[A case of a perforation indicative of early gastric cancer with various macroscopic findings and a collective review of the relevant Japanese literature].

A case of perforated early gastric cancer and a collective review of the relevant Japanese literature is reported. A 83-year-old woman with free air discovered by an abdominal X-ray, was referred to our hospital. On operating, a perforated ulcer was found within and lesions, indicative of a IIc + III type early gastric cancer. Macroscopically, there was an elevated lesion at the antrum of the stomach as well. Histologically, these lesions were connected and moderately differentiated adenocarcinoma. Another lesion was also found at the anterior wall and diagnosed as IIb type gastric cancer.

[A case of malignant melanoma, axillary lymph node metastasis proved by fine needle aspiration biopsy cytology].

We report the case of a 57-year-old man with a malignant melanoma in the subunguinal region of his right thumb associated with the palpable axillary lymph nodes. Its metastasis was confirmed by fine needle aspiration biopsy cytology. Prognosis if a malignant melanoma is protean by its stage. Thus information regarding the axillary metastasis is mandatory for deciding the therapeutic plan. Fine needle aspiration biopsy cytology is a useful technique for diagnosing lymph node metastasis.

[Results of treatment of primary malignant tumor of the small intestine].

The results of treatment of 40 cases of primary malignant tumor of the small intestine are reported. The malignant tumors included 14 leiomyosarcoma, 12 malignant lymphoma and 14 adenocarcinoma. Our standard treatment for these tumors was segmental resection of the small intestine with its mesentery including the regional lymph nodes and we used additional adjuvant chemotherapy for some cases of malignant lymphoma and adenocarcinoma. Curative resection was performed in 57.1% of leiomyosarcoma, 50.0% of malignant lymphoma and 61.5% of adenocarcinoma. Cumulative 5-year survival rates were leiomyosarcoma 21.4%, malignant lymphoma 53.0% and adenocarcinoma 43.3% respectively. Serosal infiltration, lymph node metastasis and operative curability proved to be the prognostic factors. In particular, the 5-year survival rate of patients on whom curative resection could be performed was significantly better for each tumor type than that of patients for whom this was not possible, the figures being leiomyosarcoma 40.0%, malignant lymphoma 83.3% and adenocarcinoma 83.3%.

Effects of magnesium and sodium ions on the outer membrane permeability of cephalosporins in Escherichia coli.

Both Mg2+ and Na+ stimulated the outer membrane permeation of negatively charged cephalosporins in Escherichia coli without any significant alteration of the permeation of a zwitterionic cephalosporin. Such stimulation was not observed in an E. coli mutant lacking porins. The stimulation was caused by the direct interaction between the cations and the porin pores, which resulted in a decrease in cation selectivity of both the Omp F and Omp C porin pores.

Simple method of aequorin loading into platelets using dimethyl sulfoxide.

To investigate changes in Ca2+ concentrations in platelet cytoplasm during the activation, aequorin was loaded into platelets with an incubation of dimethyl sulfoxide (DMSO) in platelet suspension. When washed human platelets (about 5 X 10(9) platelets/microliter) were incubated with 10 microM aequorin and 6% DMSO (final concentrations) for 2 min at room temperature, a part of aequorin penetrated through the platelet membrane into the cytoplasm. The leakage of LDH was very slight indicating membrane damage by DMSO treatment being negligible. The platelet membrane and cytoplasmic components preserved their normal features under electron microscope. Platelet aggregation and ATP release were not affected by the incubation. The amount of permeated aequorin was the largest when DMSO was added stepwisely 6 times for 2 min to reach 6% final concentration. Though about 1/5,000 to 1/10,000 of added aequorin penetrated into platelets, the platelets emitted enough luminescent signals by additions of collagen, thrombin and A 23187. The DMSO method is very simple and can save the incubation time (only 2 min at room temperature) to load aequorin into platelets.

Differential effect of sodium butyrate on cyclic AMP phosphodiesterase activities in butyrate sensitive and resistant mastocytoma cells.

The effect of sodium butyrate on the intracellular cyclic AMP levels and the activities of cyclic AMP-regulating enzymes were examined in two types of mastocytoma p-815 cells in culture: one type (S cell) was sensitive and the other (R cell) was resistant to the induction of differentiation by sodium butyrate. In the presence of sodium butyrate, adenylate cyclase activity increased in both S and R cells to the same degree, whereas the level of cyclic AMP was elevated only in S cells. Cyclic AMP phosphodiesterase activity increased in R cells but not in S cells. Cyclic AMP phosphodiesterase activities of two cell populations differed in their response to sodium butyrate and they seem to have an important role in regulating cellular level of cyclic AMP that might be an important factor in controlling cell differentiation.
