Metabolism of 2-naphthylamine and benzidine by rat and human bladder organ cultures.

The metabolism of benzidine and 2-naphthylamine, two aromatic amines which are carcinogenic for the human, was investigated in human and rat bladder organ cultures. There was little oxidative metabolism of either carcinogen in either species. In particular, N-hydroxy-2-naphthylamine, a proximate carcinogen of 2-naphthylamine could not be detected. In contrast, large amounts of the acetylated metabolites, N-acetylbenzidine, N,N-diacetylbenzidine and N-acetyl-2-naphthylamine were formed both in rat and human bladder cultures. The results suggest that metabolism of these carcinogens in situ in the bladder is unlikely to contribute to their carcinogenic effect but instead may have a positive protective role.

Assessment of methods for cytomegalovirus antibody testing of blood donations.

As part of a programme to establish a panel of individuals with high titres of anticytomegalovirus (CMV) IgG for the production of CMV Ig, the CMV serostatus of 139 normal blood donors was determined by indirect haemagglutination assay, enzyme immunoassay, and complement fixation. The indirect haemagglutination assay was the most accurate and convenient method for the detection of CMV antibody negative donations. Plasma for production of hyperimmune CMV IgG could best be selected by an IgG-specific enzyme immunoassay procedure.

Splits of the HLA DR2 antigen.

The reaction of ninety DR2 positive persons with 20 anti-DR2 sera were studied. Four of the panel and members of two families did not react with two of these anti-DR sera and were considered to have the short-split "DR2.2". DR2 positive samples reacting strongly with all anti-DR2 were designated "DR2.1".

Hepatitis B surveillance: 4-year survey of staff in a National Blood Transfusion Service.

The prevalence of HBsAg and anti-HBs in personnel of the Canadian Red Cross Blood Transfusion Service (BTS) during 1979 was not found to be significantly different from that of staff first employed in 1978-1979 or from that found among voluntary blood donors. Between 1976 and 1979, 2525 individuals were tested, of whom 13 seroconverted to HBsAg-positive; 54% of these cases appeared to be accident-related. During the same period 29 staff developed anti-HBs; of these 21% were apparently accident-related. Actual rates of seroconversion were not calculated because of staff turnover. The data suggest that BTS employees do not constitute a 'high-risk' group.

Metabolism and binding of benzo(a)pyrene and 2-acetylaminofluorene by short-term organ cultures of human and rat bladder.

The ability of organ cultures of normal human and rat bladder to metabolize the polycyclic hydrocarbon, benzo(a)pyrene (BP), and the arylamine, 2-acetylaminofluorene, has been studied. Cultures were maintained for 0 to 6 days in a chemically defined medium before incubation with [3H]BP (0.3 to 0.5 microM) or 2-[14C]acetylaminofluorene (18 to 25 microM) for 24 hr. Ethyl acetate-soluble and water-soluble metabolites were produced from both compounds by both species. The ethyl acetate extracts from [3H]BP-treated human cultures contained 9,10-dihydro-9,10-dihydroxybenzo(a)pyrene, 7,8-dihydro-7,8-dihydroxybenzo(a)pyrene, and 3-hydroxybenzo(a)pyrene. Rat bladder cultures produced similar metabolites but in slightly different proportions. Ethyl acetate-soluble products of 2-[14C]acetylaminofluorene from human cultures contained 7-hydroxy-2-acetylaminofluorene, 9-hydroxy-2-acetylaminofluorene, 2-aminofluorene, and N-hydroxy-2-aminofluorene. Rat bladder cultures produced similar metabolites, but 2-aminofluorene was found in relatively higher proportion. Hydrolysis by beta-glucuronidase of the water-soluble products produced from both carcinogens gave ethyl acetate-extractable derivatives. These hydrolyzable glucuronide conjugates were relatively more abundant following metabolism of the carcinogens by the rat than by the human cultures. Covalent binding to DNA occurred with [3H]BP in both human (19.7 +/- 13 pmol/mg DNA) and rat cultures (22.8 +/- 8.6 pmol/mg DNA). As with other human tissues, considerable variation (50-fold) was observed between individuals. The results demonstrate that both human and rat bladder epithelium can metabolize known potent carcinogens and, in the case of BP, can effect covalent binding between the products of metabolism and the urothelial cell DNA. In theory, carcinogenesis in the urinary bladder could thus be initiated by carcinogens produced or excreted in the urine without the necessity for their prior metabolism elsewhere in the body.

Practical application of synthetic A and B blood group immunoadsorbents.

The effectiveness of synthetic blood group A and B immunoadsorbents in removing anti-A and anti-B from human sera was evaluated. The sera contained specific alloantibodies and were destined for use as diagnostic reagents. Both Synsorb-A(TM) and Synsorb-B(TM) removed all saline anti-A or anti-B activity from 20 sera tested. All trace of anti-A activity detectable by the indirect antiglobulin test was removed from only 20% of sera; anti-B with similar reactivity was removed from 66% of sera. Specific (wanted) alloantibody titres were reduced by synsorb-A in 60% of sera, but were unaffected by Synsorb-B. Despite these shortcomings, such immunoadsorbents are of value particularly when cells of appropriate phenotype are unavailable for absorptions.

