Intracellular production of IL-2, IL-4 and IFN-gamma by peripheral blood CD3+ cells in intermittent allergic rhinitis.

BACKGROUND: Allergic inflammation is mainly driven by type 2 T helper cells. The aim was to assess the changes in production of type 1 and 2 cytokines by CD3+ T cells dependent on natural exposure to allergens in subjects with intermittent allergic rhinitis (IAR) and in non-atopic subjects. MATERIAL: A total of 13 patients with IAR and 13 healthy non-atopics were recruited into the study. 11 patients with IAR were examined during the grass pollen season and 11 patients outside the season, 9 of them were assessed on both occasions. METHODS: A flow cytometric assessment of intracellular expression of IL-2, IL-4 and IFN-gamma by CD3+ cells was performed. For statistical analysis non-parametric tests were used. RESULTS: A tendency to decreased production of IL-4 outside the season was observed (6.94% [3.42-13.33] in season vs. 2.06% [0.7-3.6] out of season). The production of IL-4 was higher in the rhinitic group in the season than in the control group (1.93% [1.07-4.97], p=0.0034) and production of IL-2 was higher both in and outside the season (9.1% [3.94-15.09] and 10.0% [4.79-25.35] vs. 3.64% (2.64-5.03), p=0.037 and 0.045, respectively). IL-4/IL-2 and IL-4/IFN-gamma ratios were higher in the IAR group in the season than outside the season. CONCLUSION: A tendency towards a switch from a predominant type 2 response during natural allergen exposure to its suppression outside the season was found, together with a stable type 1 response.

Expression of CD20 on acute lymphoblastic leukemia cells in children.

CD20 determinant expressed on B precursors is associated with regulation of proliferation, apoptosis and maturation of these cells. The acute lymphoblastic leukemia "common" type (cALL) based on expression of CD20 is subdivided in type I and II. However, the clinical significance of CD20 expression on cALL and significance of cALL type I and II discernment are not fully elucidated. The association of CD20 expression with the expression of multidrug resistance molecule (MDR), CD34, atypical immunophenotypes of leukemia cells and response to induction therapy were determined in the group of 147 patients with acute lymphoblastic leukemia (ALL) B progenitor type (ALL-proB -14 patients) and common type (cALL-133 patients). The expression of CD20 on leukemia cells was studied routinely at diagnosis before the therapy. This expression was noted on leukemia cells of 6 ALL-proB patients (42.8%) and 66 cALL patients (49.6%). The expression of CD20 showed no association with the expression of CD34, CD22 and MDR. The reverse association was observed between CD20 expression and the presence of co-expression of myeloid (CD13, CD33, CD65, CD15) and T lymphoid determinants (CD2, CD5, CD7) on leukemia cells. The effect of induction therapy analyzed as time of blast cells cytoreduction in peripheral blood and time of reaching the complete remission showed the slower clearance of peripheral blood from blast cells associated with expression of CD20. There was no association of CD20 expression with the time of reaching the hematological remission. The above results suggested a "protective" role of CD20 against co-expression of other determinants (myeloid and lymphoid) and no association with the results of induction therapy.

[The influence of severe infections on the natural killer cells in neonates with vary gestational age].

UNLABELLED: In 27 infected neonates (16 boys and 11 girls) with birth weight ranged from 1200 to 4500 g (mean 2670 +/- 827 g) and gestational age from 31 to 41 weeks (mean 36.7 +/- 4.1), among them 11 (41%) with birth asphyxia, between 1 and 7 day of life flow cytometric immunophenotyping of the relative sizes of natural killer cells (CD3-/CD16-56+) using monoclonal antibodies of Becton Dickinson were made. The following infections were diagnosed in the neonates: bacterial septicemia (gram-positive in 13 cases, gram-negative in 5 cases), pneumonia in 6 cases and purulent meningitis in 3 cases. Control group consisted of full-term, healthy, eutrophic neonates born in Military Medical Academy in Lódź between 1994-95 years, in which immunological studies were made with the same method and dr Banasik, agreed for using her results for statistical analysis. It was stated, that in infected full-term neonates the mean relative size of NK cells was 10.7 +/- 5.1% and in prematures 12.8 +/- 7.2% and did'n differ significantly from the control. Kind of infection had no significant influence on the mean relative size of these cells, because in septic neonates was 11.7 +/- 5.7% and in other infected babies 11.6 +/- 7.2%. It was found that infected neonates without respiratory insufficiency and shock had significantly higher (13.2 +/- 6.4%) relative size of NK cells than healthy. The lowest value (7.0 +/- 4.8%) of NK cells during infections in dead neonates were noted (4 cases, among them 2 septic with shock and 2 pneumonic prematures with intraventricular haemorrhages). CONCLUSION: The severity and bad results of the treatment in neonatal infections may be due to deficit of NK cells.

