[The distribution and natural degradation of cyanide in goldmine waste-solid and polluted soil].

The farmland and river were seriously polluted by cyanide because one goldmine tailing dam collapsed in 1995. 3 and 4 years after the accident, the cyanide distribution in the polluted farmland and the abandoned tailing dam was studied. The results indicated that natural degradation of cyanide in soil section was slower than in natural water body. The cyanide transference in soil section was similar to freely soluble salts. In arid and semiarid area, cyanide can be highly enriched in the salt shell which content degrading 4 years even higher than the fresh tailing slurry. One side the viscidity layer in the soil section can partly prevent cyanide transference to groundwater, on the other side the result can cause the cyanide highly enrich in the viscidity layer. According to character of cyanide natural degradation in soil the measurement of prevention and cure soil pollution by goldmine tailing dam collapsing was brought forward.

[The influence of ecological factors on simultaneous nitrification and denitrification in MBR].

The influence of dissolved oxygen(DO), C/N and pH on simultaneous nitrification and denitrification in the membrane bioreactor was studied. The experimental results showed that only under condition that each ecological factor was maintained relative steady, simultaneous nitrification and denitrification might proceed smoothly. When DO, C/N and pH of feed water was 1 mg/L, 30 and 7.2, the removal rate of COD, NH4+, and TN were 96%, 95% and 92% respectively.

[Construction of a dicistronic expression plasmid vector containing double-valent hepatitis B surface gene].

OBJECTIVE: In order to enhance vaccine response, we constructed a dicistronic expression plasmid containing double HBsAg immunogenes. METHODS: At first, pcDNA3.1 plasmid vector was digested with NheI and EcoRI to get the coding sequence of the small (S) surface protein of HBV, then cloned into pCI-neo vector and named it pCI-S. By PCR amplification, the product of IRES-S was digested with SalI & BamHI, and cloned into pBluescript IIK+S to generate pBKS-IRES-S vector, then subcloned to the pCI-S plasmid to generate pCI-S-IRES-S, which is a dicistronic plasmid of double value HBsAg genes. RESULTS: Two plasmids we constructed were digested with related restriction nucleic enzymes. Sequence analysis of HBsAg and IRES-S gene did not reveal any mutation. CONCLUSIONS: The construction of dicistronic plasmid of divalue HBsAg immunogenes has been well cloned, which is convenient for further research on cell expression and gene immunization in animals.

[Relation between activities of hepatic and gastric alcohol dehydrogenases and formation of chronic alcoholic liver diseases].

OBJECTIVE: To investigate the role of hepatic and gastric alcohol dehydrogenases (ADH) in different pathologic stages of alcoholic liver disease (ALD). METHODS: Thirty-nine rats were divided into either model or control group at random. The ALD model was established by infusing alcohol into the stomach. The activity varieties of hepatic and gastric ADH were observed by optical microscopy after hepatic and gastric tissues had been stained by enzyme histo-cytochemistry assay and the activity alterations were also determined with a LUZEX-F analysis instrument as a semi-quantitative method. RESULTS: The activity of hepatic ADH gradually increased, but gastric ADH gradually decreased in ALD. There was a significant difference between model group and control one (P<0.05). CONCLUSIONS: The activity alterations of the hepatic and gastric ADH may play an important role in the onset and development of ALD.

[Dynamic change and expression of matrix metalloproteinase-2, -9 in alcoholic liver disease in rats].

OBJECTIVE: To evaluate the dynamic change and expression of matrix metalloproteinase-2 (MMP-2) and matrix metalloproteinase-9 (MMP-9) in alcoholic liver disease in rats. METHODS: Alcoholic liver disease rat model was established by gavage. The expression of MMP-2 and MMP-9 was studied by immunohistochemistry, and the result was analyzed by picture quantitative analysis technique. RESULTS: The expression of MMP-2 and MMP-9 was significantly higher in rats of model group than that of control group (P<0.05). The level of MMP-2 and MMP-9 increased gradually with the progress of alcoholic liver disease. Immunoreactive MMP-2 was expressed in endothelial cells of hepatic artery and portal vein, sinusoidal endothelial cells and sinusoidal cells. Immunoreactive MMP-9 was observed in sinusoidal cells and hepatic cells around the vein. CONCLUSIONS: MMP-2 and MMP-9 increa with the progress of alcoholic liver disease. They might be involved in the development of alcoholic liver disease.

