Orally active peptidomimetic RGD analogs that are glycoprotein IIb/IIIa antagonists.

Peptidomimetic RGD (Arg-Gly-Asp) analogs, which bind to glycoprotein (GP) IIb/IIIa on the surface of activated platelets, have been shown to inhibit platelet aggregation. Consequently, such RGD analogs can be used for the treatment of unstable angina pectoris and myocardial infarction. However, the low oral bioavailability for this class of compounds has been hindering their clinical development. Although many factors affect the oral activity of a drug, the limited membrane permeability of RGD analogs due to charge and high polarity is thought to be a major factor leading to the low oral activity of such compounds. Another factor is the metabolic lability of some such RGD analogs in the presence of proteases and peptidases. During the last 5 years, major progress has been made in the development of orally active RGD analogs. To improve the metabolic stability of RGD analogs, N-alkylation and C-terminal modification methods have been used successfully. To improve the membrane permeability of RGD analogs, two major strategies have been used. The first one is the strategy of prodrug. Along this line, simple ester prodrugs, double prodrugs, triple prodrugs, and cyclic prodrugs have been prepared with improved membrane permeability and oral activity. The second approach used is the de novo design of centrally constrained RGD analogs with improved oral bioavailability while maintaining the desired potency against GP IIb/IIIa. The lessons learned from the modification of RGD analogs could also help the future design of other peptidomimetic drugs with improved oral bioavailability.

Tyrosine phosphorylation of an erbB-2 related p90 protein induced by estrogen in human breast epithelial cells.

Estrogen induced erbB-2 tyrosine kinase activity in human breast epithelial cells irrespective of estrogen receptor expression. MCF10A is an immortal normal human breast epithelial cell line which does not express estrogen receptor. After treatment of MCF10A cells with estradiol-17beta (E2), a phosphorylated 90 kDa protein which co-immunoprecipitates with p185erbB-2 is detected. The response is transient, detected after 1-5 min exposure to E2, and dose dependent, occurring at 10-10 M E2. A similar response was observed for MCF10A cells transfected with an estrogen receptor, estrogen receptor expressing MCF-7 cells, and estrogen receptor-negative MDA-MB-435 cells but at 10-11 M E2. Overexpression of c-erbB-2 in MCF10A cells prolonged the phosphorylated p90 response to E2.

Acute exposure to pulsed 2450-MHz microwaves affects water-maze performance of rats.

Rats were trained in six sessions to locate a submerged platform in a circular water maze. They were exposed to pulsed 2450-MHz microwaves (pulse width 2 micros, 500 128;pulses/s, average power density 2 mW/cm(2), average whole body specific absorption rate 1.2 W/kg) for 1 h in a circular waveguide system immediately before each training session. One hour after the last training session, they were tested in a probe trial during which the platform was removed and the time spent in the quadrant of the maze in which the platform had been located during the 1-min trial was scored. Three groups of animals, microwave-exposed, sham-exposed, and cage control, were studied. Microwave-exposed rats were slower than sham-exposed and cage control rats in learning to locate the platform. However, there was no significant difference in swim speed among the three groups of animals, indicating that the difference in learning was not due to a change in motor functions or motivation. During the probe trial, microwave-exposed animals spent significantly less time in the quadrant that had contained the platform, and their swim patterns were different from those of the sham-exposed and cage control animals. The latter observation indicates that microwave-exposed rats used a different strategy in learning the location of the platform. These results show that acute exposure to pulsed microwaves caused a deficit in spatial "reference" memory in the rat.

[Expression of FADD in primary hepatocellular carcinoma and its relationship with hepatic apoptosis].

OBJECTIVE: To investigate the features of Fas-associated death domain (FADD) expression in HCC and to determine its relationship with hepatic apoptosis. METHODS: Immunohistochemistry method was used to detect FADD expression in HCC. In situ end-labeling (ISEL) was employed to determine apoptotic cells. RESULTS: Ten cases (25. 6%) of HCC were detected to express FADD protein. The positive rate in HCC was lower than that in non-cancerous adjacent liver tissues which showed as high as 62.5% cases harbored FADD protein (P<0.05). Six out of 18 (33.3%) cases of mid to well differentiated HCC had detectable FADD protein. It is a little higher, but with no statistic significance, than that in poorly differentiated HCC which showed only 19% cases were positive for FADD. In those of grade I/II, 8 cases (38.1%) were FADD positive, while only 11.1% (2/18) cases of grade III/IV had FADD protein (P<0.05). No relationship was found between FADD expression and other clinical features, such as sex, age, tumor size or metastasis. ISEL positive cells could be seen in all cases of HCC. The hepatic apoptosis was associated with FADD expression because more apoptotic cells were detected in those cases which showed moderate to strong FADD positive, as compared with negative or weak FADD positive cases (P<0.05). While no relationship was found between FADD expression and hepatic apoptosis in non-cancerous adjacent liver tissues. CONCLUSION: Loss of FADD expression plays an important role in HCC carcinogenesis. The apoptosis of cells is associated with FADD expression in HCC.

