Renal parenchymal volume during and after acute pyelonephritis measured by ultrasonography.

A total of 47 children with acute pyelonephritis were investigated using water delay ultrasonographic equipment (Octoson) for determination of renal parenchymal volume by the stepped section technique. Thirty two patients were repeatedly investigated every to every other week up to seven weeks. Median renal parenchymal volume during acute pyelonephritis of the right kidney was 2.70 cm3/kg body weight and of the left kidney 3.10 cm3/kg; this was significantly larger than the volume of control kidneys, which was 1.82 and 2.07 cm3/kg, respectively. The most enlarged kidneys were found among the youngest children. A significant successive decrease in renal size was found during the first four to five weeks after the acute pyelonephritis. Because of enlargement of the kidneys during acute pyelonephritis we suggest that the first renal size determination to be used for following renal growth should be performed after at least four to six weeks.

Frequency and distribution of mitomycin C-induced structural chromosome aberrations in lymphocytes from non-Hodgkin lymphoma patients.

Lymphocytes from 15 untreated patients with non-Hodgkin lymphoma (NHL) and 15 controls were exposed to 0.08 micrograms/ml mitomycin C, and the frequency and distribution of structural chromosome aberrations (chromatid and chromosome gaps, breaks, and exchanges) were analyzed in 100 mitoses per subject. The mean frequencies of aberrant cells, and gap, break, and gap + break events were 8.7, 0.9, 9.7, and 10.6 in the NHL group and 11.6, 1.1, 12.7, and 13.8 in the control group. None of the differences between the two groups was significant (P greater than 0.05). The distribution of breakpoints was nonrandom (P less than 0.001) in both groups, with a particularly marked excess of breaks in 9q11. The other breakage-prone bands were 1q11 and 1q21 in the NHL group and 1p11, 1q11, 2q31, and 16q11 in the control group. None of these hot spots coincided with any of the 60 bands known to be involved in primary chromosome abnormalities in NHL.

No difference in general well-being during antihypertensive treatment with atenolol or metoprolol CR.

Atenolol and metoprolol are the two most widely used beta 1-adrenoceptor antagonists in the treatment of essential hypertension. Differences in their physico-chemical properties have been the basis of a number of clinical studies whose primary or secondary objective has been to compare subjective well-being during treatment with these beta-selective blockers. The results are, however, contradictory, mainly due to factors such as open study design, different dose regimens and dissimilar study populations. The aim of the present study was to evaluate and compare subjective well-being during treatment with atenolol (50 mg o.d.) and metoprolol CR (100 mg o.d.) in a randomized double-blind, cross-over study (2 x 6 weeks) in hypertensives not previously treated with either of the drugs studied. Two self-administered questionnaires (MSE-profile and Jern quality of life questionnaire) were filled in both before randomization and before follow up visits at 1, 3 and 6 weeks in each treatment period. Furthermore, subjective symptoms, blood pressure and heart rate were monitored. At the end of the study, patients were requested to state what treatment they preferred. Atenolol and metoprolol CR were found to be equally effective in reducing blood pressure (from 159/98 to 144/87 and 144/88 mm Hg, respectively, n = 74). The MSE-profile and the Jern quality of life questionnaire could not detect any differences between the two treatments as regards general well-being. Other subjective symptoms (e.g. diarrhoea, bradycardia, cold hands and feet) were uncommon and equally distributed among atenolol and metoprolol patients. 31 of the patients preferred metoprolol CR, 23 atenolol, while 20 had no preference.(ABSTRACT TRUNCATED AT 250 WORDS)

Atrial natriuretic peptide deposited as atrial amyloid fibrils.

Deposition of amyloid in the atria is exceedingly common in the aging heart. We have extracted amyloid fibrils from atria and purified a major protein which had N-terminal amino acid sequence identical to that of atrial natriuretic peptide (ANP). Antisera to ANP and to the amyloid fibril protein both labelled atrial muscle cells and atrial amyloid in an identical way.

Lipomas have characteristic structural chromosomal rearrangements of 12q13-q14.

Cytogenetic analysis was performed in 10 consecutive lipomas; 6 had typical benign histology whereas four had foci of atypia. Three tumors had supernumerary ring chromosomes, 6 had different balanced rearrangements, and one had a normal karyotype. Chromosome 12 was involved in 5 of the balanced rearrangements and, although less certainly, in all the ring chromosomes, with breakpoints localized to 12q13 or q14. The other rearranged chromosomes were numbers 2, 3, 7, 8, 11, 17 and 22. These results demonstrate the non-random involvement of chromosomal region 12q13-q14 in benign lipogenic tumors. The combined data from this and previous studies on benign and malignant lipogenic tumors indicate that different levels of cytogenetic specificity exist within this group of neoplasms. We suggest that myxoid liposarcoma development requires the recombination of 2 specific chromosomal bands (12q13 and 16pII), whereas for some types of benign lipogenic tumors structural changes in 12q13-q14 may be sufficient for neoplastic growth.

