The quantitation of alternative pathway complement function by timed lysis assay.
A simple timed lysis assay is described for quantifying haemolytic complement activity in human serum. Classical pathway complement function was determined by measuring the time taken to lyse 50% of a standard suspension of antibody-coated sheep erythrocytes; the time required for 50% lysis of a standard rabbit erythrocyte suspension was similarly used to evaluate alternative pathway function. Because target erythrocytes prepared on different days gave slightly different 50% lysis times, it was necessary first to construct a series of calibration curves for converting 50% lysis times into CH50 U/ml. For this purpose, a range of dilutions of the standard human serum, of known haemolytic activity, was tested against erythrocytes prepared on 10 separate occasions. The standard serum was subsequently included with each batch of unknown sera and used to select the appropriate calibration curve for direct conversion of the 50% lysis time into CH50 U/ml. Eleven samples of normal human serum were tested by both the timed lysis assay and by the dilution methods of Mayer (1971) (classical) and Platts-Mills and Ishizaka (1974) (alternative pathway). Comparable results were obtained in all cases.
One-way mixed lymphocyte culture: differences between Chinese and Caucasian stimulator cells.
In "standardized" one-way mixed lymphocyte cultures, a pool of blood lymphocytes from randomly chosen Chinese subjects was a significantly stronger stimulus to Caucasian responder lymphocytes than was a pool of allogeneic cells from Caucasians. In contrast, Chinese lymphocytes did not respond more strongly to Caucasian cells than to Chinese cells. This could not be explained by an inherent racial difference in the capacity of blood lymphocytes to undergo blastogenesis. The implications of these findings for organ transplantation and routine testing of cell-mediated immune function are discussed.

Inhibition of the mixed lymphocyte reaction by fractionated niridazole urine dialysate.
Urine dialysate from rats treated orally with 25 mg/Kg 3H-labeled niridazole was fractionated by DEAE-Sepharose column chromatography and was found to contain three radioactive metabolites and no parent compound. When human niridazole urine dialysate (NUD) was fractionated under identical conditions, fractions corresponding to the three rat NUD metabolites were found to inhibit the human one-way MLR. No inhibition was obtained with fractionated control urine dialysate. It was concluded that nonimmunosuppressive niridazole is metabolized by rats and man to produce three active compounds with the ability to suppress the in vitro response to alloantigens.

An immunoradiometric assay for the acidic ferritin of human heart: application to human tissues, cells and serum.
Human tissues contain ferritin molecules with a range of isoelectric points but immunoassays for detecting serum ferritin have generally employed antibodies to the more basic liver or spleen proteins. To study the distribution of more acidic ferritins in tissues and serum acidic ferritin has been isolated from normal human heart and a two-site immunoradiometric assay for this protein developed. This assay gives little cross-reaction with spleen ferritin. Tissue ferritins have been fractionated by anion exchange chromatography and assayed with both spleen and heart antibodies. The spleen ferritin assay detects the more basic ferritin and the heart ferritin assay the more acidic ferritin. Acidic ferritins were found in heart, kidney, reticulocytes and HeLa cells. In sera from normal subjects and patients with iron overload, myocardial infarction, leukaemia and carcinoma only low concentrations of heart ferritin were found, although in the pathological sera spleen ferritin concentrations were generally raised. Circulating ferritin contains only a small proportion of molecules with the immunological characteristics of acidic heart ferritin.

Depot penicillin as prophylaxis in accidental wounds.
A series of 285 patients attending an accident and emergency department with wounds which required suturing was divided at random into two equal groups. The first group received a single dose of depot penicillin at the time of suture and the second group received no antibiotic cover. The incidence of subsequent wound infections was not significantly reduced in patients who received penicillin.
Ethanol metabolism in male American Indians and whites.
Ethanol metabolism rates were investigated in a group of 17 male American Indians and a group of 17 male whites of similar age, education, weight, and drinking history. A moderate dose of alcohol (0.52 g/kg) was administered orally to each subject, and blood alcohol levels were determined by breath analysis. The American Indian group demonstrated a significantly faster rate of alcohol metabolism than the white group. The peak blood alcohol level was significantly higher for the American Indians than for the whites, and the American Indian group demonstrated a significantly faster absorption rate than the white group.
Ethanol metabolism and memory impairment in American Indian and white women social drinkers.
American Indian women metabolized a moderate dose of ethanol significantly more rapidly than did White women of similar age, education, weight and drinking history; however, the two groups demonstrated a similar memory decrement due to ethanol.
Immunosuppressive properties of sera and urine dialysates from kidney-graft recipients treated with azathioprine, prednisolone, and niridazole.
Niridazole, an antischistosomal agent, was given to renal transplant recipients in addition to azathioprine and prednisolone, as there is experimental evidence that this combination of drugs is highly immunosuppressive. Sera obtained from kidney-graft recipients during the first two weeks after transplantation were examined for their ability to inhibit the one-way mixed lymphocyte reaction (MLR). Sera from seven patients receiving azathioprine, prednisolone, and niridazole (triple-drug treatment), five patients receiving azathioprine and prednisolone, and two other patients treated with niridazole alone for schistosomiasis produced MLR inhibition by comparison with pretreatment (control) sera.A mean of 78% inhibition was observed with sera taken after one day's treatment with the three-drug combination, whereas this level of in-vitro immunosuppression occurred only after eight days of treatment with azathioprine and prednisolone. Niridazole alone produced an effect similar to azathioprine and prednisolone. Concentrated dialysate of urine from a patient receiving triple-drug treatment not only inhibited the MLR but also significantly prolonged the survival of heterotopic heart allografts in rats, whereas dialysate from the same patient after niridazole had been stopped gave less MLR inhibition and failed to prolong heart allograft survival.Since niridazole thus increased the in-vitro and in-vivo immunosuppressive action of azathioprine and prednisolone, we suggest that this triple-drug combination might be useful for preventing early acute kidney graft rejection.
