Study on serum-free media for genetically engineered CHO cells producing prourokinase.

Based on the initial optimization of DMEM:F12(1:1), two kinds of serum-free media, 11G-SG-SFM and 11G-SE-SFM, were formulated by using orthogonal experiments for the growth of genetically engineered CHO cell line 11G and the production of recombinant prourokinase (pro-UK) in a suspension culture, respectively. Growth of 11G cells in 11G-SG-SFM was comparable with that observed in DMEM:F12 + 5% NBS and CHO-S-SFM. 11G cells grew slowly in 11G-SE-SFM, but secreted a relatively high level of pro-UK. The pro-UK productivity of 11G cells in 11G cells in 11G-SE-SFM reached 800 to 1,000 IU/10(6). d-1 and was about 80% and 20% higher than in DMEM:F12 + 5% NBS and CHO-S-SFM, respectively.

Neurotrophic effects of Schwann cells in culture on spinal anterior horn neurons.

OBJECTIVE: To study the neurotrophic effects of Schwann cells in culture on damaged spinal anterior horn neurons. MATERIALS AND METHODS: Dissociated from rat sciatic nerves undergoing Wallerian degeneration by trypsin-collagenase. Schwann cells were cultured for 96 hours to form a trophoblast, onto which embryonic day 14 rat spinal anterior horn neurons plated on glass coverslip and cultured for 4 hours were transferred for cocultivation without cell-cell contact between Schwann cells and spinal anterior horn neurons. The effect of survival and neurite outgrowth activity promoting factors secreted from Schwann cells on spinal anterior horn neurons were examined under phase-contrast microscope at 24, 48, 72 and 96 hours after cocultivation. RESULTS: When cultured for 24 hours, almost all the spinal anterior horn neurons, that had been damaged in the course of dissociation from spinal nervous tubes, survived, and some of them grew with short neurites. 96 hours after cocultivation, about 1/3-1/2 of the neurons survived and their bodies enlarged with neurites by more than six times of their body diameters. Contrarily, none of the neurons in the control group (no Schwann cell trophoblast) survived simultaneously. CONCLUSIONS: By the model of Schwann cell-spinal anterior horn neuron cocultivation separated by glass coverslip, it was well confirmed that Schwann cell can secrete a neuron survival promoting factor and a neurite outgrowth promoting factor, which undoubtedly play a crucial role in peripheral nerve regeneration.

Reduced tumorigenicity of metastatic human lung cancer cell subline (PGCL3) transfected with hRAR beta gene.

The recombinant PSG5-RAR beta plasmid and the G418-resistant PSV2 neo plasmid (10.1) were cotransfected into PGCL3 cells by coprecipitation with calcium phosphate. The transfectants CR3 and CR4, which expressed the RAR beta gene, were identified by Northern blot hybridization. The results showed that the in vitro growth and invasion of CR3 and CR4 were dramatically reduced compared to the control-transfected cell (CSV1). Furthermore, the colony-forming abilities in soft agar and the tumorigenicity in nude mice of CR3 and CR4 were abrogated. Our results suggests that RAR beta functions not only as a receptor mediating the RA action, but also as a suppressor in lung tumorigenesis.

[Clinical significance of determinations of blood catecholamine, glucose and isulin in burn patients].

UNLABELLED: Blood catecholamine (CA), glucose and insulin were determined in 30 severe burn patients, of whom 22 were males, and 8 females. Mean burn area was 58.6 percent TBSA. These patients were in hypovolemic shock. RESULTS: CA release was persistent in severe burn patients showing two peaks in shock period and infection period (P < 0.01). The quantity of epinephrine (E) was normal, while norepinephrine (NE) was presistently high (P < 0.01), and it was over two fold of the normal value. The value seemed to reflect the severity of the injury. When CA did not show a rise in the shock period, or lowered abruptly in the sepsis period, the patients were in high risk. Increasing CA produced increase in gluconeogenesis with insufficiency in SI secretion.