Stress stimuli-induced histopathological changes in the prostate: an experimental study in the rat.

To elucidate a suggested relationship between stress stimuli and prostatis, a histological study on the genital organs of seven rats was performed. The specimens were histopathologically examined after a 10-day period, during which the rats had been submitted to standardized stress stimuli, and compared with corresponding organs from a control group of seven male rats. Inflammatory histopathological changes and large numbers of inflammatory cells were seen in the prostatic glands of all rats submitted to the long-term stress stimuli. The findings agreed with those from the prostatic glands of human males with prostatis. No corresponding changes were observed in the control group. There was no evidence indicating a bacterial origin. The results indicate a connection between stress stimuli and prostatitis.

Blood perfusion of the male genital organs--an experimental study in the rat.

Male genital blood flow and its regulation have not been fully clarified. In the present study we employed a radioactive microsphere technique to estimate the blood flow simultaneously in the major urogenital organs of male rats. The recorded genital flow rates varied among the rats. The relative blood perfusion rates of the testicles, seminal vesicles and kidneys did not differ from one animal to another. However, the blood flow in the prostate gland was not correlated to the perfusion of any of the other investigated urogenital organs.

Genital blood flow in male rats subjected to stress stimuli.

A common opinion among physicians is that some symptoms from the prostate gland are stress-dependent. However, no experimental support for this view has been presented. In the present study blood perfusion of the major genital organs of male rats were measured after they had been subjected to experimental "short- and long-term" stress stimuli. The blood-flow measurements were made by means of a radioactive microsphere technique. The used experimental "short-term" stress reduced the prostatic blood flow by approximately 50 percent. A decreased blood flow and a concomitant atrophy, indicating that the reduction in blood flow has been in force over a prolonged period of time, were noticed in all examined genital organs of the rats subjected to prolonged stress stimuli. The decreases in blood flow and weight were most pronounced in the prostate gland.

Rapid lateral diffusion of lectin-labelled glycoconjugates in the human colonic adenocarcinoma cell line HT29. Comparison with the synthetic lipid analogue diI-C14.

The lateral diffusion of lectin-labelled glycoconjugates was studied in the human colon carcinoma cell line HT29 using fluorescence photobleaching techniques. HT29 cells were grown in either Dulbecco's modified Eagle's medium with glucose (25 mM; DMEM-Glu) or with galactose (25 mM; DMEM-Gal). Cell cultivation in the DMEM-Gal medium was assumed to promote a transformation of the cells to become small-intestinal-like with characteristic microvilli and associated enzymes. The diffusion of glycoconjugates labelled with fluoresceinated Triticum vulgaris agglutinin (Wheat germ agglutinin; WGA), Ricinus communis agglutinin-I (RCA-I), Concanavalia ensiformis agglutinin (ConA), Ulex europaeus agglutinin-I (UEA-I) and Arachis hypogaea agglutinin (PNA) was in all cases rapid, with a diffusion constant (D) ranging between 0.4 and 0.8 X 10(-8) cm2 s-1. As a comparison the diffusion of the fluorescent synthetic lipid analog diI-C14 was characterized by D = 0.8 - 1.0 X 10(-8) cm2 s-1. The diffusion of lectin-labelled surface components could not be related to the presence of microvilli on HT29 cells grown in DMEM-Gal, which ought to yield an apparently lower diffusion rate. The results indicate either that surface glycoconjugates in HT29 cells are dominated by glycolipid, or that the labelled glycoproteins are more or less free to diffuse in the plane of the membrane.

Regulation of the lateral diffusion of WGA-labeled glycoconjugates in human leukocytes. Comparison between adult granulocytes and differentiating promyelocytic HL60 cells.

The regulation of the membrane mobility of glycoconjugates in human polymorphonuclear leukocytes (PMNL) was studied by comparing adult PMNL with promyelocytic HL60 cells before and after stimulation of differentiation in HL60 cells with phorbol-myristate acetate (PMA) with respect to lateral diffusion of wheat germ agglutinin (WGA)-labeled glycoconjugates. For this purpose we developed a novel variant of microscope equipment for the study of fluorescence recovery after photobleaching (FRAP) and continuous fluorescence microphotolysis (CFM) using a mini-computer for handling of shutters, data acquisition, and calculations. This equipment is presented in the report. We found that PMA-induced differentiation in HL60 cells reduced the lateral diffusion coefficient (D) of WGA-labeled membrane entities from about 1.5 to 1.0 x 10(-10) cm2/s, which was close to that found for adult blood PMNL, i.e., 1-1.2 x 10(-10) cm2/s. The lateral mobility (D x 10(10)) of succinylated WGA (S-WGA) was 2.3 and 1.7 cm2/s in undifferentiated and PMA-differentiated HL60 cells, respectively, indicating that WGA might have cross-linked membrane receptors, resulting in the slower diffusion. The results are discussed in relation to the effect of phagocyte maturation on the mobility of membrane components.