[The influence of perinatal risk factors on the number of CD3+ lymphocytes and CD25+ cells in full-term neonates].

In 39 full-term neonates aged from 1 to 7 days of life, among them 24 with perinatal risk factors and 15 healthy as the control group, immunophenotyping of CD3+ and CD25+ cells using Becton Dickinson flow cytometer and monoclonal antibodies and CD3+ and CD25+ was performed. It was stated, that the mean relative and absolute number of CD3+ T lymphocytes in high risk neonates did not differ from the mean values in control group. The premature rupture of fetal membranes in mothers caused a significant increase of mean relative number of CD3+, whereas it did not influence on the absolute number of these cells. Significant increase of mean relative and absolute number of CD25+ cells in neonates with perinatal risk factors in comparison with the control group was noted. Mean relative and absolute number of CD25+ cells were significantly higher in neonates with non-infectious risk factors than in neonates with infectious risk factors. Authors emphasize that, evaluation of CD25+ cells is useful in the assessment of immunological changes in high risk neonates.

[Influence of mode and time of delivery on leukocyte subpopulations in umbilical cord blood].

DESIGN: The aim of this article was to study the differences between lymphocyte subsets of cord blood newborns according to the mode of delivery. MATERIALS AND METHODS: Lymphocytes of 15 infants born by vaginal route and 14 infants born by caesarean section were immunophenotyping by flow cytometer. RESULTS: Significant higher percentage of CD8+ was found in group of vaginal delivery. We also found high coefficient of linear regression between birthweight and CD23+, CD19+, CD4+/CD25+, NK, CD8+, CD4+, CD+3. CONCLUSION: No effect of operative delivery on percentage and absolute sizes of cord blood lymphocytes subsets except significant higher percentage of CD8+ were found.

[Evaluation of serum sialic acid concentration in chronic renal failure children on continuous ambulatory peritoneal dialysis and hemodialysis].

In various pathological conditions elevated serum sialic acid level, as the result of increased metabolism of glycoproteins and glycolipids, is observed. The study aimed at the evaluation of sialic acid concentration in serum of children on continuous ambulatory peritoneal dialysis (CAPD) and maintenance haemodialysis (HD). Examination was performed in 27 end-stage renal disease (ESRD) children including 11 on CAPD, 16 on HD treatment. Ten healthy children served as the control group (K). In CAPD group assessment was carried out during the routine monthly check up, in HD group--before (HD-1) and after (HD-2) dialysis session. Sialic acid concentration was determined using method of Shamberger. In CAPD children we obtained significantly increased serum sialic acid concentration comparing to controls and HD children. Increased serum sialic acid concentration was also found in HD children comparing to controls. There was no significant difference between the sialic acid concentrations before and after dialysis session. Analysis of correlation revealed positive correlation of sialic acid concentration with haemoglobin concentration and hematocrit in CAPD group. In children on HD treatment we showed positive correlation of sialic acid level with renal replacement therapy duration and creatinine concentration after HD. Elevated serum sialic acid concentration in dialysed children could be the result of non-specific organism response, characterised by tissue and organ damage, towards the materials of extracorporeal circulation (HD) or the presence of dialysis solution in peritoneal cavity (CAPD).

Gene discovery and product development for grain quality traits.

