[Determination of hyperin in Sanguisorba Officinalis L. by high performance liquid chromatography].

An HPLC method for the determination of hyperin in Sanguisorba officinalis L. was investigated. 0.5 g of dried powdered crude drug was weighed accurately in a 50 mL volumetric flask added with 40 mL methanol, supersonicated for 30 min and then made up to volume with methanol. The sample solution was separated on a Shim-pack ODS column(6.0 mm i.d. x 150 mm, 5 microns) with methanol-0.025 mol/L phosphoric acid (adjusted to pH 3.0 by using triethylamine)(50:50) as the mobile phase and detected at UV-370 nm. Calibration curve of hyperin was obtained for the concentration range of 0.046-0.2 microgram. The corresponding regression equation was Y = 154 x 10(4) X-0.28 x 10(4), r = 0.9998. The recovery was 98.7% and relative standard deviation was 1.9%. The results showed that this method is simple, specific and accurate.

[A clinical analysis of 64 cases of pemphigus].

OBJECTIVE: Reasonable therapeutic dosage regimen for pemphigus are analyzed to find the factors related to prognosis. METHODS: A retrospective study is made on 64 cases of pemphigus who were admitted to our hospital between 1986-1996. RESULTS: Sixty-two of 64 cases with pemphigus were treated with systemic administration of corticosteroids. The average dosage of prednisone for controlling the diseases were 0.958 mg.kg-1.d-1 for mild-type of pemphigus (12 cases) and 1.145 mg.kg-1.d-1 for severe-type (50 cases) respectively. Furthermore, 26 of 62 cases were treated with corticosteroids only, and the average dosage of prednisone for controlling the diseases was 1.072 mg.kg-1.d-1 while in 36 cases treated with corticosteroids combined with immunosuppressors, the average dosage of prednisone was 1.178 mg.kg-1.d-1. The rate of side-effects and complications were 15.4% in patients with corticosteroids alone and 36.1% in patients with combined therapy. Six of eight patients who died were diagnosed pemphigus vulgaris and seven with a course of disease less than 3 years. CONCLUSIONS: We recommend a moderate to high dosage of corticosteroids be the first choice to control pemphigus. Those who were effective with steroids are not recommended to use additional immunosuppressors. Patients with pemphigus vulgaris usually have a poor prognosis while those who could survived more than 4 years usually have a better outcome.

[Effect of 8-(N,N'-diethylamino)-n-octyl-3,4,5-trimethoxybenzoate on [Ca2+]i and the release of NO in cultured endothelial cells of the calf middle cerebral artery].

The effect of 8-(N,N'-diethylamino)-n-octyl-3,4,5-trimethoxybenzoate(TMB-8) on [Ca2+]i and the release of NO in cultured endothelial cells of the calf middle cerebral artery was studied by a system of measurement of AR-CM-MIC, using Fura-2/AM as a fluorescent indicator and the oxy-haemoglobin method. In the presence of extracellular Ca2+ 1.3 mmol.L-1, the resting [Ca2+]i and the extinction difference(delta E) of between 401 and 410 nm were not changed by TMB-8 12.5 and 25 mumol.L-1, but were increased by TMB-8 50 and 100 mumol.L-1. The rise of delta E induced by TMB-8 50 and 100 mumol.L-1 was blocked completely by L-NAME. The elevation of [Ca2+]i was related to the release of NO. The result suggested that TMB-8 increased resting [Ca2+]i of endothelial cells of the calf middle cerebral artery and induced NO release.

[In vitro sensitivity of oral gram-negative bacteria to the bactericidal activity of defensins].

Oral gram-negative bacteria, including Actinobacillus actinomycetemcomitans, Porphyromonas gingivalis, Capnocytophaga sputigena, Provotella intermedius, Provotella melaninogenicus, have been associated with destructive periodontal infection. An ultrasensitive antibacterial assay was applied to determine the sensitivity of a battery of oral gram-negative bacteria to the bactericidal effects of the isolated human neutrophil defensins (HNP) and two kinds of rabbit defensins NP1 and NP2. All species tested were killed by HNP, NP1 and NP2 except that porphyromonas gingivalis was resistant to HNP. However, there was strain-to-strain variation in sensitivity. It was concluded that the oral gram-negative bacteria were sensitive to the cidal mechanism involved in defensinmediated bacterial killing.

