In the present study, we used netilmicin for the postoperative complications and obtained the following results. 1. Netilmicin was used in totally 29 cases; 23 with localized peritonitis, 4 with generalized peritonetis and 2 with wound infection. Excellent effects were obtained in 2 cases, effective results in 24 cases and ineffective results in 3 cases. The effective rate was 89.7%. 2. Adverse reactions or abnormal clinical laboratory test results which might be attributable to the use of netilmicin were not noticed.

Effects of 4-ethoxy-3-methyl-5-morpholino-3(2H)-pyridazinone (emorfazone, M 73101) on thermic edema in rats were investigated, and following results were obtained: Oral administration of emorfazone (100 and 200 mg/kg) significantly inhibited the edema formation resulting from immersing the rat paws in a water bath maintained at 46.5 degrees C for 30 min. Using the coaxial perfusion technique it was found that in the 15-min period from 15 to 30 min after immersing the highest amount of bradykinin-like substance was obtained, and the release of kininogen, kinin-forming enzyme and kininase was markedly increased. Emorfazone showed a significant inhibitory action against release of bradykinin-like substance and kininogen and a tendency to decrease the release of kinin-forming enzyme. However, emorfazone had no direct effect on the content of kininogen, kinin-forming enzyme and kininase activity in rats plasma. In conclusion, it is suggested that kininogen and kinin-forming enzyme released into the extravascular space may be of importance for the biosynthesis of bradykinin-like substance, and that the inhibitory effect of emorfazone on the release of bradykinin-like substance into the extravascular space may be involved in the mechanism of analgesic and anti-inflammatory action.

Surgical specimens of carcinoma in the papilla of Vater were studied histologically. Adenomatous residue which suggested that the carcinoma was derived from a pre-existing adenoma was found in 18 cases (81.8%) out of 22. The histological transition of adenoma into carcinoma was also frequently seen.
We investigated the mechanism of the elevation of serum corticosterone level by 4-ethoxy-2-methyl-5-morpholino-3(2H)-pyridazinone (M73101) and its participation in the anti-inflammatory action. M73101 when given to rats intraperitoneally at doses of 50-200 mg/kg, caused an increase in serum and adrenal corticosterone in a dose-dependent manner. Oral administration of 200 mg/kg of M73101 also elevated the serum corticosterone level. Such action of M73101 was completely abolished by adrenalectomy, hypophysectomy, or the pretreatment with pentobarbital and morphine. In vitro experiment demonstrated that M73101 had no direct effect on adrenocortical function. These results indicate that the response to M73101 must be medicated through the release of ACTH from the adenohypophysis, which is probably due to the secretion of corticotropin releasing factor from the hypothalamus. M73101 at an oral dose of 200 kg/mg significantly decreased the volume of exudative fluid and the number of leucocytes in carrageenin-induced pleurisy of intact rats. However, the inhibitory action of this drug on cell mobilization decreased by adrenalectomy but not on exudative fluid, indicating that anti-inflammatory actions of M73101 may be due in part to pituitary-adrenocortical stimulation.

Analgesic, anti-inflammatory and other related actions of 4-ethoxy-2-methyl-5-morpholino-3(2H)-pyridazinone (M73101) were investigated in experimental animals, and the following results were obtained: Analgesic activity of M73101 was more potent than that of other anti-inflammatory drugs except for aminopyrine in phenylquinone test in mice. M73101 showed the most potent analgesic activity among the drugs tested in Randall-Selitto test in rats. The mode of analgesic action of M73101 resembled that of aminopyrine. M73101 possessed potent inhibitory activities on acute inflammatory edema and suppressed the permeability of capillary vessels. M73101 inhibited histamine release from isolated rat mast cells (in vitro) and rat skin (in vivo) by the condensation product of N-methyl-homoanisylamine formaldehyde (compound 48/80), and leucocyte emigration in carrageenin rat pleurisy. M73101 was much less active than phenylbutazone and other anti-inflammatory drugs in causing gastric lesion. Considering from therapeutic index, M73101 was found to be much superior to mepirizole, tiaramide, benzydamine, phenylbutazone and acetylsalicylic acid.

Little is known about the pathogenesis of pancreatolithiasis, and although several theories have been proposed, no experimental models of pancreatic calculi production are documented in the literature. This study examines such a model in which partial or complete pancreatic duct obstruction was surgically created in dogs. We found that (1) pancreatic calculi were demonstrated in 46.7% of the dogs (14 of 30) with partial outflow obstruction for 4 months or longer; (2) no pancreatic calculi were found in any of the dogs with complete pancreatic duct obstruction; (3) all calculi produced were localized in the ductal system; (4) the organic and inorganic composition of canine and human pancreatic calculi were quite similar; and (5) connective tissue proliferation and mucous cell metaplasia of the ductal epithelium, often seen in association with clinical pancreatolithiasis, are also detected in the pancreata of dogs that have had partial obstruction of pancreatic excretion. These findings suggest that an experimental model of pancreatolithiasis can be accomplished in dogs by the partial obstruction of pancreatic excretion.
