Structural studies of the capsular polysaccharide elaborated by Haemophilus influenza type d.

The structure of the capsular polysaccharide elaborated by Haemophilus influenzae type d has been investigated, methylation analysis and n.m.r. spectrometry being the principal methods used. It is concluded that the polysaccharide is composed of repeating units having the structure: leads to 4)-beta-D-GlcpNAc-(1 leads to 3)-beta-D-ManpNAcA-(1 leads to. In addition, single residues of L-alanine, L-serine, or L-threonine, in the proportions 2:2:1, are linked, through their amino groups, to C-6 of the 2-acetamido-2-deoxy-beta-D-mannopyranosyluronic acid residues. The degree of substitution (75-85%) varies for different preparations.

Structural studies of the O-specific side-chain of the lipopolysaccharide from Escherichia coli O 55.

The structure of the O-specific side-chains of the lipopolysaccharide from Escherichia coli O 55 has been investigated, methylation analysis, specific degradations, and n.m.r. spectroscopy being the principal methods used. It is concluded that the O-specific side-chains are composed of pentasaccharide repeating-units having the following structure [where Col stands for colitose (3,6-dideoxy-L-xylo-hexose)].(See formula in text).

Structural studies on the hexose region of the core in lipopolysaccharides from Enterobacteriaceae.

The structures for the hexose regions of cores from Enterobacteriaceae lipopolysaccharides have been investigated, using specific degradations and 1H NMR studies as the principal methods. Complete structures for these regions in the Salmonella, the Escherichia coli R1, R2, R3, R4, the E. coli K12 and E. coli B cores are proposed. Some complementary information on the structure of the heptose region has also been obtained.

Structural studies of the capsular polysaccharide from Streptococcus pneumoniae type 1.

The capsular polysaccharide from Streptococcus pneumoniae type 1 is composed of D-galactopyranosyluronic acid residues and 2-acetamido-4-amino-2,4,6-trideoxy-D-galactopyranosyl residues. The latter sugar, previously unknown in Nature, was not isolated but was identified from the products obtained on deamination of the polymer. Using n.m.r. spectroscopy, methylation analysis, and Smith degradation as the principal methods of structural investigation, it is concluded that the polysaccharide is composed of trisaccharide repeating-units having the structure: leads to 3)-alpha-Sugp-(1 leads to 4)-alpha-D-GalpA-(1 leads to 3)-alpha-D-GalpA-(1 leads to, in which Sug denotes the new sugar.

Close scavenging experimental and preliminary clinical studies of a method of reducing anaesthetic gas contamination.

Long-term exposure to low concentrations of anaesthetic gases is potentially hazardous. In spite of efforts to reduce the exposure of personnel by increased air conditioning, by scavenging of excess circuit gases and by improvements in anaesthetic equipment, high concentrations of gases have still been measured. Problems arise during mask induction, in paediatric anesthesia, and in certain situations with unavoidable leaks, especially in small rooms but also in recovery rooms. Experimental and preliminary clinical studies have shown that nitrous oxide pollution can be considerably reduced by using a separate close scavenging system in the area where high gas concentrations occur. The geometrical and flow dimensions of a close scavenging device used in paediatric anaesthesia are described.

Structural studies of the lipopolysaccharide from Salmonella typhimurium 902 (ColIb drd2).

Salmonella typhimurium infected with the plasmid ColIb drd2 gave rise to changes in the composition of the bacterial lipopolysaccharide. Bacteria carrying the wild-type ColIb, the revertant of drd2 to the wild type, or the noncolicinogenic strain resulting from the elimination of ColIb drd2, showed no changes in the sugar composition of the lipopolysaccharide. The structure of the O-specific side chains of the lipopolysaccharide produced by S. typhimurium 902, infected with derepressed ColIb mutants has been investigated. As a result of these studies, it is proposed that the O-specific side chains are composed of chemical repeating units with the following structure: (formula: see text).

Detection of extracranial carotid stenosis by computed tomography.

Ten patients with transient ischaemic attacks (T.I.A.) within the carotid territory were examined by aortic-arch angiography and computed tomography (C.T.) of the neck. C.T. showed evidence of carotid calcifications in all 17 angiographically abnormal arteries, and there was no evidence of calcification in the 3 normal arteries. C.T. of the neck promises to be a useful technique for the investigation of patients with T.I.A.s.

Structural studies of the capsular antigen of Haemophilus influenzae type c.

The structure of the capsular antigen from Haemophilus influenzae type c has been investigated, n.m.r. spectroscopy being the principal method used. It is concluded that the antigen is composed of repeating-units having the following structure: formula: (see text). O-Acetyl groups are present in approximately 90% of the repeating-units.

Structural studies of the capsular polysaccharide of Klebsiella type 33.

The structure of the capsular polysaccharide from Klebsiella type 33 has been investigated. Methylation analysis, various specific degradations, graded hydrolysis with acid, and n.m.r. spectroscopy were the principal methods used. It is concluded that the polysaccharide is composed of pentasaccharide repeating-units having the following structure. (formula, see manual). The D-galactopyranosyl group, with pyruvic acid linked as a ketal to O-3 AND O-4, was degraded on treatment of the fully methylated polysaccharide with strong base. It is proposed that methyl pyruvate is eliminated, in an E2 type of reaction.

Structural studies on the hexose region of the Enterobacteriaceae type R3 core polysaccharide.

The structure of the hexose region of the Enterobacteriaceae type R3 core polysaccharide, which is the common core in Shigella flexneri and some Escherichia coli lipopolysaccharides (LPS), has been investigated. The principal method used in these studies involved methylation analysis of the original and modified materials. The polysaccharide was modified by N-deacetylation-deamination and by Smith degradation. As a result of these studies, the following structure is proposed: alpha-D-Glcp-(1 leads to 2)-alpha-D-Glcp-(1 leads to 2)-[alpha-D-GlcNAcp-(1 leads to 3)]-alpha-D-Galp-(1 leads to 3)-alpha-D-Glcp-(1 leads to.