MCF-10A-NeoST: a new cell system for studying cell-ECM and cell-cell interactions in breast cancer.

PURPOSE: There is a continuing need for genetically matched cell systems to model cellular behaviors that are frequently observed in aggressive breast cancers. EXPERIMENTAL DESIGN: We report here the isolation and initial characterization of a spontaneously arising variant of MCF-10A cells, NeoST, which provides a new model to study cell adhesion and signal transduction in breast cancer. RESULTS: NeoST cells recapitulate important biological and biochemical features of metastatic breast cancer, including anchorage-independent growth, invasiveness in three-dimensional reconstituted membranes, loss of E-cadherin expression, and increased tyrosine kinase activity. A comprehensive analysis of tyrosine kinase expression revealed overexpression or functional activation of the Axl, FAK, and EphA2 tyrosine kinases in transformed MCF-10A cells. CONCLUSIONS: MCF-10A and these new derivatives provide a genetically matched model to study defects in cell adhesion and signaling that are relevant to cellular behaviors that often typify aggressive breast cancer cells.

[The morphological and histological identification of Boschniakia rossica].

In this paper the descriptions and microscopic characters of Boschniakia rossica were recounted. The evidences of the identication were presented. The differences between Boschniakia rossica and its similar species Cistanche deserticola and Cynomrium songaricum were also summed up. Boschniakia rossica: Stem thin. Cross section hollow, brittle in texture, with cortex. Stele vascular bundles arranged in ring, pith obvious. Cistanche deserticola and Cynomrium songaricum: Stem thick and strong. Cross section solid, without cortex. Hard in texture. Pith unobvious. The stele vascular bundle of Cistanche deserticola arranged in wavy ring. That of Cynomrium songaricum irregularly arranged.

Inhibitory effect of recombinant TGFalpha-PE40 on neointimal proliferation after arterial balloon injury.

AIM: To investigate inhibitory effects of recombinant transforming growth factor alpha-Pseudomonas exotoxin 40 fusion protein (TGFalpha-PE40; TP40) on neointimal proliferation after arterial balloon injury. METHODS: Forty male rabbits fed a cholesterol rich diet were randomly divided into TP40 15 microg, 30 microg, 60 microg, physiologic saline control, and normal artery groups (n=8). Rabbits in the treatment groups were treated by local administration of TP40 (15 microg, 30 microg, and 60 microg per rabbit) 24 h postinjury, and those in the control group were treated by physiologic saline 24 h postinjury. Remained 8 rabbits in normal artery group were treated by TP40 (60 microg). Optical microscope, electron microscope, and computer image analysis were used to study arterial segments 2 weeks after treatment. RESULTS: Irregular thickening of the arterial intima, large amounts of smooth muscle cells (SMC) within the neointima, and stenosis of the arterial cavity were observed in the physiologic saline control group. Great inhibition of intimal proliferation and prevention of stenosis of the arterial cavity were observed in the TP40 treated groups that were examined 2 weeks postinjury by optical microscope. The uninjured carotids were histologically normal. Lots of destructural and necrotic SMC were observed in media in TP40 60 microg group by electron microscope. Computer image analysis showed that the neointimal area of the TP40-treated groups was markedly smaller than that of the saline control group (P < 0.01). CONCLUSION: Recombinant TP40 greatly inhibited neointimal proliferation after arterial balloon injury.

Effect of Salvia miltiorrhiza Bunge injection on anticardiolipin antibody production induced by beta2 glycoprotein.

