Radioimmunoassay of human cardiac acidic isoferritin: a new index for hepatic cancer.

OBJECTIVE: To investigate human cardiac acidic isoferritin as a specific index of hepatic cancer. METHODS: Acidic isoferritin was isolated and purified from human heart muscle. A radioimmunoassay for the acidic isoferritin in human serum has been developed, on the equilibrium method. The antiserum was obtained from rabbits immunized with purified acidic isoferritin. The 125I-acidic isoferritin was prepared by the chloramine-T method. The data were processed using the automated smoothed spline function data processing program. RESULTS: The intra- and inter-assay CV of acidic isoferritin RIA were 1.65% and 9.71%, respectively, and the recovery rate was 102%. The antiserum provided a linear response from 7.0 to 369.6 micrograms/L with ED50 of 27.50 micrograms/L. The cross reactivity with AFP, CEA, lactoferrin and transferrin was negligible, and that with ferritin was 1.74%. The serum acidic isoferritin concentration showed a considerable variation in different sex and age groups. The serum acidic isoferritin was measured in liver diseases including hepatic cancer, hepatic cirrhosis and acute and chronic hepatitis. Its sensitivity for diagnosis of hepatic cancer was 73.05%, independent from the severity of hepatic injury. In 8 malignant tumors studied, acidic isoferritin appeared the most valuable in the diagnosis of hepatic cancer, with its positive, negative, false positive and false negative rates all being ideal. CONCLUSIONS: Acidic isoferritin may turn to be a rather specific index of hepatic cancer. Combination of monitoring both acidic isoferritin and AFP would raise the positive detection and specificity in the diagnosis of hepatic cancer.

[Repair of massive bone defect with free vascularized fibular graft].

In the study of repair of massive bone defect with free vascularized fibula graft, 13 cases were reported, in which traumatic defect in 7 cases, segmental resection of bone from tumors in 5 cases and osteomylitis in 1 cases. They all were treated successfully with vascularized fibular graft. After a follow-up of 6 months to 7 year, bone healing was observed with satisfactory and rehabilitation of functions. In one case, fatigued fracture occured twice due to early walking. It was concluded that free vascularized fibular graft was very helpful in the repair of massive bone defect, but prolonged external fixation after operation might be important to prevent fractur of grafted bone.

8-(N,N-diethylamino)-n-octyl-3,4,5-trimethoxybenzoate reduced [Ca2+]i elevation induced by histamine, serotonin, and glutamate in cultured calf basilar artery smooth muscle cells.

AIM: To study the effects of 8-(N,N-diethylamino)-n-octyl-3,4,5- trimethoxybenzoate (TMB-8) on intracellular free calcium ([Ca2+]i) in cultured calf basilar artery smooth muscle cells. METHODS: [Ca2+]i was examined by a system of measurement of AR-CM-MIC, using Fura 2-AM as a fluorescent indicator. RESULTS: In the presence of extracellular Ca2+ 1.3 mmol.L-1, histamine (His), serotonin (5-HT), and sodium glutamate (Glu) markedly increased the [Ca2+]i which was attenuated by TMB-8. In Ca2+ free Hanks' solution containing egtazic acid 0.1 mmol.L-1, TMB-8 not only reduced the resting [Ca2+]i, but also inhibited the elevation of [Ca2+]i evoked by His and 5-HT. CONCLUSION: TMB-8 reduced the resting [Ca2+]i and attenuated His-, 5-HT-, and Glu-induced increases of [Ca2+]i in basilar artery smooth muscle cells.

Effects of indomethacin on joint damage in rat and rabbit.

AIM: To study the effects of indometacin (Ind) on joint damages. METHODS: The volume of noninjected hind paw and interleukin-1 (IL-1) production from peritoneal macrophages and articular synoviocytes induced by lipopolysaccharides were assayed in adjuvant arthritis (AA) rats. Measurements of synovial fibroblast proliferative response and proteoglycan synthesis of cartilage from rabbits were used. RESULTS: The secondary inflammatory reactions in AA rats on d 18, 21, and 24 were suppressed by i.g. Ind 2 mg.kg-1.d-1 for 9 d. Ind promoted IL-1 production from both macrophages and synoviocytes in AA rats. Ind 10 mumol.L-1 enhanced the proliferation of rabbit synovial fibroblasts and suppressed the proteoglycan synthesis of articular cartilage in response to IL-1 in vitro. CONCLUSION: Ind is unfavorable to the repair of joint destruction.

