Total sequence, flanking regions, and transcripts of bacteriophage T4 nrdA gene, coding for alpha chain of ribonucleoside diphosphate reductase.

As a part of a study of the role of bacteriophage T4-coded ribonucleoside diphosphate reductase in the switch-on and regulation of T4 DNA replication, we report the cloning and sequencing of the nrdA gene, coding for the alpha protein chain of the enzyme. The open reading frame of the nrdA gene begins 558 base pairs downstream of the 3' terminus of the td gene (thymidylate synthase). nrdB, encoding the beta chain of the enzyme, initiates 700 base pairs from the termination of nrdA. A high degree of similarity is found between the deduced amino acid sequence of the 754-residue alpha chain and the corresponding chain reported for nrdA of Escherichia coli; 56% of the residues are identical, with some segments reaching 84%. Some structural aspects of the derived alpha 2 subunit of the T4 enzyme are explored. By the S1 nuclease protection method, the RNA formed after T4 infection contains two prereplicative transcripts for nrdA, T3 and TU, and one postreplicative transcript, TL. T3 is found in low concentration. While the 5' termini of T3 and TL occur at sites near nrdA, TU apparently is a multicistronic transcript initiating farther upstream. The regulation of nrdA expression is examined in light of these findings.

An evaluation of predisposing factors for testis cancer in Ireland.

All testis cancer in Ireland (1980-1985) was analysed (n = 240). Incidence was highest in young adults (5.8/10(5)/year in those aged 25-34 years) with a moderate level in the elderly (1.8/10(5)/year in those over 75 years). Associations elsewhere with maldescent and social class were confirmed but cryptorchidism (12%) was more common than expected. Significant infertility was not identified. Associations with urban domicile (77%, p = 0.001), mental handicap (2.8%, p less than 0.05), recent vasectomy (1.2%, p = 0.009) and certain occupations were observed.

Non-seminomatous germ cell testis cancer in Ireland (1980-1985). Management, results and prognostic variables with relevance to national management protocols.

All non-seminomatous germ cell testis cancer in Ireland (1980-1985) was analysed (n = 131) and 3-year survival was: stage I = 95% (n = 58), stage II = 54% (n = 36), stages III and IV = 24% (n = 34). Countrywide results were not as favourable as those achieved in specialist units and this supports the development of a national strategy to maximise disease control. No management protocols existed during this study and the variety of approaches helped to identify the most suitable types of treatment in this broad context.

Physician-dependent prognostic variables in the management of testicular cancer.

A study of 246 cases of germ cell testicular cancer in Ireland between 1980 and 1985 confirmed that the personnel and the manner of management can significantly influence the outcome. Inadequate staging by omitting marker assays or CT scan reduced prognosis. Failure to use standard chemotherapy (PVB, BEP, POMBACE, VAB) or a reduced dosage diminished survival. Regular investigations are necessary during treatment, including marker assays, chest X-ray and CT scan. Frequent monitoring is important in later follow-up. Management by a urologist improved survival, especially if he was involved from the outset. The results also favoured the concept of combined management by a urologist and an oncologist.

Effects of pregnancy and lactation on the metabolism of bovine adipose tissue.

The incorporation of [2-14C] acetate into the triglycerides of adipocytes from bovine omental adipose tissue was at a minimum during peak lactation but increased again in late lactation. Similar but somewhat greater changes were observed for adipocytes from subcutaneous adipose tissue. In both tissues the activities of four enzymes concerned with fatty acid synthesis were lowest during peak lactation. In omental tissue lipoprotein lipase activity was lowest during peak lactation but there was a marked increase in its activity in the udder during lactation. In both omental and subcutaneous adipocytes the rate of glycerol release at peak lactation was higher than at all other stages of pregnancy and lactation. At peak lactation plasma insulin levels were low but growth hormone levels were higher than at all other stages of pregnancy and lactation. These results suggest that lipogenic precursors were diverted away from adipose tissue in early lactation and stored triglycerides were made available for use elsewhere in the body.

Violent delinquency: an examination of psychopathic typologies.

This study sought to determine the combined effects of psychopathy, low intellectual functioning, and love deprivation on violent delinquency. Low-intellectual-functioning psychopaths were significantly more violent than were low or high-intellectual-functioning nonpsychopaths or high-intellectual-functioning psychopaths. Love deprivation was more strongly related to violence than was psychopathy/intellectual functioning, and severely love-deprived, low-intellectual-functioning psychopaths were the most violent.

Love deprivation, Wechsler performance greater than verbal discrepancy, and violent delinquency.

This study explored the relationship among love deprivation, Performance greater than Verbal discrepancy, and violent crime in a sample of juvenile probationers. Love deprivation and P greater than V discrepancy was significantly related to violent delinquency after adjusting for the effects of both race and social class, two variables often closely associated with violent delinquency. Also, love deprivation was strongly related to P greater than V discrepancy. We speculated that high performance IQ scores relative to verbal scores are a function of a lack of disruption in the short-term memory mechanisms of high P greater than V subjects, that this nondisruption is a function of hyporeactive autonomic nervous systems, and that this hyporeactivity could itself be a function of early love deprivation.

Patients' delay in the presentation of testis cancer in Ireland.

A study of 217 cases of testis cancer in Ireland revealed a longer duration of symptoms (median 2.8 months, mean 10 months) than elsewhere: 32% of patients waited at least 6 months before seeking medical advice. Delay was associated with metastases (P = 0.001), diminished prospects of cure (P = 0.001) and increased mortality (P = 0.002) but not with marker status or complexity of treatment required. Eleven per cent did not notice a detectable swelling and 25% waited at least 3 months after such a discovery. These facts and the incidence of maldescent (12%, with more than two-thirds untreated) highlight the need for better health education in this area.

Violent crime, sociopathy and love deprivation among adolescent delinquents.

Ever since Wechsler observed that the adolescent sociopath characteristically scored higher on the performance section of the Wechsler IQ scale relative to the verbal section, psychologists have been debating its meaning. This study examines the relationships between P-V discrepancy scores, love deprivation, and juvenile delinquency among a sample of juvenile probationers. P greater than V scores were significantly related to love deprivation and violent crimes. It was determined that much of the explanatory power of P greater than V with regard to violent delinquent behavior was mediated by love deprivation. We also found that P greater than V scores could not be explained away by social class, low verbal scores, or lower full-scale scores. Only love deprivation was found to be significantly related to P greater than V scores in our path model, thus offering some support to the assertions of some authors that early emotional stresses affect subsequent autonomic nervous system function.