AIM: To explore the therapeutic effect and the mechanism of Chinese herbs on antiphospholipid syndrome (APS) by observing the effect of Salvia miltiorrhiza Bunge injectio (SmBI) on anticardiolipin antibody (aCL) induced by beta2 glycoprotein I (beta2-GP I). METHODS: Sixty female mice randomly fell into 6 groups: group A, B, C, D was injected through abdominal cavity with different dosage of SmBI daily; after 14 d, group A, B, C, E was immunized with 150 microg of purified human beta2-GP I in complete Freund's adjuvant subcutaneously; group F as control. The titre of aCL were detected by enzyme linked immunosorbent assay; subsets of T cell were grouped by streptavidin-biotin complex technique; and the activity of IL-2 was measured by MTT chromatometry. RESULTS: (1) Compared with group E, the absorbance (A) of aCL in group A, B, and C was decreased (P < 0.05 or P < 0.01). By linear correlation, the dosage is negatively correlated with the A values of aCL in 1, 2, and 3 weeks (P < 0.01). (2) Compared with group E, TH/TS ratio was reduced in group A, B, and C (P < 0.05 or P < 0.01); there is no significant differences between group D and F (P>0.05). By linear correlation, the dosage is negatively correlated with TH/TS ratio (P < 0.01). (3) Compared with E, the activity of IL-2 in group B and C decreased significantly (P < 0.01). By linear correlation, there is negative correlation between dosage and IL-2 activity (P < 0.01). There is no significant difference between D and F (P > 0.05). (4) There is positive correlation between TH/TS ratio and IL-2 activity in different dilutions (P<0.01). CONCLUSION: The mechanism of suppressive effect of SmBI on aCL induced by beta2-GP I may be realized by resuming the elevated TH/TS ratio and IL-2 activity. The state that SmBI have no effect on normal mice indicates that SmBI has selective immunoregulative functive.

Antinociceptive effect of astragalosides and its mechanism of action.

AIM: To study the effect and mechanism of astragalosides (AST) related to the antinociceptive activity. METHODS: The standardized formalin test was performed to induce the direct stimulation of nociceptors followed by inflammatory process in the Kunming strain mice. The involvement of opioid and nitric oxide was studied by subcutaneous injection of morphine with/without naloxone 30 min before formalin test, or peritoneal injection of L-arginine with/without L-NAME 20 min before formalin. RESULTS: AST 20, 40, and 80 mg/kg significantly lowered pain score of the second phase of formalin response as compared with control group (P<0.01). The maximum analgesic effect of AST 40 mg/kg was found at 4 h after the administration of AST (34.4 % inhibition at the second phase). Injection of morphine 5 mg/kg significantly inhibited pain response of both phases (P<0.01) and this was reversed by naloxone 2 mg/kg (P<0.01). However, naloxone did not alter the effect of AST on the second phase. Antinociceptive effect of AST 40 mg/kg was partially blocked by L-arginine 400 or 800 mg/kg (P<0.01). CONCLUSION: AST has an antinociceptive effect on formalin test in mice that is not mediated by the endogenous opioid system but related to its inhibitory effect on the production of NO.

[AFLP analysis on genetic structure of planted Acacia auriculiformis population in Heshan].

AFLP analysis of Acacia auriculiformis populations in 3 different planted communities with the same provenance and same planting time indicated that the genetic heterozygosity was quite high in the plantation population. 90.73% of total genetic variance distributed within a population, while only 9.27% among populations. The most distinct genetic structural difference was found between pure plantation and legume mixed plantation, next was between legume and non-legume mixed plantation, and the most similar was between pure and non-legume mixed plantation, which revealed that the genetic variation in this Acacia auriculiformis population was relatively higher than that from the natural forests by isoenzymic analysis data. The genetic differentiation was not the result of gene flow, but the result of the artificial selection and the microenvironmental changes.

[A novel antiapoptosis gene, survivin, bcl-2, p53 expression in cervical carcinomas].

OBJECTIVE: To study the expression of a novel inhibitor gene of apoptosis, survivin, in cervical carcinoma and its relationship with the expression of bcl-2, p53 genes. METHODS: Using streptavidin-biotin peroxidase (SP) method, we examined the expression of survivin, bcl-2 and p53 genes in 10 normal cervical tissues and 59 cervical carcinoma tissues. RESULTS: Survivin was expressed in 41 of 59 (69.5%) cases of cervical carcinomas. In contrast, normal cervical epithelium did not express survivin. Overexpression of survivin was related to the tumor grade and clinical stage. Survivin positive cases were strongly associated with bcl-2 expression (36/45, 80.0% versus 5 of 14 cases, P < 0.005) and p53 expression (33/43, 76.7% versus 8 of 16 cases, P < 0.05). CONCLUSIONS: Apoptosis inhibition by survivin, alone or in cooperation with bcl-2 and p53 may participate in the onset and progression of cervical carcinoma. Survivin could be a new diagnostic/therapeutic target in cervical cancer.

[Analysis of loss of heterozygosity on 19p in primary gastric cancer].

