Altered enzyme profile is possibly related to hepatocarcinogenesis by clofibrate.

Sequential analysis of a few biochemical markers was carried out in rat liver exposed to the hypolipidemic drug, clofibrate. A transformation marker, gamma-glutamyltranspeptidase (GGT), proliferation markers, polyamines, differentiation markers, arginase and ornithine transaminase (OTA), were chosen for the study. GGT activity was significantly reduced with an increase in glutathione concentration. Polyamine synthesis was markedly elevated 5 h following clofibrate administration. However, chronic exposure evoked only a moderate increase in polyamine profile. Hepatic arginase activity decreased significantly during the course of drug treatment. Progressive decrease in OTA, accompanied by hyperornithinemia, suggested decreased catabolism of ornithine. It is felt that these effects of clofibrate on enzyme systems unrelated to its lipid lowering action have far-reaching implications in hepatocarcinogenesis.

A rapid prognostic electrophoretic assay for serum lipid conjugates in carcinoma uterine cervix and breast.

The levels of cholesterol and lipid associated sialic acid in sera of patients with carcinoma uterine cervix and breast cancer were determined using agarose electrophoresis and precipitation method. Elevated level of lipid associated sialic acid was found, that was specifically reflected only in LDL and VLDL fractions. In carcinoma uterine cervix patients, cholesterol in low density lipoprotein increased with corresponding decrease in high density lipoprotein and there was no change in total as well as VLDL cholesterol content. But breast cancer patients were found to have higher concentrations of both total and high density lipoprotein cholesterol. A good positive correlation was found between electrophoresis and precipitation methods for the lipoprotein analysis. We recommend electrophoretic method for rapid routine analysis.

Short-term biocompatibility studies of hydrogel-grafted collagen copolymers.

Synthetic hydrogels are an interesting class of biomaterials. Hydrogels were prepared by the graft copolymerization of either HEMA and MMA or HEMA and GDMA onto soluble collagen using different cross linking agents. The tissue compatibility of these hydrogels was studied by implantation in rats. It was observed that there were no untoward rejection phenomena of these gels when implanted in vivo except for the typical healing process. The hydrogels were well tolerated and might well serve as good tissue equivalents.

Host liver changes in leukemic mice: effect of alpha-difluoromethyl ornithine, an inhibitor of polyamine synthesis.

Elevated levels of polyamines and gamma-Glutamyl Transpeptidase were seen in the liver of P388 leukemia bearing mice. There was also increase in specific activity of beta-Hexosaminidase and the B/A isoenzyme ratio. Administration of a 2% solution of alpha-Difluoromethylornithine (DFMO) immediately after inoculation of tumor cells prevented increases in polyamines in liver, but did not have any effect on gamma-Glutamyl Transpeptidase. Almost normal ratio of beta-Hexosaminidase B to A was maintained during treatment. Endogenous ornithine level was not altered both in treated and untreated mice. However, proline level was elevated in liver of untreated mice and DFMO prevented this increase. Glutathione levels were altered both by leukemia and DFMO in the host liver. The effect of drug was more prominent in the early stages rather than during terminal stages of leukemic growth.

Induction of gamma-glutamyl transpeptidase by hexachlorocyclohexane.

Hexachlorocyclohexane (BHC) induced gamma-Glutamyl transpeptidase in rat liver. The enzyme was partially purified from normal BHC fed and fetal liver and also from hepatoma. The gel filtration and electrophoretic properties of the BHC-induced enzyme was compared against that of the other three. Chemical induced hepatoma showed an additional peak of activity in Sephadex G-200 filtration. The other enzymes could be cleaved by papain to give a fraction which cochromatographed with the additional peak of hepatoma enzyme. BHC-induced enzyme and normal enzyme had similar electrophoretic mobility but differed from that of hepatoma and fetal liver enzyme which showed a slightly slower movement.

A simplified procedure for the estimation of arginine in plasma and urine using arginase.

To ensure reliable arginine estimation, other compounds giving positive results with Chinard's ninhydrin reaction were removed by simple ion-exchange chromatography. Eluted arginine was converted to ornithine by arginase and estimated using Chinard's ninhydrin reaction. Plasma arginine level in adults was in the range 56--98 mumol/l, and urine arginine in the range 0.58--1.77 mg/day.