A case of myelodysplastic syndrome with high platelet counts and a t(3;8)(q26;q24).

A 62-year-old male patient with refractory anemia with excess of blasts had a t(3;8)(q26;q24) as an acquired chromosomal abnormality in the bone marrow. This aberration has not been previously reported as a sole anomaly in any type of neoplasm, including myelodysplastic or myeloproliferative disorders. The findings of disturbed thrombocytopoiesis with high platelet counts and micromegakaryocytes in the bone marrow support the idea that alteration of the transferrin receptor gene, localized to 3q26, may be of pathogenetic significance.

Clonal chromosome abnormalities in two liposarcomas.

Two liposarcomas were analyzed with chromosome banding technique. The sole chromosomal abnormality in one of the tumors, a mixed type (myxoid and round cell) liposarcoma, was t(12;16)(q13;p11), a rearrangement previously reported to be associated with myxoid liposarcoma. The other tumor, a pleomorphic liposarcoma, displayed massive numerical rearrangements (modal chromosome number 94-112), and numerous, mostly unidentifiable, marker chromosomes. The following clonal structural aberrations were recognized: del(1)(p22), del(1)(q23), t(7;?)(p22;?), i(17q), and t(19;?)(q13;?).

Blood clonal B cell excess (CBE) at diagnosis in patients with non-Hodgkin lymphoma (NHL). Relation to clinical stage, histopathology and response to treatment.

Untreated non-leukemic (lymphocytes less than or equal to 4.0 X 10(9)/l) patients with non-Hodgkin lymphoma (NHL) of B cell type often show an excess of B cells in peripheral blood bearing the same light chain isotype as the lymph node tumor cells which may indicate a leukemic spread of the disease. The ratio between kappa- and lambda-bearing lymphocytes (normal range 1.0-3.3) was studied to evaluate the prognostic significance of clonal B cell excess (CBE) at diagnosis in 110 NHL patients. In total 43% had a CBE in peripheral blood. Fifty-two per cent of the patients in clinically advanced stages had CBE and 30% of the patients in stages I and II. CBE was detected in 49% of all patients with low-grade malignant lymphoma and in 32% with high-grade malignancy. Patients with a normal kappa:lambda ratio at diagnosis entered complete remission more often than those with CBE (P less than 0.01). In patients with high-grade but not with low-grade malignant lymphomas remission duration was longer for those with normal kappa:lambda distribution than for the patients with an abnormal ratio (P less than 0.01). Survival was not statistically significantly influenced.

Electrical membrane breakdown: a possible mediator of the actions of electroconvulsive therapy.

The voltage across electrodes during electroconvulsive therapy (ECT) is two or three orders of magnitude greater than the membrane potential levels at which electrical breakdown occurs. This paper attempts to relate the interelectrode voltage to the membrane potential of individual neurones. For certain neurones the calculated values exceed the breakdown level. The study indicates that electrical breakdown does take place during ECT. It is suggested that electrical breakdown could explain some effects and side-effects of ECT.

Qualitative and quantitative changes in the guinea pig organ of Corti after pure tone acoustic overstimulation.

A direct relationship between delivered sound energy and morphological overstimulation has been formulated in the so-called equal energy principle. The validity of this principle was systematically tested by studying the effects of acoustic overstimulation on the guinea pig cochlea by means of scanning electron microscopy (SEM) and light microscopy (LM) techniques. The animals in the present study were exposed to 3.85 kHz pure tones of various durations and intensities and allowed a recovery period of 4 weeks before the morphological changes in the cochlea were assessed. A 4-grade scale was devised in order to achieve a qualitative as well as a quantitative SEM evaluation of the cochlear damage. The results of the examinations by LM and SEM were compared and analysed statistically. No significant difference was observed between groups of animals exposed to a sound energy of 9.4 and 37.5 Pa2.h, irrespective of sound intensity (114 or 120 dB SPL). Groups of animals exposed to a higher energy level (150 Pa2.h) differed significantly from those exposed to higher and also lower sound energy levels. Groups exposed to different intensities (108, 114 and 120 dB SPL) did not differ significantly within this energy level.