[Construction of the prokaryotic expression plasmid pYNenv encoding the gp120 env gene of human immunodeficiency virus type 1 of yunnan strain].

The recombinant envelope protein of human immunodeficiency virus type 1(HIV-1) of Chinese Yunan epidemic strain was expressed in E. coli for confirming the antigenic domains of the protein. The recombinant prokaryotic expressive plasmid pYNenv was constructed by inserting the env DNA fragment which encode the gp120 of HIV-1 Yunnan strain into pBV220. The env DNA fragment was obtained by nested polymerase chain reaction with the HIV-1 infected patients peripheral blood macrophage and monocyte genomes from Yunan HIV-1 epidemic area as template. The recombinant plasmid pYNenv was confirmed by restriction enzyme analysis. The recombinant protein was expressed in E. coli DH10b by pYNenv after vibration culture at 30 degrees C for 20 hours then at 42 degrees C for 5 hours and confirmed by SDS-PAGE electrophoresis. The protein can specifically react with the serum of HIV-1 infected patient from Yunan epidemic area by Western blot assay. The result showed that the recombinant protein can be used as antigen for detection of HIV-1 membrane glycoprotein antibody as well as for the further study of the role of envelope protein in pathology.

[Detection of proviral gene of human immunodeficiency virus type 1 from dried blood spots].

A method for detection of proviral human immunodeficiency virus type 1 DNA fragment encoded pol gene in dried blood spots on filter paper by nested primers polymerase chain reaction (nPCR) has been developed. The filter paper was pre-saturated and dried by EDTA proteineas K. About 50 microliters whole blood from HIV1 infected individuals was collected and onto the piece of filter paper, and the paper was air dried stored sealed in a plastic bag at 4 degrees C or room temperature for 1 to 64 weeks. The PCR with outer primers was performed in 0.5% ml tube with the piece of filter paper. Following the PCR with the inner primers was finished. The results showed that template DNA could detected in piece of filter papers which stored at 4 degrees C for 40 weeks as well as stored at room temperature for 24 weeks. The results could directly determined by ethidium bromide staining of the PCR products when it compared with the PCR products of standard positive control and DNA molecular weight marker in electrophoresis analysis on agarose gel. The method is rapid, specific, and sensitive and has the ability to detect as little as 10 proviral template DNA. The collected samples could mail from basic laboratory to center laboratory and the method could be applied in confirming and screening of HIV1 infection.

[Studies on reduction of hexavalent chromium by fungi].

Several fungal strain, isolated from activated sludge, sewage and soil, were able to grow in the medium containing 300-500 mg/L K2Cr2O7, and strain BS-1 was capable of resistance to K2Cr2O7 up to 900 mg/L. After 4-6 d growing of the strain BS-1 and other 3 strains in the liquid medium added 200 mg/L K2Cr2O7, the Cr(VI) content of the medium disappeared completely. These strain were identified as Penicillium sp. BS-1, BS-3, Aspergillus niger BR-4, Aspergillus flavus BX-1 respectively. The results from the detection of the fungal culture by UV-absorbance spectrum and chemical analysis showed that high-toxic Cr(VI) was reduced to lesstoxic Cr(III). Reaction conditions and effectors of Cr(VI) reduction by intact cells of Penicillium sp. BS-1 were studied. The optimum temperature was 30 degrees C, the optimum pH was 7.0. Addition of glucose (0.25%) enhanced Cr(VI) reduction, but high concentrations of Cr(VI) inhibited the reduction of Cr(VI) to Cr(III).

[Mechanism of specific effects of transient electromagnetic pulses on cell model].

In this paper, a facilitated cell model has been built to analyze the electromagnetic pulses (EMP) induced transmembrane potentials. The analysis shows that the athermal biological effects are caused by micropores on the membrance, not directly by transmembrane potentials. These micropores are enlarged by weak transient EMP and finally lead to membrane openings to make ions cross. The electroporation effects are used to explain the specific response of biological cells to transient EMP.