Effects of synthetic (-)-huperzine A on cholinesterase activities and mouse water maze performance.

AIM: To compare the effects of synthetic and natural (-)-huperzine A (Hup A) on cholinesterase and mouse water maze performance. METHODS: Spectrophotometry was used to determine cholinesterase activity. Mouse water maze was used to evaluate nootropic effect. RESULTS: The IC50 of synthetic Hup A for acetylcholinesterase (AChE) of rat cortex and rat erythrocyte membrane determined in vitro were 64.7 (52.6-79.5) and 53.9 (43.6-66.6) nmol.L-1, respectively, and for butyrylcholinesterase of rat serum was 53.6 (44.9-63.8) mumol.L-1. Synthetic Hup A 0.12-0.48 mg.kg-1 ig produced a dose-dependent inhibition of brain AChE in mice. Synthetic Hup A 0.05 mg.kg-1 ig attenuated scopolamine-induced impairment of spatial memory. The efficacy of synthetic Hup A was the same as natural Hup A. CONCLUSION: Synthetic Hup A yielded an in vitro and in vivo pharmacological profile of activities similar to that of natural Hup A.

[Applied anatomy of the pedicled patella transposition for repair of the superior articular surface of the medial tibial condyle].

To investigate the feasibility of using the pedicled patella for repaire of the superior articular surface of the medial tibial condyle, 37 lower limbs were studied by perfusion. In this series, there were 34 obsolete specimens and 3 fresh specimens of lower legs. Firstly, the vessels which supply to patella were observed by the methods of anatomy, section and casting mould. Then, the form and area of the patellar and tibial medial conylar articular surface were measured in 30 cases. The results showed: (1) the arteries supplied to patella formed a prepatellar arterial ring around patella, and the ring gave branches to patella; (2) medial inferior genicular artery and inferior patellar branches of the descending genicular arterial articular branch merge and acceed++ to prepatellar ring at inferior medial part of patella; (3) the articular surface of patella is similar to the superior articular surface of the tibial medial condyle on shape and area. It was concluded that the pedicled patella can be transposed to medial tibial condyle for repaire of the defect of the superior articular surface. The function of the knee can be reserved by this method.

[Isolation of antibacterial polypeptides of human cervical mucus].

The acid-soluble extract of human cervical mucus was obtained by solving mucus with 5% acetic acid in the presence of protease inhibitor. The antibacterial activity of the acid-soluble extract was analyzed by gel overlay technique. The result showed that two protein bands which were designated human cervical protein-1 (Hcp-1), human cervical protein-2(Hcp-2) were potently antibacterial against E. coli 25922 and S. aureus 25923. Tricine-SDS-PAGE analysis indicated that Hcp-1, Hcp-2 actually contained three and four protein bands respectively. The molecular weights of Hcp-1 were 6.7 kd, 10 kd, 15.4 kd; these of Hcp-2 were 4.4 kd, 6.7 kd, 9 kd, 15.4 kd. Our studies suggested that human cervical mucosa might secrete some currently-unknown antibacterial polypeptides which play an important role in the cervical defense against infection.

[Distribution of morphine in acute morphine-treated rats].

The distribution of morphine in acute morphine-treated rats was studied by immunohistochemistry (PAP). Samples were examined for morphine distribution and localization in internal organs at 10-160 min after intraperitoneal injection of morphine. The results showed that morphine was distributed in the heart, hepatocytes, epithelial cells of the bile canalicular cells in liver, spleen, kidney, adrenal glands, stomach parietal cells, small intestinal villus, pancreas islet, sublingual glands and submandibular glands. Morphine was found in the cardiac tissue, liver, spleen, pancreas islet, small intestinal villus at 10 min after injection and in the cardiac tissue, stomach parietal cells at 160 min after injection. The cause of the postmortem the distribution of morphine in internal organs at different times after injection was discussed. The results of this study may be used as a basis for the choice of samples for the forensic toxicological analysis.