[Result on the screening of scoliosis among school students in Beijing area].

An investigation of the morbidity of scoliosis has been carried out in Beijing area including both urban and rural area. The survey involved 21759 elementary school students aged 8 to 14. The result showed that the morbidity of scoliosis was 1.06 per cent. Most patients had idiopathic scoliosis while congenital scoliosis possessed only 5.19% of the whole patients. Prevalence in female students was slightly higher than in males. In terms of morbidity of scoliosis, there was no significant difference seen in the areas of study. The use of Adam-forward-bending test for original examination in the investigation seemed to be simple and effective. The spine measure ruler is qiute simple in production and easy to handle. Using the ruler, one will get less exposure to X-ray. The film which shows the whloe spinal AP and lateral view would lay a final and ture basis for diagnosis.

[Effect of acupuncture on immunomodulation in patients with malignant tumors].

In order to investigate the role of acupuncture in the regulation of cellular immune function, the changes of T lymphocyte subsets (CD3+, CD4+, CD8+), soluble interleukin-2 receptor (SIL-2 R) and beta-endorphine (beta-EP) in the peripheral blood of patients with malignant tumors before and after acupuncture were observed with double blind method. Forty patients were divided randomly into two groups, 20 for each. One group treated with acupuncture and the other one for control. Results showed that acupuncture has the effect of enhancing the cellular immunity of patient with malignant tumor. Acupuncture treatment could increase the percentage of T lymphocyte subsets CD3+, CD4+ and the CD4+/CD8+ ratio (P < 0.01) and the level of beta-EP, as well as decrease the level of SIL-2 R (P < 0.01). The correlation analysis of the three criteria showed there was a positive correlation between beta-EP and T lymphocyte subsets and a negative correlation between beta-EP and SIL-2 R, there was also a negative correlation between T lymphocyte subsets and SIL-2 R. Based on these results, a discussion on the acupuncture immunomodulation network was conducted in this article in order to explore the possible mechanism of acupuncture on immunomodulation.

[A model of osteoporosis induced by retinoic acid in male Wistar rats].

An animal model of osteoporosis induced by retinoic acid was successfully established in 3-month-old male Wistar rats. The animals were given the drug 70 mg.kg-1.d-1 for 14 d intragastrically and sacrificed on day 29. The proximal tibia and middle tibia of the rats were processed undecalcifiedly for quantitative bone histomorphometry. Compared with the control rats, the cancellous and compact bone volume of the model rats were reduced markedly. The bone tissue microstructure showed some obvious pathological changes that the trabecular number were decreased, the separation of trabecular and medulla ossium cavity became large, the thickness of trabecular and cortex of bone were decreased. The mechanism of bone loss in the model rats was that the osteoclast was activated by retinoic acid which promoted bone resorption. Other changes in the model rats were also observed such as: the body weight, and the weights of seminal vesicle and prostate were decreased, the adrenal glands and spleen showed hyperplasia and hypertrophy. No change of the blood concentration of calcium, phosphorus, alkaline phosphatase, alanine transaminase, estradiol and testosterone in the model rats was observed.

Overexpression of c-myc gene without gene amplification in human hepatocellular carcinoma.

OBJECTIVE: To clarify the relationship between c-myc gene amplification and expression in human hepatocellular carcinoma (HCC) tissue. METHODS: Levels of amplification, transcription and P62 myc expression of c-myc gene in tumor tissues and corresponding non-tumor tissues were systematically studied using Dot blot hybridization, in situ hybridization and immunohistochemical technique, respectively. The 14 pairs of specimens were resected from patients with HCC. RESULTS: The amplification of c-myc gene was not found in tumor and non-tumor tissues. Overexpression of c-myc mRNA transcription was detected in 11 tumor tissues and 10 non-tumor tissues. The positive rate of P62 myc was 85.7% (12/14) in tumor tissues and 92.9% (13/14) in non-tumor tissues, respectively. A close correlation was noticed between c-myc mRNA and P62 myc expression. The results suggested that c-myc gene overexpression was not related to its amplification. CONCLUSIONS: The overexpression of c-myc mRNA and gene product without amplification may be characteristic in HCC.