OBJECTIVE: To investigate the loss of heterozygosity (LOH) frequency of microsatellite loci in primary gastric cancer samples and locate the deleted regions on 19p in which might exist human gastric cancer related genes. METHODS: The LOH of microsatellite loci on chromosome 19p was analyzed using PCR-SSLP-silver stain method in 43 primary gastric cancers and their paired normal tissues. RESULTS: In 43 primary gastric tumors, LOH was detected on the site for D19S424(29.63%), D19S216(11.53%), D19S406 (33.33%), D19S413(8.57%), D19S221(13.15%), D19S226(8.00%), D19S411(6.45%), D19S883(6.89%), and D19S886(10.71%), microsatellite instability (MSI) was found at the same time at locus D19S886 (17.85%). CONCLUSION: The most common LOH occurrence at D19S406 and D19S424 might imply the existence of the potential genes related to the tumorigenesis of gastric cancer in these loci.

Detection of p53 gene mutations in sputum samples and their implications in the early diagnosis of lung cancer in suspicious patients.

OBJECTIVES: To evaluate the value of detecting p53 gene point mutations in sputum samples and its validity and reliability as a surveillance index in the early diagnosis of lung cancer in suspicious patients. METHODS: Sputum samples were collected from 54 cases identified as lung cancer and 114 cases identified as pulmonary benign disease. The polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) was performed for the detection of point mutations at exons 5-8 of the p53 gene, and sputum smears were also used for each sample. RESULTS: The detected mutation rate of 55.56% (30/54) in the lung cancer group was significantly higher than that of 1.75% (2/114) in the control group (P < 0.001). In the lung cancer group, the sensitivity, specificity, and positive likelihood ratio (PLR) of p53 mutations as a diagnostic marker for lung cancer were 55.56%, 98.25% and 31.75%, respectively. Also, the detection rates were 35.19% (19/54) by smears and 55.56% (30/54) by PCR-SSCP-silver stain, respectively. The differences both in rate and consistency were statistically significant (P < 0.01 and P < 0.05, respectively). Moreover, statistical analysis showed no significant relations between p53 mutations and clinical parameters such as gender, smoking habits, histotypes and stages, but the detection rate of p53 mutations in older patients (> or = 60 years old) was significantly higher than that in younger patients (P = 0.02). One case with p53 mutations at exon 5 in the control group was confirmed to be squamous cell carcinoma after 4 years of follow-up. CONCLUSION: Detection of p53 gene alterations in sputum samples by PCR-SSCP-silver stain can be used as a follow-up surveillance index for the early diagnosis of lung cancer in suspicious patients.

[Chlamydia pneumoniae infection in patients with chronic obstructive pulmonary disease].

OBJECTIVE: To investigate the association of chlamydia pneumoniae infection with chronic obstructive pulmonary disease (COPD). METHODS: 61 patients with acute exacerbations of COPD, 35 patients with stable COPD and 26 healthy subjects were selected. Chlamydia pneumoniae-specific serum IgG, IgA, IgM antibodies were measured by microimmunofluorescence method. Chlamydia pneumoniae DNA in sputum were detected by nested polymerase chain reaction. RESULTS: The prevalence of acute chlamydia pneumoniae infection in COPD patients with acute exacerbation was significantly higher (31.1%) than that in patients with stable COPD (P < 0.05) and in healthy subjects (P < 0.01). The prevalences of chronic chlamydia pneumoniae infection in COPD patients with acute exacerbations (21.3%) and stable COPD patients (31.4%) were higher than that in healthy subjects (P < 0.05) The level of geometric mean titer of the serum IgA antibodies was the highest in COPD patients with acute exacerbations (20.5), intermediately elevated in stable COPD patients (10.8) and lowest in healthy subjects (3.6). CONCLUSION: Acute chlamydia pneumoniae infection is a rather frequent event in acute exacerbation of COPD, and chronic chlamydia pneumoniae infection may be involved in pathogenesis of COPD.

Relationship between hyperventilation and intracranial pressure in patients with severe head injury.

With high fatality rate and disability rate, the pathophysiologic changes of severe head injury are complicated. But the method of lowering intracranial pressure (ICP) through artificial hyperventilation is called in question recently. To understand the related changes of the partial pressure of carbon dioxide in artery (PaCO(2)) and the ICP at the acute period of severe head injury, a total of 64 patients with severe head injury were monitored and analyzed on the 3rd day after injury.