[Partially purified antibacterial polypeptides from granules of human large granular lymphocytes].

This was a study on the antibacterial activity of human large granular lymphocytes (LGL). In our previous work, three antibacterial fractions named HLP-1, HLP-2 and HLP-3 had been identified from the acid-soluble extracts of the granules of LGL by using the gel overlay technique. The present study demonstrated that HLP-1, HLP-2 and HLP-3 separated by preparative acid-urea polyacrylamide gel electrophoresis were potently bactericidal against E. coli MI-35P, S. aureus ATCC 25923, P. aeruginosa ATCC 27853 when tested for their antibacteiral activities by using both agarose radial diffusion assay and gel overlay technique. SDS-PAGE analysis showed that the HLP-3 was almost purified with the molecular weight of 7 kd. The HLP-1 and HLP-2 were more complex, and the ranges of molecular weight were 5.6-13 kd and 5.6-8.8 kd respectively. Our study suggests that the granules of human LGL might contain some low molecular weight antibiotic peptides which play an important role in human innate immunity.

[Antitumor effect of combined therapy with adenovirus-mediated CD suicide gene and interleukin 2 gene transfer and its immunological mechanism].

OBJECTIVE: Adenovirus harboring E. coli cytosine deaminase gene(AdCD) and adenovirus harboring interleukin 2 gene (AdIL-2) were used for the combined treatment of established tumors in vivo. METHODS: C57BL/6 mice were inoculated with B16F10 melanoma cells and 3 days later received at the tumor site injections of AdCD and/or AdIL-2 followed by i.p. injection of 5-fluorocytosine (5-Fc) 300 mg/kg per day for 10 days. RESULTS: Mice received combined therapy developed tumors more slowly and survived much longer when compared with mice treated with AdCD/5-Fc, AdIL-2, control adenovirus AdlacZ/5-FC, or PBS. To investigate the immunological mechanism of the antitumor effects of the combined treatment it was found to induce enhanced natural killer cell activity and specific cytotoxic T cell activity. FACS analysis demonstrated that AdIL-2/AdCD/5-Fc therapy increased the expression of H-2Kb and B7-1 on freshly isolated tumor cells. The CD4+ and CD8+ T cell infiltration in the tumor increased significantly. CONCLUSION: Transfer of CD suicide gene plus 5-Fc combined with transfer of IL-2 gene synergistically inhibits the growth of melanoma in mice. Besides the cytotoxic effect of 5-Fc, specific and non-specific antitumor immunity might be responsible for the therapeutic effect. The combined therapy might have therapeutic potentials for human cancer.

[The serum levels of soluble interleukin-2 receptor in patients with malignant lymphocytic proliferative disorders].

OBJECTIVE: To explore the changes of serum levels of soluble interleukin-2 receptor (sIL-2R) in lymphocytic malignancies and its clinical implication. METHODS: The serum levels of sIL-2R were measured in 60 cases of patients with various malignant lymphocytic proliferative disorders by using ELISA method. At the same time, the ratio of CD4+/CD8+ of peripheral blood mononuclear cells was also determined in 43 patients by using fluorescence activated cell sorter(FACS). RESULTS: The serum levels of sIL-2R in patients with multiple myeloma(MM), non-Hodgkin's lymphoma (NHL) or acute lymphoblastic leukemia(ALL) were significantly higher than that of normal value. The ratio of CD4+/CD8+ measured in 43 patients were markedly lower than that of normal value. In MM, NHL and ALL patients, the higher values of sIL-2R were revealed in severe cases or late stage of these diseases. The levels of sIL-2R were independant of the infection in these cases. CONCLUSION: sIL-2R levels significantly elevated as the disease progressing, and the immune function decreasing, and the sIL-2R level may be used as an effective clinical marker.