The composition of oils, proteins, and carbohydrates in seeds of corn, soybean, and other crops has been modified to produce grains with enhanced value. Both plant breeding and molecular technologies have been used to produce plants carrying the desired traits. Genomics-based strategies for gene discovery, coupled with high-throughput transformation processes and miniaturized, automated analytical and functionality assays, have accelerated the identification of product candidates. Molecular marker-based breeding strategies have been used to accelerate the process of moving trait genes into high-yielding germplasm for commercialization. These products are being tested for applications in food, feed, and industrial markets.

Lymphocyte subpopulations in full-term septic neonates.

PURPOSE: To estimate the absolute leukocyte and lymphocyte counts and relative and absolute sizes of CD19+ B lymphocytes, CD3+, CD4+, CD8+ and CD3+/HLA-DR+ T lymphocytes in full-term septic neonates and the influence of some perinatal risk factors on these lymphocyte subsets. METHODS: Twenty-one septic and mechanically ventilated full-term neonates (13 boys and eight girls) and 15 healthy full-term neonates born vaginally with an Apgar score > 9 and without hyperbilirubinemia were investigated. Two-color flow cytometric immunophenotyping with appropriate antibody panels using lysed whole vein blood was performed. RESULTS: The mean relative and absolute sizes of CD19+ B lymphocytes, CD3+/CD8+ and CD3+/HLA-DR+ T lymphocytes in septic neonates did not differ significantly from control. In contrast, the mean relative sizes of CD3+ and CD3+/CD4+ T lymphocytes and the CD4+/CD8+ ratio in septic neonates were significantly higher than in healthy neonates. With regard to the absolute size in septic neonates, only CD4+ T cells were significantly higher compared with the control group. Perinatal risk factors (birth asphyxia, gestation and delivery complications) had no significant effect on the relative and absolute counts of all estimated lymphocyte subpopulations in septic neonates. CONCLUSIONS: Increases in the relative sizes of CD3+ and CD3+/CD4+ T lymphocytes and the CD4+/CD8+ ratio in full-term septic neonates provides important information about changes in cell-mediated immunity during the early neonatal period.

[BAL from two different lung segments indicated by high resolution computed tomography (HRCT) in patients with sarcoidosis. I. Evaluation of alveolitis homogeneity and estimation of HRCT usefulness in selection of lung region for BAL].

The aim of the study was: 1) to evaluate the homogeneity of alveolitis by estimation of lymphocytes subsets in double BAL (2 x 120 ml) from two different lung segments: with the most (s.A) and with the least (s.B.) extensive involvement estimated by high resolution computed tomography (HRCT) 2) to examine the usefulness of HRCT as a guide method in selection the most reliable lung region for BAL. Examined group consisted of 28 sarcoid patients with homogeneous, regular distribution of nodular opacities in conventional chest X-ray (14 F, 14 M aged 19-54). Twelve patients showed homogeneous distribution of HRCT changes (RD) in lung parenchyma and 16 showed nonhomogeneous distribution of HRCT changes (ND) with domination of pathological changes in upper lobes. Eleven healthy volunteers served as controls. BAL lymphocytes subpopulations (CD3, CD19, NK, CD4, CD8, HLA-DR, CD25,) were estimated by flow-cytometry. Among patients from ND group in BAL from s.A we found the significantly higher (p < 0.01) mean total cell yield, the significantly higher (p < 0.05) mean values of % of lymphocytes (45.2 vs 36.8%) and CD4/CD8 ratio (5.3 vs 4.4) than in BAL from s.B. Also the mean values of absolute number of lymphocytes and lymphocytes CD4, HLA-DR, CD25 in ND group were significantly higher in BAL from s.A than in BAL from s.B. In RD group and in controls no significant differences between BAL findings from s.A and s.B were noticed. Our results suggest that: 1) alveolitis process is not fully homogeneous, it's intensity is greater in upper lobes with most extensive involvement of HRCT changes 2) HRCT can serve as a useful method for selection the most reliable lung region for BAL.

