Chromosomal assignment of the genes for human aldehyde dehydrogenase-1 and aldehyde dehydrogenase-2.

Chromosomal assignment of the genes for two major human aldehyde dehydrogenase isozymes, that is, cytosolic aldehyde dehydrogenase-1 (ALDH1) and mitochondrial aldehyde dehydrogenase-2 (ALDH2) were determined. Genomic DNA, isolated from a panel of mouse-human and Chinese hamster-human hybrid cell lines, was digested by restriction endonucleases and subjected to Southern blot hybridization using cDNA probes for ALDH1 and for ALDH2. Based on the distribution pattern of ALDH1 and ALDH2 in cell hybrids, ALDH1 was assigned to the long arm of human chromosome 9 and ALDH2 to chromosome 12.

Granular cell tumor of the orbit.

An 18-year-old woman had a 3-month history of increasing proptosis and visual loss secondary to an inferior orbital mass. Surgical exploration revealed an encapsulated mass in the orbit. After this mass was excised good vision was restored. The diagnosis of granular cell tumor was made as a result of various microscopic studies. The location of the tumor within the orbit is unusual and its histologic origin is uncertain. Immunohistochemistry by peroxidase-antiperoxidase method revealed that this tumor had S-100-protein-positive cells and negative results for lysozyme and alpha-1-antitrypsin. This fact suggests that the origin of the granular cell tumor may be due to the Schwann cell.

[Ultrasonographic studies on endometrial carcinoma].

Pre-operative ultrasonographic examinations on endometrial carcinoma were carried out in 34 cases in Tokushima University Hospital for 9 years since 1976. The results were compared with post-operative histological findings. The cancer mass was seen as higher echoic part than normal myometrium and its characteristic echo patterns were grouped into 5 types (4 cases of anechoic, 3 of linear, 4 of cystic, 14 of small mass and 9 of large mass). Small and large mass types were seen in 86% of the cases whose cervix, pelvic lymph nodes or parametrium were involved. The degree of myometrial invasion could be estimated by measuring the thickness of the myometrial echo uninvolved, and its coefficient correlation was 0.65. Depending on the increase in tumor echo size, myometrial invasion sometimes developed more deeply. Cervical involvements were estimated accurately in 86% by ultrasonography. Ultrasonographic estimation of the invasion on endometrial carcinoma was thought to be useful.

Existence of glucose-6-phosphate dehydrogenase-like locus on chromosome 17.

Hybridization of DNA samples prepared from flow-sorted human chromosomes with a cDNA probe for the X-linked glucose-6-phosphate dehydrogenase (G6PD) suggested the existence of the G6PD-like locus on chromosome 17. Southern hybridization analysis of endonuclease-digested DNA samples from the human-mouse hybrid cell line with human chromosome 17, and from control human and mouse cells, proved that not only X chromosomes, but also chromosome 17, contain DNA sequences that are hybridizable with the G6PD cDNA probe. The G6PD-like locus on chromosome 17 could be a putative pseudogene or a functional gene for the fetal brain-specific G6PD isozyme or other protein.