Pollution of water sources and removal of pollutants by advanced drinking-water treatment in China.

The pollution of water resources and drinking water sources in China is described in this paper with basic data. About 90% of surface waters and over 60% of drinking water sources in urban areas have been polluted to different extents. The main pollutants present in drinking water sources are organic substances, ammonia nitrogen, phenols, pesticides and pathogenic micro-organisms, some of which cannot be removed effectively by the traditional water treatment processes like coagulation, sedimentation, filtration and chlorination, and the product water usually does not meet Chinese national drinking water standards, when polluted source water is treated. In some drinking-water plants in China, advanced treatment processes including activated carbon filtration and adsorption, ozonation, biological activated carbon and membrane separation have been employed for further treatment of the filtrate from a traditional treatment system producing unqualified drinking water, to make final product water meet the WHO guidelines and some developed countries' standards, as well as the Chinese national standards for drinking water. Some case studies of advanced water treatment plants are described in this paper as well.

[Proliferation and apoptosis of hepatic stellate cells and effects of compound 861 on liver fibrosis].

OBJECTIVE: To investigate the proliferation and apoptosis of the hepatic stellate cell (HSC) in vitro and in vivo and the effects of Chinese herb, compound 861. METHODS: The in vitro study was carried out on the culture of hepatic stellate cell line. Various concentrations of compound 861 were added and incubated. Cell proliferation was detected with MTT colorimetric assay. Cell apoptosis was detected by electron microscopy, flow cytometry and TUNEL. The subjects of in vivo study were patients with chronic hepatitis B. RESULTS: Compound 861 could significantly inhibit HSC proliferation and increase the apoptosis rate of HSC dose-dependently and time-dependently compared with the control group. After compound 861 incubation for 48h, the apoptosis rate of HSC was 25.9% compared with 9.2% in control group (P<0.05). The clinical study elucidated that the HSC was activated and proliferated in patients with chronic hepatitis B. After compound 861 treatment for 6 months, the number of activated HSC decreased and the apoptosis of HSC could be seen in the liver biopsy. CONCLUSION: Apoptosis of activated HSC exists in vitro and in vivo, stimulating its apoptosis may play an important role in the treatment of liver fibrosis.

[Evaluation for biocompatibility of poly-(3-hydroxypropyl)-DL-aspartamide].

To evaluate the biocompatibility of poly-(3-hydroxypropyl)-DL-aspartamide(PHPA), we utilized the laboratory methods, including subcutaneous implantation, histological examination, blood biochemical and micronucleus test. Histological examination revealed a nonspecific foreign body reaction around the PHPA and a slight inflammation around the small blood vessels of the liver on the 13th day, but all these reactions diminished or disappeared gradually after 34 days of implantation. The structures and functions of liver and kidney were normal. The results indicate that PHPA possesses a good nature in biocompatibility.

Detection of transfusion transmitted virus in hepatic and extra hepatic tissues using in situ hybridization.

OBJECTIVE: To investigate the location, distribution and significance of transfusion transmitted virus (TTV) in the liver and extra hepatic tissues. METHODS: A 123 bp fragment of TTV DNA within ORF(2) was amplified by PCR reaction and cloned into PCR2.1 vector. DNA sequencing revealed that the inserted fragment was highly isogenous with Japan ABO11494 strain. The inserted fragment was labeled with digoxigenin. Segment of liver, spleen, kidney, stomach and intestine tissues from 22 patients dead of liver diseases were detected by in situ hybridization with this digoxigenin labeled TTV DNA probe. RESULTS: TTV DNA was detected in 8 livers, 4 kidneys, 3 spleens, 2 intestines, and 2 stomachs. The positive cells of TTV DNA in livers were higher than that in other tissues. No obvious pathological topographical relationship of tissue inflammatory reaction with positive cells was found. CONCLUSION: TTV can infect hepatic and ex-hepatic tissues and may cause persistent infection.

[AFLP analysis of photoperiod-sensitive genic male sterile(PGMS) rice mutant lines].

The reaction conditions for rice AFLP assay were optimized. The relative efficiencies for polymorphism detection of RFLP, RAPD and AFLP were compared through the analysis between a pair of PGMS allelic mutant lines(NK58S and NK58F). Results indicated that the efficiency for polymorphism detection in rice is in the order of AFLP > RAPD > RFLP, and also indicated that AFLP is a powerful DNA molecular marker technique for polymorphism detection, especially in the cases of extremely low polymorphism, such as isogeneic lines and allelic mutant lines. The advantages and disadvantages of these three molecular marker systems were discussed. Using AFLP in conjunction with bulked segregating analysis, 5106 AFLP loci were screened and 9 of them showed polymorphism between NK58S and NK58F, 4 of the polymorphic AFLP products were cloned, Southern bloting analysis showed that two of them were single copy sequences while the other two were low copy sequences in rice genome.