The effect of blockage of macrophage on tumor-suppressive activity of polyactin B.

OBJECTIVE: To investigate the tumor-suppressive activity of polyactin B (PB) and the life-span of tumor bearing mice treated with PB and the anti-tumor mechanisms of PB by blockage of macrophage in vivo. METHODS: Thirty-two S180 sarcoma-bearing (s.c.) BALB/c inbred mice were divided into 4 groups, i.e., control group (8 mice), PB group (8), Trypan blue (TrB) group (8) and PB + TrB group (8). Five days after PB injection (i.p.), the tumor size of the mice was measured, and the ratio of TNA to TTA in HE section of the tumor maximal area was calculated and the pathological features of the tumors were observed. Forty S180 sarcoma-bearing (i.p.) ICR mice, which were randomly divided into 4 groups as above, were used for observation of the life-span. RESULTS: PB could strongly inhibit the growth of S180 sarcoma with the tumors regressing completely in 3 of 8 mice, prolong the life-span of mice bearing S180 sarcoma, increase the infiltration of macrophages and lymphocytes in the periphery and inside of the tumors. In addition, there were more prominent hemorrhagic necrosis, neutrophil infiltration and microthrombi in small vessels around the necrotic areas in the tumors of PB treated group. The tumor-suppressive effect of PB disappeared after macrophage system blockage with TrB. In PB + TrB, TrB, control and PB groups, the size of tumors reduced, and the life-span of mice increased in successive order. CONCLUSIONS: (1) PB is a strong activator of macrophage. (2) In vivo the tumor-suppressive effect of PB is initially mediated by PB activated macrophage system. Then other immunocompetent cells, e.g., T-cells, NK-cells, etc, activated directly and/or indirectly by PB, may play a very important role in tumor suppression. (3) PB-activated macrophages are more sensitive to TrB blockage. Therefore, using TrB + PB to block macrophage system is more effective than TrB alone. (4) Blockage of macrophage function might influence the life-span of the mice.

[Therapeutic effects of combined suicide gene and cytokine gene therapy on erythroleukemia-bearing mice].

OBJECTIVE: To explore the antitumor effect of combined transfer of suicide gene and GM-CSF gene in erythroleukemia-bearing mice. METHODS: Adenovirus harboring E. coli. cytosine deaminase (CD) gene (Ad-CD) and/or GM-CSF gene (Ad-GM-CSF) were used for the treatment of erythroleukemia-bearing mice. The mice were inoculated with FBL-3 erythroleukemia cells subcutaneously and 3 days later received Ad-CD followed by 5-fluorocytosine (5FC) treatment with or without Ad-GM-CSF. RESULTS: The mice received Ad-CD/5FC and Ad-GM-CSF developed tumors more slowly and survived much longer than those received Ad-CD/5FC alone, Ad-GM-CSF alone, control virus Ad-LacZ/5FC or PBS. Combined transfer of CD gene and GM-CSF gene induced a higher specific CTL activity than control therapies did. Pathological examination illustrated that there were tumor necrosis and massive lymphocyte infiltration in the mice after the combined therapy. CONCLUSION: Combined transfer of suicide gene and cytokine gene could synergistically inhibit the growth of erythroleukemia cells in the mice and induce tumor specific immunity of the host.

[Construction of the internal standard RNA and DNA templates for MRP gene quantitative RT-PCR analysis].