Mammary-tumour incidence in Sprague-Dawley rats treated with 7,12-dimethylbenz(a)anthracene: effect of pregnancy and lack of effect of unilateral lactation.

Mammary teat removal (thelectomy) was performed unilaterally in female Sprague-Dawley rats at 35 days of age. They were given 7,12-dimethylbenz(a)anthracene (DMBA) when aged either 55 days or 79 days. One third were unmated; one third were mated one week and one third mated more than 3 weeks after DMBA administration. Animals were killed when tumour-positive or after one year, when mammary lesions had developed in 99% of rats. The mean latent period for adenocarcinomas was 18.9 +/- 2.0 weeks. Benign mammary tumours, mainly secretory adenomas, developed significantly later (39.2 +/- 1.7 weeks). The rapid unilateral involution of the thelectomized glands at parturition had no effect on the localization of either adenocarcinomas or benign mammary tumours. Pregnancy and delayed DMBA administration markedly reduced the incidence of adenocarcinomas; lactation had no significant effect. In a separate experiment, precocious puberty induced with pregnant-mare-serum gonadotrophin in 30-day-old female Sprague-Dawley rats enabled their first pregnancy and lactation to be completed by 80 days of age. Parity before carcinogen administration significantly delayed the development of adenocarcinomas.

Antibody detection and identification by AutoAnalyzer haemagglutination systems.

The fundamental aims of antibody detection, the limitations imposed by present manual methodology, and what can be expected through the use of single-channel AutoAnalyzers are described. A combination of three different channels gives adequate power of detection and breadth of coverage for routine needs. Special techniques are also available that are useful the study of antigen : antibody reactions and autoimmune diseases.

Influence of local vascularity on hormone receptors in mammary gland.

Procedures, such as teat removal (thelectomy) or teat duct ligation, which prevent removal of milk, lead to rapid involution of the lactating mammary gland; performed unilaterally they have been used previously to study the biochemistry of involution, enabling a comparison of normal and involuting glands in the same animal against the same systematic hormonal environment. Both the protein hormone prolactin and the steroid hormone oestrogen are of importance in the development and function of the mammary gland. In the present experiments, female Sprague-Dawley rats were unilaterally thelectomised and the binding to the mammary gland of prolactin and oestrogen was examined through pregnancy, lactation and weaning. There was an effect of thelectomy during lactation only, when levels of both receptors increased in the intact lactating gland but failed to rise in the thelectomised, involuting gland. Capillary closure is known to occur in the mammary glands of rats after 36-48 h of milk accumulation. The rate of delivery of hormones to the tissue will be drastically reduced and it is concluded that this, rather than systemic hormone levels, is of importance in controlling receptor levels.

The non-covalent binding of benzo[a]pyrene and its hydroxylated metabolites to intracellular proteins and lipid bilayers.

The non-covalent interactions of benzo[a]pyrene (BP) and several of its hydroxylated metabolites with ligandin, aminoazodye-binding protein A (Z-protein, fatty acid binding protein) and lecithin bilayers have been studied by equilibrium dialysis, an adsorption technique and fluorescence spectroscopy. Binding affinities expressed as v/c (where v = moles of BP or BP metabolite bound per mole of protein or lipid and c = unbound concentration), were measured at concentrations sufficiently low that there was no self-association of the unbound compounds as judged by their fluorescence characteristics. 3-Hydroxybenzo[a]pyrene (BP-3-phenol), 4,5-dihydro-4,5-dihydroxybenzo[a]pyrene (BP-4,5-dihydrodiol) and 7,8-dihydro-7,8-dihydroxybenzo[a]pyrene (BP-7,8-dihydrodiol) bind more strongly (v/c = 10(5)-5 x 10(5) l x mol-1) to all three binders than does BP itself (v/c = 10(4)-7 x 10(4) l x mol-1). 9,10-Dihydro-9,10-dihydroxybenzo[a]pyrene (BP-9,10-dihydrodiol) binds to ligandin with an affinity similar to those of the other BP metabolites studied here, but binds much less strongly to both protein A and lecithin (v/c = 10(4) and 3 x 10(4) x mol-1, respectively). The low affinity of BP-9,10-dihydrodiol for lecithin would account for earlier findings that on incubation of BP with isolated rat hepatocytes, this metabolite egressed from the cells to the extracellular medium much more readily than either BP-4,5-dihydrodiol or BP-7,8-dihydrodiol. Calculations based on these results suggest that within hepatocytes BP and its metabolites, including BP-9,10-dihydrodiol, will be found almost exclusively associated (> 98%) with lipid membranes.