Dose-proportional and stereospecific pharmacokinetics of methylphenidate delivered using an osmotic, controlled-release oral delivery system.

Methylphenidate hydrochloride (HCl) is frequently used for the treatment of attention deficit/hyperactivity disorder (ADHD). A study was conducted in healthy subjects to evaluate the dose-ranging pharmacokinetics of 18, 36, and 54 mg methylphenidate HCl delivered using an oral, osmotic, controlled-release formulation (OROS). Plasma concentrations of l-methylphenidate were 40-fold lower than those of d-methylphenidate, whereas plasma concentrations of d-alpha-phenyl-2-piperidine acetic acid (d-PPA) and l-PPA, the major metabolite of methylphenidate, were comparable. Mean AUCinf values for d-methylphenidate were 42.2, 80.9, and 120 ng.h/mL for the 18, 36, and 54 mg doses, respectively, increasing dose proportionally. AUCinf values for l-methylphenidate were only approximately 1% of d-methylphenidate (0.43, 0.96, and 1.82 ng.h/mL for the 18, 36, and 54 mg dose groups, respectively). In contrast, AUCinf values of d- and l-PPA were comparable. The dose-normalized d- and l-methylphenidate plasma concentration-time profiles for the three treatment groups were superimposable. Similarly, dose-normalized plasma concentrations of d- and l-PPA were superimposable. Methylphenidate metabolism, measured as the ratio of d-methylphenidate AUCinf to d-PPA AUCinf and as l-methylphenidate AUCinf to l-PPA AUCinf, was similar for the three dose groups, indicating that methylphenidate metabolism was not affected by increasing dose. OROS (methylphenidate HCl) exhibits dose-proportional and linear pharmacokinetics.

[Expression of collagen XVIII mRNA in rat liver fibrosis].

OBJECTIVE: To measure and study quantitatively the collagen XVIII mRNA in normal and fibrotic rat livers. METHODS: We used ribonuclease protection assay to investigate the collagen XVIII mRNA expression in rat liver fibrosis induced by complete bile duct occlusion (BDO). The expression level of procollagen 1 (XVIII) mRNA was compared with that of procollagen 1 (I) and tissue inhibitor of metalloproteinase 1 (TIMP1). RESULTS: mRNA levels of procollagen and TIMP 1 increased 20- and 4-fold in BDO rat livers, respectively. In contrast, hepatic procollagen 1 mRNA level increased only 1.8-fold in fibrotic rat livers. CONCLUSION: C XVIII mRNA is upregulated slightly in liver fibrosis, which is probably correlated with the fact that CXVIII is mainly expressed by hepatocytes.

Analysis of DYS19 and DYS287 polymorphisms in the Han population and three other ethnic groups of northeast China.

The allelic distribution of the Y-chromosome specific microsatellite DYS19 in the Han population and in the Daur, Oroqen and Ewenki ethnic groups (Northeast China) was analyzed by PCR and denatured polyacrylamide gel electrophoresis. The allelic distribution in the Han population group is as follows: A = 2.90%, B = 26.09%, C = 26.09%, D = 29.98%, E = 15.94%. This allelic distribution differs statistically significant from that observed in the three other ethnic groups (p < 0.05). Furthermore the polymorphism of the Y-chromosome specific Alu insert sequence DYS287 was tested in these four groups. However, no Alu sequence insert was found.

[Estimates of breeding value of Inner Mongolia cashmere goats using animal model BLUP method].

In this study, single trait animal model of best linear unbiased prediction (BLUP) procedures was used to obtain estimates of 3,981 Inner Mongolia cashmere goats' breeding value for body weight and cashmere yield. The data were collected during 1989-1998 at Arbas cashmere goats farm, Etuoke banner, Inner Mongolia. Multiple traits of BLUP were used to get estimates of total breeding values (TBV) of all goats. The model included age and sex-herd-year as fixed effects, and individual additive effect and individual permanent environment as random effects. The selection based on breeding value and phenotypic value was compared. The results showed as following: (1) There was a large difference between weaning weight selection and TBV selection of ram lamb; (2) The difference between phenotypic selection and TBV selection of gimmer hogg reached a significant level (P < 0.01); (3) The difference between phenotypic selection and TBV selection of ram hogg reached a significant level (P < 0.01); (4) Rank correlation between body weight selection and body weight' EBV selection and that between cashmere yield selection and cashmere yield' EBV selection were not significant (P > 0.05). The study concluded precision of individual phenotypic selection was low and animal model BLUP was suitable to select Inner Mongolia cashmere goats. At last, this study gave a series of methods of selecting sires based on objective reality.

