[Non random position of metaphasic chromosomes. IV. Study of translocations t(7;14)(p14;q12) and t(7;14)(q35;q12)].

A study is reported on the distances between rearranged chromosomes and between their normal homologues in the case of t(7;14)(p14;q12) and of t(7;14)(q35;q12), in lymphocytes from patients with ataxia-telangiectasia (AT) and in non-affected persons. The study shows that rearranged chromosomes, in t(7;14)(p14;q12) are closer together, in non-AT persons, than their normal homologues. This is interpreted as the result of recent, repeated, and poorly transmitted mutations, in non-AT persons. On the contrary, this translocation, probably not eliminated in AT patients, becomes the most frequent.

High resolution R- and G-banding on the same preparation.

A simple method using bromodeoxyuridine (BrdU), for both cell synchronization and incorporation into replicating DNA is described. Many prophasic and prometaphasic mitoses were observed, and due to the probable blocking at different times of the cell cycle, very good R-banding and G-banding were obtained on the preparation.

Mitotic and meiotic analysis of a reciprocal translocation t(Y;3) in an azoospermic male.

A balanced reciprocal translocation t(Y;3)(q11.2q12) is detected in an azoospermic male, with a normal phenotype. Replication study shows no modification of the replication pattern of the bands of the rearranged chromosomes. Meiotic studies show a chain element at diakinesis. At the pachytene stage, the Y-body is in external contact with the sex vesicle. It is concluded that the integrity of the Y-chromosome is probably necessary for a normal meiotic process, and is more important than dosage effect due to complete or partial disomy of this chromosome.

Detection of G-C rich heterochromatin by 5-azacytidine in mammals.

A new effect of 5-ACR (5-azacytidine), observed in fibroblast or kidney cell cultures is described in the calf (Bos taurus) and in some selected primates (Cebus capucinus, Erythrocebus patas, and Cercopithecus cephus). This cytidine analogue, used at low doses during the last late S phase, prevents the condensation of R-band positive heterochromatin (G-C rich segments). Thus, 5-ACR treatment can be used as a simple method for the detection of G-C rich heterochromatin.

Non random position of metaphasic chromosomes: a study of radiation induced and constitutional chromosome rearrangements.

The rearranged chromosomes derived from reciprocal translocations or dicentric-acentric formations, observed 48 h after their induction by irradiation at Go phase, have a clear tendency to be closer together than their normal homologues. This tendency disappears in longer cultures, and does not exist when many different constitutional reciprocal translocations are considered together. It indicates that the chromosomes having exchanged segments remain adjacent at the following metaphase, and thus, that metaphase plates reflect, at least partially the interphase arrangement of chromosomes.

Tentative estimate of the risk of chromosomal disease due to radiation-induced translocations in man.

An attempt to estimate one of the parameters establishing the risk of occurrence of abnormal live-born progeny by malsegregation of radiation-induced translocation is reported. A sample of 247 2-break translocations induced by gamma-rays in human lymphocytes was studied in relation to the minimal possible imbalance they could induce in gametogenesis. These imbalances were compared with chromosomal trisomies and monosomies known to be compatible with life after birth in man. It is concluded that at least 106 out of 247 translocations should not give viable products in cases of malsegregation. A second comparison, with translocations ascertained in human subjects for various reasons, led to the conclusion that about 2/5 of the radiation-induced translocations might involve a risk of partial trisomies or monosomies. Cell survival and frequency of meiotic malsegregations are other parameters needed to make a correct estimate. A short discussion shows the difficulty of such estimates from inter-specific comparisons.

Conservation of replication chronology of homologous chromosome bands between four species of the genus Cebus and man.

Replication patterns after 5-bromodeoxyuridine incorporation are analyzed in chromosomes of four species of the genus Cebus (C. capucinus, C. albifrons, C. appella, and C. nigrivittatus). They are compared with those of man, taking as reference the banding analyses previously described. It was found that the high degree of conservation of chromosome structures between Cebus and man was accompanied by conservation of the DNA-replication sequence of the bands. It is assumed that this conservation during the course of evolution may apply to other mammals. Thus, replication patterns may be useful for ensuring interspecific comparisons. The only detected difference concerns late-replicating X chromosomes from normal female cells: The predominant lymphocyte pattern described in man is rare in Cebus, in which the usual lymphocyte pattern corresponds to that of human fibroblasts or to the minor human lymphocyte pattern.

