Histiocytosis X: treatment with topical nitrogen mustard.

The case histories of two elderly patients with cutaneous histiocytosis X treated topically with nitrogen mustard are presented. The cutaneous lesions cleared within 2 to 3 weeks, and remission was maintained with daily topical administration of nitrogen mustard. The clinical impression of improvement was substantiated by light and electron microscopic studies prior to and after therapy.

Establishment of a cell line retaining Langerhans cell characteristics.

A discontinuous gradient of polyethylene glycol (PEG) with dimethyl sulfoxide (DMSO) was used to induced fusion between guinea pig epidermal cells (GPEC) enriched in Langerhans cells (LC) and cells of a human fibrosarcoma line (T-1080) deficient in hypoxanthine phosphoribosyl transferase (HPRT). The rapidly proliferating carrier T-1080 cells lack plasma membrane ATPase activity, Fc receptors, C3b receptors, guinea pig Ia-antigens, and nonspecific esterase activity, all characteristic of LC, and absent on other guinea pig epidermal cells. GPE/T-1080 hybrid cells were selected in hypoxanthine/aminopterin/thymidine (HAT) medium and by C3b and Fc rosette formation. A resultant continuous cell line was characterized karyotypically, histochemically, ultrastructurally and immunologically.

Complement receptors on guinea pig epidermal Langerhans cells.

A population of guinea pig epidermal cells possessed the ability to form rosettes with IgM antibody-sensitized sheep erythrocytes coated with purified human C3b (SEAIgMC1-), but not with intermediates carrying C1, C4, or C2. This population of rosette-forming epidermal cells was identified as Langerhans cells (LC) ultrastructurally and by LC depletion and enrichment studies. In addition, these cells were able to bind fluid phase C3b, which inhibited any subsequent SEAIgMC1-3b rosette formation. Rosette formation was also inhibited after SEAIgMC1-3b pretreatment with beta 1H and C3b-inactivator. LC were shown to possess trypsin-resistant C3b and Fc receptors, and unlike the temperature-independent nature of LC Fc-rosette formation, the ability of LC to form C3 rosettes was a temperature-dependent process. The finding of C3b receptors on the surface of guinea pig epidermal LC supports the concept that they are of the phagocytic monocyte-macrophage family.

Histochemical analysis of Langerhans' cells.

Single cell suspensions of epidermal cells from guinea pigs were analyzed histochemically for the presence of the following enzymes: 5'-adenosine triphosphatase, nonspecific esterase, specific esterase, myeloperoxidase and leukocyte alkaline phosphatase. A population of cells was positive for nonspecific esterase, leukocyte alkaline phosphatase, and 5'-adenosine triphosphatase. This population was identified as Langerhans' cells because the number of cells stained for these enzymes paralleled the number of Langerhans' cells in the suspension. These same enzymes were shown to be present in guinea pig leukocytes of the mononuclear-phagocytic series, suggesting that they and Langerhans' cells may have a precursor in common.

Pontiac fever. An epidemic of unknown etiology in a health department: I. Clinical and epidemiologic aspects.

In July 1968, an explosive epidemic of acute febrile illness occurred at a county health department facility in Pontiac, Michigan. Illness characterized principally by fever, headache, myalgia, and malaise affected at least 144 persons, including 95 of 100 persons employed in the health department building. The mean incubation period was approximately 36 hours. Illness was self-limited, generally lasting from two to five days. Secondary cases did not occur in family contacts and second attacks did not consistently follow re-exposure in the building. A defective air-conditioning system was implicated as the source and mechanism of spread of the causative factor. However, extensive laboratory and environmental investigations failed to identify the etiologic agent. Since these investigations a bacterium similar to or identical with the agent responsible for Legionnaires' Disease has been isolated from guinea pigs exposed to the Pontiac health department building in 1968 as well as from guinea pigs exposed to water from the evaporative condenser. Paired sera from 32 cases of Pontiac Fever showed seroconversion or diagnostic rises in antibody titers to this bacterium.

Concurrent cutaneous and hepatic hemangiomata in infancy: report of a case and a review of the literature.

Fifty-eight cases of concurrent cutaneous and hepatic hemangiomatosis in infants have been reported in the world literature. A fifty-ninth case is herewith reported and the literature is reviewed. Untreated cases have a mortality rate of 81%, whereas the mortality of treated cases is 29%. The main cause of death appears to be the consequence of arteriovenous shunting in the liver. Early and aggressive treatment by prednisone, radiotherapy to the liver, partial resection of liver, and ligation of the hepatic artery, each alone or in combinations, have been effective.

Two antigenically distinct species of human interferon.

Rabbit antisera prepared against interferon produced in human fibroblast cell cultures stimulated with poly(1).poly(C) neutralized the activity of interferon preparations produced in various human fibroblast cultures timulated either with poly(1)poly)C) or with viruses. However, these antisera showed no detectable neutralizing activity against interferon produced in cultures of human leukocytes. On the other hand, most rabbit antisera against the human leukocyte interferon were active in neutralizing both homologous interferon and fibroblast interferons. A preparation of antiserum against leukocyte interferon, active against both leukocyte and fibroblast interferons, was shown by affinity chromatography to have two distinct antibody populations, one of which was specific for the fibroblast interferon. We conclude that the heterologous neutralizing activity of sera from rabbits immunized with leukocyte interferon is liekly to be due to the presence of two antigenic species of interferon. The major antigenic species of leukocyte interferon preparations (designated "Le") is distinct from huamn fibroblast interferon. The minor species of leukocyte interferon ("F") is either identical with, or closely related to, interferon produced in human fibroblast cultures.

Effect of exercise on lung function tests in hay fever.

The patients in this study were divided into three groups on the basis of V50. Group I included completely normal subjects with normal V50 before and after exercise. Group II showed a normal initial V50 which decreased following exercise. This indicates small airways dysfunction which is exaggerated by exercise. Group II showed abnormal V50 initially which remained unchanged after exercise, suggesting more permanent impairment of the small airways function. Thus significant physiologic abnormalities were identified in patients belonging to Group II and Group III.