[Three-dimensional reconstruction of experimental fibrotic liver tissue and effect of herbal compound on it].

OBJECTIVE: To observe the three-dimensional structure of fibrotic liver tissue and the effect of herbal compound 861 (Cpd861) on it. METHODS: Experimental rat liver fibrosis was induced by human albumin. Liver biopsy specimens were stained with Masson trichrome staining and liver fibrosis was divided into 6 stages. Three-dimensional structure of fibrotic and Cpd861 treated liver tissue were observed by computer aided three-dimensional reconstruction with serial slices. RESULTS: The fibrous septa of fibrotic liver were continuous and wide, the sinusoids were sparsely, irregularly distributed and dilated. After treatment with Cpd861, the fibrous septa were much less than that of without treatment. The sinusoids were more regular and arranged in radius to central veins. CONCLUSION: Observed three-dimensionally, experimental rat liver fibrosis can be inhibited and reversed by herbal compound.

Effects of TMB-8 on constriction of pial arteries in rats.

AIM: To study the effects of 8-(N,N-diethylamino)-n-octyl-3,4,5-trimethoxybenzoate (TMB-8) on constriction of pial arteries (PA). METHODS: The change of PA in rats was observed continuously and directly through cranial window using microcircular image-shearing system. RESULTS: Nimodipine (Nim) 1 mumol.L-1 produced dilatation of PA immediately and the maximal response occurred in 2 min. But the diameter was not changed by TMB-8 50 mumol.L-1. PA diameter decreased immediately after the application of CSF containing KCl. TMB-8 25, 50 mumol.L-1 apparently inhibited KCl-induced PA constriction. When persistent constriction was evoked by 5-HT, diameter of PA were increased in a concentration-dependent manner after application of TMB-8, which was inhibited by N omega-nitro-L-arginine methyl ester (L-NAME). CONCLUSION: TMB-8 inhibited the contraction induced by 5-HT or KCl in cerebral artery, probably via its calcium antagonization and nitric oxide release.

[An experimental study of human tracheal epithelial transfer of human neutrophil defensin gene].

This study was amied at the possibility of production of the human neutrophil defensin in human tracheal epithelial cells when transfected with it's gene by using liposome. The plasmid vector carrying the human neutrophil defensin HNP1 (human Neutrophil Peptide-1) gene was used to transfect the human tracheal epithelial cells cultured in serum-free complement medium by lipofection reagent. Antiserum of HNP1 was prepared and used for the immunocytochemistry staining to detect expression of HNP1 gene in human tracheal epithelial cells. The result showed that recombinant eukaryote expression plasmid-pBabe-Neo-HNP1 could be transducted into human tracheal epithelial cells by liposomal transfection method. Transducted cells showed strong positive staining for HNP by immunohistochemistry assay. It was suggested that HNP1 gene could be productively transduced into human tracheal epithelial cells and expressed by using liposome. Antibiotic peptide gene might be used for gene therapy of respiratory infections.

[Analysis of the design principle and mechanics nature of adjustable semi-circular planum external fixator].

This thesis presents our studies of an adjustable semi-circular planum external fixator and offers a relatively detailed introduction and analysis of the structure principle and mechanics nature. Because dual-direction joining pole with active joints was used, by adjusting the length of each pole, it can not only make the two semi-circular steel plates more parallely along the axis line to stabilize the fracture and lengthen the extremity, but it also can change the planum angle of the steel plate, and thus achieve the effect of orthopaedic function.

[Effects of epimedium on the expression of interleukin-6 messenger ribonucleic acid in bone of ovariectomized rat].

OBJECTIVE: To investigate the effects of the traditional herb-epimedium on the expression of interleukin-6 (IL-6) mRNA in bone of ovariectomized rat. METHODS: Forty female rats were randomly allocated into 4 groups, 10 in each: ovariectomized (OVX) group, sham operation group, OVX followed by epimedium (group 3) or nilestriol (group 4) for 3 months respectively. All rats were then sacrificed, and total RNA were directly isolated from their right tibia. Interleukin-6 mRNA expression was detected by relative semiquantitative reverse transcription-polymerase chain reaction technique. Lumbar bone mineral density (BMD) was measured by dual-energy X ray absorptiometry before sacrifice. RESULTS: The BMD of epimedium group was significantly higher than that in the OVX group (P < 0.05). On the contrary, the IL-6 mRNA expression level of epimedium group was significantly lower than that in the OVX group (P < 0.01), but higher than those in the sham and nilestrial group (P < 0.01). CONCLUSIONS: IL-6 may play a role in the pathogenesis of postmenopausal teoporosis. Epimedium can inhibit the expression of IL-6 mRNA, which may contribute to its anti-resorptive effect.