The aim of the study was to evaluate the role of lymphocytes T gd in sarcoidosis by estimation of T gd cells in double BAL (2 x 120 ml) from two different lung segments: with the most (s.A) and with the least (s.B.) extensive involvement evaluated by high resolution computed tomography (HRCT) and in peripheral blood. Eleven healthy volunteers served as controls. Lymphocytes T gd were estimated by flow-cytometry. In peripheral blood of patients with sarcoidosis the mean value of T gd lymphocytes (4.75%) did not differ from control group (5.3%). In all patients the mean values of T gd percentage in BAL from s.A (1.7 + 1.0%) and in BAL from s.B (2.1 + 1.5%) were significantly lower (p < 0.01) than the mean value in peripheral blood (4.75 + 2.4%) and were significantly lower than mean value of T gd cells in BAL from s.B bis (4.2 + 2.7%). Among subgroups ND and RD we did find any significant differences between values of T gd in BAL form s.A and s.B. Our results suggest minimal role of T gd lymphocytes in sarcoid pathogenesis.

[BALF from two different lung segments indicated by high resolution computer tomography (HRCT) in patients with sarcoidosis. III. Correlation between pulmonary function tests and HRCT changes and BAL cell count].

In 28 patients with sarcoidosis patients (14 F, 14 M aged 19-54) lymphocytes subpopulations were estimated in double BAL from two lung segments: with the most (s.A) and with the least (s.B.) extensive involvement estimated by high resolution computed tomography (HRCT). HRCT score for whole lung correlated negatively with DCO (r = 0.46, p < 0.05), D/VA (r = -0.46 p < 0.05), Cstat (r = -0.57, p < 0.05) and Cdyn (r = 0.-057, p < 0.01). HRCT-score for lung segments A and B did not correlate with BAL-cell count and lymphocytes subsets from these segments. The relationship between percentage of lymphocytes HLA-DR in BAL from s.A and d(A-a)O2 (r = 0.38, p < 0.05) and the relationship between absolute number of CD25 in BAL from s.A and DCO (r = -0.38, p < 0.05) were observed. The percentage of lymphocytes in BAL from s.B correlated negatively with D/VA (r = -0.40, p < 0.05) and the percentage of HLA-DR lymphocytes in BAL from s.B. correlated negatively with Cdyn (r = -0.45, p < 0.05). Our results suggests usefulness of HRCT in estimation of sarcoidosis advancement but not in it's activity and indicate the careful interpretation the relationships between BAL results from only one lung segment and pulmonary function parameters.

[Peak expiratory flow (PEF) rates evaluation in 3-year-old children].

In our study we examined 219 healthy 3-years old children. The children were exercised how to make a forced expiratory flow for about 7-10 days before the tests started. Subjects that had Chest Flexibility Index Rate equal 7.0 or higher were selected for further analysis. 174 (79.4%) children--131 from industrial region and 43 from control group--made PEF tests correctly. Use of three different peak flow meters ("Vitalograph"--Low Range model, mini-Wright--Low Range model, Personal-Best) made the results more reliable and determinant. Based on our results we concluded that the peak expiratory flow values obtained in 3-years old children can be used as reference values for assessment of PEF in healthy and ill children, the residents of an industrial region.

[The evaluation of peripheral blood lymphoid cells in children in remission a year after the cessation of treatment for acute lymphoblastic leukemia and non-Hodgkin's lymphoma type non-B].

Immunophenotype of peripheral blood lymphocytes was studied in 10 children after the cessation of treatment acute lymphoblastic leukemia and in 3 after non Hodgkin's lymphoma non-B. They all were in complete remission. Peripheral blood lymphocytes cells were analysed by flow cytometric analysis. Estimate surface antigens were estimated: CD3, CD19, CD4, CD8, CD3-16+ 56+. The results were compared with data obtained in children not treated because of neoplastic diseases. Our results showed that the absolute number of lymphocytes T(CD3+), TS (CD8+) and NK cells were significantly increased in the study group.

Sialic acid concentration in different stages of malignant lymphoma and leukemia in children.

Sialic acid concentration in the blood serum of 104 children suffering from leukemia and malignant lymphomas was determined. A significant higher concentration of sialic acid was found at the onset of the disease, compared to the controls. In the group of children suffering from acute lymphoblastic leukemia a dependence was noted between the stage of the disease and the concentration of sialic acid in the serum. The high concentration of sialic acid at the onset of the disease, although decreasing during treatment, rose again during relapses of the disease. It seems that determination of sialic acid concentration may be useful in monitoring treatment.
