[Differences in the intracellular potentials of human peripheral lymphocytes].

The intra-cellular potentials of human blood mononuclear cells have been measured using a microelectrode and a modified classical neurophysiological system. The results obtained in vitro suggest that different lymphocyte sub-populations and monocytes have characteristics intra-cellular potentials. We have also shown that a change from negative to positive intra-cellular potential occurs in lymphocytes stimulated by Phytohemaglutinine and Concanavaline A.

Preliminary results of phase I and II clinical trials of RFCNU, a new nitrosourea sugar derivative, in digestive tract tumours.

RFCNU or (chloro-2-ethyl)-l-(ribofuranosyl-isopropylidene-2', 3' paranitrobenzoate-5')-3 nitrosourea, a new synthetic nitrosourea derivative, which has been shown to have, in mice, among all nitrosourea derivatives tested, the longest maximallly efficient dose interval (MEDI) and which is not immunosuppressive at the smallest dose of MEDI, gave in a phase II trial on digestive tract tumours (at the dose of 400 mg/m2 per month determined by the phase I trial), 30% objective remissions among which 13% were greater than 50%.

Treatment of the mouse Lewis tumor by the association of radiotherapy and immunotherapy with Bacillus Calmette-Guérin.

Three groups of 6 to 8-week-old male mice with transplanted C57BL/6 X DBA/2F1 tumor were irradiated by a single dose of 60Co localized on the tumor in association with i.p. injections of lyophilized Bacillus Calmette-Guérin. When the animals were sacrificed, tumor parameters (weight, surface area, and volume) were determined. Lung metastases were counted using a binocular magnifying glass. When Bacillus Calmette-Guérin injections were begun the day after the irradiation, tumor growth and the number of lung metastases were significantly decreased in comparison with the control group (p less than 0.01). However, there was no significant difference between the control group, which received radiotherapy alone, and the groups in which Bacillus Calmette-Guérin treatment was started after the 4th day postirradiation. This emphasizes the importance of timing in combined radioimmunological treatment.

Relationship between Hassall's corpuscles and thymocytes fate in guinea-pig foetus.

The development and structure of the Hassall's corpuscles are studied with the light and electron microscope in 53 guinea-pig foetuses from 30 to 58 days gestation. The different stages of their life cycle are described. Prior to the appearance of Hassall's corpuscles, no medullary lymphocytes can be found in the early medulla. Hassall's corpuscles are seen initially between 35-42 days gestation. They are surrounded by large and medium-size lymphocytes, while medullary lymphocytes are in close contact with the "external cells" which exhibit cytoplasmic characteristics of a secretory function. The data reported in this paper suggest that Hassall's corpuscles are not the only sites of lymphoid cells destruction but are also "privileged" areas for maturation of medullary lymphocytes at least for the guinea-pig.

The oncostatic and immunosuppressive action of new nitrosourea derivatives containing sugar radicals.

Four new nitrosourea derivatives represent an appreciable progress in the treatment and cure of L1210 leukemia. Their therapeutic index is higher than that of CCNU and MeCCNU. Of these compounds, RFCNU may prove the most promising, as its therapeutic index is the highest of those for all the four compounds studied; moreover, unlike the other products, it is not immunosuppressive, whether administered before or after the antigen.

Granulomatous hepatitis caused by B.C.G. injection during immunotherapy of a malignant melanoma.

The authors report a case of granulomatous hepatitis after treatment by B.C.G. in a patient with malignant melanoma. The patient was treated for one year by an association of chemotherapy and lyophilized B.C.G. (Pasteur Institute). Aside from the rarity of the localization the importance of this observation resides in the discovery of B.C.G. after auramine staining in the liver suggesting that the hepatic lesions were linked to the in situ presence of mycobacteria.

[Inhibition of lymphocytes pre-incubated with human chorionic gonadotropin of the blastic transformation of peripheral murine lymphocytes].

Thymocytes cultured in medium containing human chorionic gonadotrophin hormone (HCG) are capable of blocking the response of peripheral splenic lymphocytes to mitogens in medium containing no HCG. This effect is not due to cytotoxicity. Our results suggest that the inhibition of lymphocyte response by serum from gestating subjects could be due to the presence of large quantities of HCG. This would be, at least partially, and by a direct or indirect mechanism, responsible for foetal acceptance by the mother. The implications of these findings in cancerology are manifest.

Immunological overshoot phenomenon following cancer chemotherapy: significance in prognosis evaluation of solid tumors.

Immunological function was studied in 48 patients before, during and after cancer treatment, using cutaneous tests, spontaneous rosettes test and the PHA and PWM lymphocyte stimulation tests. Chemotherapeutic drugs were used individually or in combination for 5 days a month and were preceeded by a cellular synchronization using Vincristine (1 mg/m2/day). When treatment was discontinued, we observed in 30 patients increased rates of spontaneous rosettes and of thymidine uptake. We noticed in several cases that the cutaneous tests became positive even if they were negative before and during treatment. The I.O.P. appeared 5 days after chemotherapy was discontinued and lasted 8 to 12 days. The topography of the neoplasm had no influence on the I.O.P. Failure to manifest I.O.P. is not indicative of a negative response of the tumor to chemotherapy (9 positive responses to chemotherapy out of 18 patients without I.O.P.) But the I.O.P. was a fairly constant feature in patients with positive response to chemotherapy and with favorable prognosis (28/30).

[Thymic ectopia and parathyroid tissue in the pangolin (Manis tricuspis Rafinesque)].

Thymic ectopies are under study in 45 Pangolin's thyroids (Manis tricuspis Rafinesque). Their frequency seems to be independent of the sex of the animal but this frequency also appears to be in relation to the age of the animal. The topographic and morphological studies suggest a close relationship between these inclusions made out of thymic tissue and the presence of parathyroid islets included into the thyroid capsule. Some pictures, showing a connection between parathyroid cells and thymis parenchym elements, plead in favour of a functional interelation between these different structures as the Mc Manus experiments suggest it.

[Isolation and electrophoretic characteristics of 3 lymphocyte populations of normal human blood].

Cell electrophoresis allows separation of normal human blood lymphocytes into two main groups which are a function of their relative rates of migration, with regard to the reference speed (1 mum.sec.-1V-1.cm): the lymphocytes which have a greater mobility than this value seem to be T-lymphocytes (80,1 per cent for 42 healthy adults); on the contrary, B-lymphocytes have an inferior mobility (19,9 per cent). Two known methods are used for the selection of the lymphoid populations: spontaneous rosetting with sheep's red blood cells, which are characteristic of T lymphocytes, and adherence to nylon wool columns, which is dominant in the case of B-lymphocytes. This method confirms the fact that T-lymphocytes have a rapid migration and B-lymphocytes a slow migration. We have isolated a third population, having neither the T markers, nor the B markers. It has a very homogeneous migration, centered on the two classes 1,05 and 1,10 mum.sec.-1.V.-1.cm.
