-GL decreased by 19.6% compared with normal BSA-NP. The amount of GL molecule was 9.2% of the total determined SRAG of BSA-NP. The mean diameter of the BSA-NP-GL was 73 nm with round shape. The stability of BSA-NP-GL was constant when it was stored at 25 degrees C and 37 degrees C during 10 days. CONCLUSION: BSA-NP-GL was successfully prepared, which is considered to establish an experimental foundation for further research on its ability for targeting to hepatocytes.

Tetra-arsenic tetra-sulfide for the treatment of acute promyelocytic leukemia: a pilot report.

In the past 6 years, we treated 129 patients who had acute promyelocytic leukemia (APL) with a new arsenic agent, oral tetra-arsenic tetra-sulfide (As(4)S(4)). Nineteen of the patients had newly diagnosed APL, 7 had first relapse, and 103 had hematologic complete remission (HCR). HCR was achieved in all patients with newly diagnosed APL and in all those with hematologic relapse. Of 16 patients with newly diagnosed disease and available cytogenetic and molecular analyses, 14 had cytogenetic and molecular complete remission (CR). Cytogenetic and molecular CR was also obtained in 5 of the 7 patients with hematologic relapse. In the HCR group, 35 of 44 patients positive for PML-RARalpha at baseline became negative. In the newly diagnosed group, estimated disease-free survival (DFS) rates for 1 and 3 years were 86.1% and 76.6%, respectively, with a median follow-up time of 13.5 months (range, 2-40 months). In the HCR group, DFS rates for 1 and 6 years were 96.7% and 87.4%, respectively, with a median follow-up of 23 months (range, 2-71 months). Treatment with As(4)S(4) was well tolerated, with only moderate side effects, including asymptomatic prolongation of corrected QT interval, transient elevation in liver enzyme levels, rash, and mild gastrointestinal discomfort; neither myelosuppression nor appreciable long-term side effects occurred. Degeneration or apoptosis of APL promyelocytes was observed during As(4)S(4) therapy. Pharmacokinetic studies showed that the agent was absorbed rapidly. Most urinary arsenic excretion occurred within the first 24 hours. Both blood and urinary arsenic levels declined after discontinuation of As(4)S(4). Our results show, for the first time, that As(4)S(4) treatment alone is highly effective and safe in both remission induction and maintenance therapy in patients with APL, regardless of disease stage.

[A quantitative assay for telomerase activity in peripheral blood mononuclear cells from patients with acute leukemia].

To establish a quantitative assay for telomerase activity and analyze the telomerase activity in peripheral blood mononuclear cells (PBMNC) from patients with acute leukemia, a fluorescent dye, PicoGreen, was added to the products after telomere repeat amplification protocol. The samples were excited at 480 nm and the fluorescence emission intensity was measured at 520 nm using a spectrofluorometer. Telomerase activity was detected in PBMNCs from 20 cases of normal individuals and 25 patients with acute leukemia. The results showed that the fluorescence of PicoGreen binding to double-stranded DNA specifically was enhanced with increase of DNA quantities. In conclusion, the met hod is rapid, simple and quantitative, the telomerase activities of PBMNCs from acute leukemia patients are significantly higher than that of the normal controls.

[Changes of both knowledge levels and health-related behaviors of preventing cardio-cerebro-vascular diseases in population after intervention].

Comprehensive intervention measures for preventing cardio-cerebro-vascular diseases of community population were carried out in Beijing, Shanghai and Changsha urban areas, and then was evaluated to its effects on knowledge, attitude and behavior (KAB) levels and health-related behaviors of population by quasi-trial designation. Results showed that the net KAB leveling were separately 9.89 in 1995, 4.74 in 1997 and 10.66 in 2000 in comparison with base investigation, and statistically significant difference was found. Of 6 health-related behaviors, caring to health knowledge, being influenced by health knowledge and exercising were significantly influenced, next controlling salt consumption. No change of smoking and alcohol drinking was found. Among different target population, these changes among women were higher than man as well as at home or retired than at work, and hypertension than normtension. As aging and education time decreasing, both absolute and relative effects of health education were better. It suggests that comprehensive intervention with health education as its main measure promote KAB levels and transfer health-related behaviors of population. It is great important to preventing cardio-cerebro-vascular diseases of community population.

[Evaluation of effects of health education on prevention of cardio-cerebro-vascular diseases in community population].

A comprehensive community-based intervention with health education as the major measure for preventing cardio-cerebro-vascular diseases in selected urban residents in Beijing, Shanghai and Changsha cities during the periods of the ninth "five-year" plan (1996-2000) was conducted, and its effects on knowledge, attitude and behavior (KAB) changes were evaluated by quasi-trial designation. Results show that the net KAB level increases by 6.01 scores in three cities and by 6.12 scores in patients suffering from hypertension after intervention, and these changes in scores are statistically significant. The net percentage of hypertension patients who took blood pressure measurement regularly within every 3 months and the net percentage of them who took anti-hypertension drugs regularly for therapy increased by 9.65% and 7.33% respectively after the intervention and the former increased was statistically significant. It is concluded that health education may promote the improvement of knowledge, attitude and behavior changes of participants and is of great importance for preventing cardio-cerebro-vascular diseases in communities.

A study on cometabolic bioventing for the in situ remediation of trichloroethylene.

Cometabolic bioventing for removal of TCE in the unsaturated zone was studied in a soil column study using methane as growth substrate. A numerical model was developed for simulating the behavior of TCE during cometabolic bioventing. The model parameters were estimated independently through laboratory batch experiments or from the literature. Simulations were found to provide reasonable agreement with the experimental data. The experimental data show that a total TCE remediation efficiency of over 95% was obtained. The volatilization-to-biodegradation ratio of TCE was about 7:1 and T ( c ) values ranging from 0.0078 to 0.07 were obtained in this methane-driven system. Due to the toxicity of the high TCE concentrations to the microbial biomass in the initial stages of the experiment, cometabolic biodegradation was enhanced and was more efficient in the later stages of cometabolic bioventing.