[The value of serum CA125 assays in the diagnosis of uterine adenomyosis].

OBJECTIVE: To investigate the value of serum CA125 assays in the diagnosis of uterine adenomyosis. METHODS: Fifty-five consecutive patients with uterine-adenomyosis and twenty with leiomyoma of uterus, diagnosed by surgery and pathology were studied. The blood samples were taken one day before operation. Twenty normal healthy women were served as controls. CA125 levels were determined by radioimmunoassay method. RESULTS: The median (M) CA125 levels (Ql-Qu) for patients with adenomyosis, leiomyoma and normal controls were 102.1 (56.3-182.1)kU/L, 34.6 (33.7-43.8)kU/L and 33.1 (32.7-33.8)kU/L respectively. The differences among the three groups were all significant (P < 0.01). The CA125 positive rates (CA125 > 50kU/L) for the above three groups were 80.0%, 10.0% and 5.0% respectively. Patients with adenomyosis had a higher CA125 positive rate than those with leiomyoma or normal controls (P < 0.001). For patients with adenomyosis the CA125 levels were positively correlated with uterine size (r = 0.33, P < 0.05). The adenomyosis patients treating with sex hormone preparations were found to have lower CA125 levels in comparison with non-users (P < 0.05). The mean CA125 level measured in sixteen patients decreased significantly one week postoperation. CONCLUSIONS: Serum CA125 assay is of great assistance to the diagnosis of uterine adenomyosis as well as to the differential diagnosis between adenomyosis and leiomyoma of uterus.

[A spectral analysis of heart rate variability during postural tilt].

We analyzed the spectral characteristics of heart rate variability during the 4-minute periods just before and after postural tilt and during the 4-minute period just before the occurrence of syncope or the end of the test. 6 subjects with syncope had a progressive increase in low-frequency segment during these three periods, and highfrequency segment was significantly lower than that of the 10 subjects without syncope during the three periods. Those with syncope had a significantly higher segment ratio of low-frequency segment to high-frequency than those without syncope during these three periods. Thus, the imbalance of sympathetic and parasympathetic neural tones plays an important role in the syncope induced during postural tilt. The ratio of low-frequency segment to high-frequency is a practical measure which reflects the accuracy and sensibility of interaction of sympathetic and parasympathetic neural tones.

[Studies on microscopic characteristics of atmospheric particles].

Distribution of 15 elements on the surface of and morphological characteristics of 776 atmospheric particles collected from six air monitoring spots in Taiyuan City, Shanxi Province, were analyzed with scanning electronic microscopy and energy dispersive X-ray spectrometry, to study their chemical compositions in air pollution. Results showed silicon and aluminum were abundant in the particles, accounting for 49.4% and 25%, respectively, which indicated they were major components of the particles. Contents of lithophilic elements, such as silicon, aluminum and potassium, on the particle surface were decreased relatively with gradual decrease of particle diameter. Silicon and aluminum in the particles were detected frequently, also, accounting for 96.55% and 89%, respectively, which indicated alumina-silicate particles accounted for the dominant part in atmospheric aerosol, and dust particles from varied sources showed different characteristics and distributions.

[Effect of hemin in treating hemorrhagic anemia and toxicity].