OBJECTIVE: To construct the internal standard RNA and DNA templates for quantitative reverse transcription-polymerase chain reaction (RT-PCR) and quantitatively measure the mRNA expression of MRP gene. METHODS AND RESULTS: A 292 bp fragment was amplified by PCR from plasmid pRC/RSV-MRP containing full-length MRP cDNA and inserted into the Sma I site of PUC19 vector, constructing a recombinant plasmid-pUMRP292. A 238 bp HGV fragment was amplified by PCR from plasmid pUHGV and inserted into the Cla I site of pUMRP292, constructing a new recombinant plasmid-pUMRP 292/HGV as the internal DNA competitive template for quantitative PCR of MRP. A 530 fragment containing the 292 bp of MRP and the 238 bp of HGV was cut down by EcoR I and Xba I pUMRP292/HGV and cloned into the transcriptional vector pSP72 by the same enzyme sites, constructing a recombinant plasmid-pSMRP292/HGV, and then pSMRP292/HGV was cleaved with EcoR V and transcripted in vitro by SP6 RNA polymerase, obtaining a 530 bp mutant MRP-RNA positive strand as the internal RNA competitive template for quantitative RT-PCR. CONCLUSION: The established quantitative RT-PCR assay is simple, rapid and sensitive, and the DNA competitive template is more simple, economic and reliable than the RNA one.

[Correlation of serum markers with fibrosis staging in chronic viral hepatitis].

OBJECTIVE: To assess the correlation between serological fibrosis markers and staging of fibrosis in chronic hepatitis. METHODS: One hundred and fourteen (114) biopsy and 13 autopay liver specimen were reviewed pathohistologically. Serum samples for liver function tests and fibrosis marker assays including serum P-III-P, IV collagen, laminin (LN), hyaluronic acid (HA), h-TIMP-1 were collected simultaneously right before the biopsy or autopsy. HE, sirius red stains as well as staining for reticulum and elastic fiber were done. In besides, IV collagen, laminin, activated stellate cell were assayed by immunohistochemical methods. RESULTS: Among 127 patients, 92 cases of chronic viral hepatitis including early cirrhosis and 18 cases of hepatic cirrhosis were examined pathologically (17 alcoholic liver disease excluded). The overall inflammatory index and fibrotic scoring were noticed to be significantly correlated with each other (r = 0.82, p < 0.01); The markers, such as HA, P-III-P, LN, IV collagen, h-TIMP-1 were positively correlated with the fibrotic staging and scoring (r = 0.610, 0.575, 0.607, 0.641, 0.606 respectively). The higher the marker level, the more serious the fibrotic change. In addition, (18 cases) patients with active cirrhosis (16 cases) showed higher serum marker level than that of the early cirrhosis and the serum markers in the later were higher than that in the non-active cirrhosis. CONCLUSION: Serum fibrosis markers are fairly well correlated with the staging of fibrosis and are considered to be the valuable references for clinical diagnosis. The close correlation between inflammation process and fibrosis suggests that the inflammatory process plays an important role in fibrogenesis.

Bcl-2 gene and its family genes Bax, Bcl-Xl as well as Fas/Apo-1 and their clinical significance in acute leukemia.

OBJECTIVE: To study the suppressing genes of apoptosis, namely Bcl-2, its family genes Bax, Bcl-Xl, and the inducing gene of apoptosis Fas/Apo-1. METHODS: The techniques of cytoimmuno-histiochemical stains, Western blotting and Northern blotting were used. RESULTS: It was found that the antigens of Bcl-2 in acute myelogenous leukemia (AML) and acute lymphocytic (ALL) was higher than that in the normal (P < 0.01). At same time, Bcl-2 was obviously lower expression in complete remission (CR) group than that in non-remission (NR) one by retrospective analysis (P < 0.01). Though Bcl-2 was low expression in CR with Western blotting, there was no statistical significance (P > 0.05). In CR group the expression of Bcl-2 mRNA was obviously lower than that of NR one (P < 0.01). Even though leukemia expression of Bax with cytoimmuno-histiochemical stain was also lower than that in the normal people, there was no difference between CR and NR with cytoimmuno-histiochemical, Western blotting and Northern blotting (P > 0.05). There was difference of Bcl-Xl mRNA in two groups (P < 0.01). The expression Fas/Apo-1 in leukemia was lower than that in normal people (P < 0.01). But in CR and NR, there was no difference with cytoimmuno-histiochemical stain and Western blotting. CONCLUSIONS: The Changes of genes and their proteins are significant theoretically and clinically. The antigen expression of Bcl-2 and the expression of Bcl-2 mRNA may be considered as a prognostic index for AML.