[Cytogenetics of Cercopithecus (mona) campbelli. Comparison with other cercopithecus species and man (author's transl)].

The karyotype of C. (mona) campbelli campbelli is described after applying various banding techniques. After comparing this karyotype with those of numerous other Cercopithecus species it becomes possible to situate C. (m.) c. campbelli in the phylogeny of the group. A comparison with the human karyotype, after high resolution banding, shows that both species differ by some forty rearrangements, essentially fissions and inversions. A fission of the chromosome homologous to the human no. 10 occurred in this taxon, thus reproducing a chromosome present in Prosimii and Lagomorpha (rabbit), which can be considered as a reverse mutation.

[The karyotypes of Colobus vellersus and of C. palliatus: comparison with Cercopithecidae and man].

The karyotypes of two species of African Colobidae, Colobus vellerosus and C. palliatus, as well that of their hybrid, are described and compared. The two species differ by four pericentric inversions, indicating species divergence a relatively long time ago. Chromosome banding analogies with Papioninae, Cercopithecinae, and man are described. A common origin for Papioninae, Cercopithecinae, and Colobidae is indisputable; but, there are no arguments for considering that a close relationship exists between Colobidae and Hylobatidae as suggested by some authors.

Chromosome banding analogies between a prosimian (Microcebus murinus), a platyrrhine (Cebus capucinus), and man.

The comparative study of the karyotypes of Microcebus (Lemur, Cheirogaleid) and Cebus (Platyrrhini, Ceboid) shows important similarities between nearly all large chromosomes of these two genera. Since a relationship has been previously found between the karyotypes of Homo and Cebus, a correspondence may be found between many chromosomes of man and those of Lemurs. This study enlarges the field of comparative cytogenetics at the level of the primate order.

High frequencies of inversions and translocations of chromosomes 7 and 14 in ataxia telangiectasia.

The R-banding of more than 1100 lymphocytes and fibroblasts from 11 patients with ataxia telangiectasia (AT) showed rearrangements of chromosomes 7 and/or 14 in about 7% of the cells. Among these rearrangements, pericentric inversion of chromosome 7 was the most frequent, and the paracentric inversion of chromosome 14 not very rare. These inversions are believed to be fairly specific of AT, and their absence in the previously reported literature may be due to technical problems. Inversions were also observed in the lymphocytes of presumed heterozygote carriers with a lower frequency, and may be of some help for the detection of healthy heterozygote carriers of the AT gene.

The use of high resolution banding in comparative cytogenetics: comparison between man and Lagothrix lagotricha (Cebidae).

New techniques for obtaining high resolution banding are used in comparative cytogenetics. They make possible a direct comparison of the chromosomes of two distantly related primates, man and Lagothrix lagotricha (Platyrrhine). For many chromosomes, the homoeologies of banding are surprisingly clear. These new results strongly strengthen the interpretations proposed with the use of more conventional banding techniques.

Chromosomal evolution in Malagasy lemurs. V. Chromosomal banding studies of Lemur fulvus albifrons, Lemur rubriventer and its hybrids with Lemur fulvus fulvus.

The karyotype of Lemur fulvus albifrons, L. rubriventer and its hybrids with L. fulvus fulvus were compared with that of L. fulvus. fulvus. The two subspecies of L. fulvus have the same karyotype. L. rubriventer differs from L. fulvus by five Robertsonian translocations and also by increased heterochromatin in the juxtacentromeric region of the X chromosome. The karyotypes in the present study compared with those of other lemurs allowed us to propose a general scheme representing the chromosomal evolution of the genus Lemur. In this genus Robertsonian translocations represent 29 of the 33 chromosomal rearrangements which have so far been identified. We cannot affirm that sterility of the hybrids is exclusively due to the accumulation of Robertsonian translocations.

Chromosomal evolution of the great apes and man.

A study of all the chromosomal changes that have occurred during evolution of the great apes and man gives an indication of the phylogeny of these species. Their karyotypes differ from one another by a dozen chromosomal changes. The karyotype of their ancestor was very similar to that of living orangutans. Reconstruction of the sequence of the chromosomal changes indicates that, after the isolation of the orang-utan, a common ancestral population existed for the other species. Then, each branch leading to the living genera was independently isolated. The karyotype of the chimpanzee is an intermediary stage between that of man and the gorilla for some chromosomes. The human karyotype is less differentiated than that of the chimpanzee or the gorilla.