AIM: To study the effect of hemin in treating hemorrhagic anemia and toxicity. METHODS: Fifty rats with hemorrhagic anemia were randomly divided into 5 groups with different dosage of hemin (93, 168, 300 mg.kg-1.d-1), ferrous gluconate (FG 300 mg.kg-1.d-1), and water, ig for 7 d. Twenty mice fed with hemin (6.0 g.kg-1.d-1) in 24 h for observing acute toxicity effects. Long-term toxicity were observed in 80 rats given hemin (0.65, 1.3, 2.6 g.kg-1.d-1) in 3 months. RESULTS: Hb of the rats of corresponding groups were 66-->121, 71-->141, 66-->148, 69-->140, and 67-->112 g.L-1. There were no adverse effects observed on acute toxicity test. No abnormalitis were found in hemogram, liver renal function test, and autopsy. CONCLUSION: Hemin had a better effect than FG and no adverse effect was found in hemin.

IFN-alpha treatment suppresses the development of experimental autoimmune myasthenia gravis.

Myasthenia gravis (MG) is an Ab-mediated autoimmune neuromuscular disease and is linked to MHC class II beta-chain polymorphism. Corticosteroids and azathioprine are the primary immunosuppressive drugs used in the treatment of MG. These drugs have significant side effects and have limited efficacy. Therefore, drugs with fewer side effects and greater efficacy are being sought. IFN-alpha is a potent immunomodulator and has been shown to down-regulate MHC class II expression on lymphoid cells. MHC class II expression is critical for the development of experimental autoimmune myasthenia gravis (EAMG). Because of the immunomodulating effects of IFN-alpha and its effect on the MHC class II expression, we tested the therapeutic efficacy of IFN-alpha on EAMG induced by immunization with acetylcholine receptor (AChR) in CFA. IFN-alpha (10(5) IU three times weekly for 5 wk) treatment started 1 wk after the second immunization with AChR in CFA, when autoimmunity to AChR is well established, reduced the incidence of clinical EAMG by more than 50% in two separate experiments (p = 0.04 and 0.008). Therefore, IFN-alpha could be a potential agent for the control of MG, and other Ab-mediated autoimmune diseases.

TCR gene usage in experimental autoimmune myasthenia gravis pathogenesis. Usage of multiple TCRBV genes in the H-2b strains.

Experimental autoimmune myasthenia gravis (EAMG) is an Ab-mediated autoimmune disease. The pathogenic auto-antibody production depends on the activation of CD4+ cells after their TCR interact with dominant T cell epitopes within acetylcholine receptor (AChR) in the context of the MHC class II molecule. In vitro analysis suggested that the TCRBV6 was the predominant TCR that recognized AChR and one of the dominant epitopes, alpha 146-162, in C57BL6 (B6, H-2b) mice. However, in vivo depletion of TCRBV6 cells in H-2b mice by anti-TCRBV6 mAb neither suppressed the in vitro immune response to AChR nor prevented development of EAMG. Moreover, B10.TCRc (H-2b) strain with a genomic deletion of TCRBV genes including TCRBV6, and B10.V beta 8.2 transgenic mice with a restricted TCRBV8S2 T cell repertoire, responded to AChR, alpha 146-162, and developed EAMG after immunizations with AChR/CFA. These data suggest that more than one TCRBV-bearing cell having the affinity for AChR-dominant peptides is involved in pathogenesis. Therefore, depletion of a single TCRBV (e.g., TCRBV6) with mAb may not be sufficient to completely suppress the response to AChR and development of EAMG. However, if a similar amino acid sequence in the TCR-VDJ (e.g., CDR3) region among different TCRBV gene(s) could be involved in recognizing the dominant AChR epitope(s), then motif-specific mAb reactive to the common motif within the VDJ region of different TCR could be used to eliminate the T cell clones involved in EAMG.

Cytotoxic ent-kaurene diterpenoids from Isodon gesneroides.

From Isodon gesneroides, three new cytotoxic diterpenoids, gesneroidins A, B and C, together with one known diterpenoid, dawoensin A, were isolated, and the structure determination and unambiguous assignment of their stereochemistry and NMR spectral data were made by a combination of one-and two-dimensional NMR techniques, computer modelling calculations and X-ray analysis.
