Physiological and structural analysis of light-harvesting mutants of Rhodobacter sphaeroides.

Two mutants of Rhodobacter sphaeroides defective in formation of light-harvesting spectral complexes were examined in detail. Mutant RS103 lacked the B875 spectral complex despite the fact that substantial levels of the B875-alpha polypeptide (and presumably the beta polypeptide) were present. The B800-850 spectral complex was derepressed in RS103, even at high light intensities, and the growth rate was near normal at high light intensity but decreased relative to the wild type as the light intensity used for growth decreased. Mutant RS104 lacked colored carotenoids and the B800-850 spectral complex, as well as the cognate apoproteins. This strain grew normally at high light intensity and, as with RS103, the growth rate decreased as the light intensity used for growth decreased. At very low light intensities, however, RS104 would grow, whereas RS103 would not. Structural analysis of these mutants as well as others revealed that the morphology of the intracytoplasmic membrane invaginations is associated with the presence or absence of the B800-850 complex as well as of carotenoids. A low-molecular-weight intracytoplasmic membrane polypeptide, which may play a role in B800-850 complex formation, is described, as is a 62,000-dalton polypeptide whose abundance is directly related to light intensity as well as the absence of either of the light-harvesting spectral complexes. These data, obtained from studies of mutant strains and the wild type, are discussed in light of photosynthetic membrane formation and the abundance of spectral complexes per unit area of membrane. Finally, a method for the bulk preparation of the B875 complex from wild-type strain 2.4.1 is reported.

Transvesical and transabdominal closure of vesicovaginal fistulas.

Between 1973 and 1985, a total of 46 patients were operated for vesicovaginal fistula. Depending on the extension and localization of the fistula, transvesical or transperitoneal closure was performed. In 12 patients ureteral neoimplantation was made. Fistula recurrence was observed in 6 cases. Based on their favourable experience, authors recommend this surgical solution for the closure of fistulas adjacent to the ureteral orifices and/or for that of larger fistulas.

The detection of complement binding immune complexes by means of 51Cr-labelled indicator cells.

A radioimmunoassay is described for the detection of circulating complement binding immune complexes. The method was found to be highly sensitive but of limited specificity. It is based on the ability of the complexes to bind added complement (C1q-component) so that they are not available for the lysis of 51Cr-labelled chicken red blood cells in a haemolytic system. Sera from 241 patients and 87 healthy persons were studied for the presence of circulating immune complexes. The complement binding capacity is dependent on the disease groups. Patients with immune complex diseases showed a high significant difference to the healthy persons. The experimental conditions and the quality control of the method were described. There was found a very good correlation between complement binding capacity and concentration of heat-aggregated immunoglobulin.

Bilateral urological tumours.

In the 5-year period between January 1, 1977 and December 31, 1981, 6 patients with bilateral urological tumours localized in various organs were treated at the Department of Urology, Debrecen University Medical School. As compared to earlier results, the survival of patients with bilateral adrenal renal and testicular tumours may be improved by the early diagnosis, immediate and adequate treatment, by a careful follow-up lasting for years and, when needed, by a delayed intermittent chemotherapy.

[Immunologic significance of the inflammatory stromal reaction in malignant tumors].

In an investigation of correlation and factor analysis the relations between cell-mediated stromalreactions and immunological parameters are checked in 63 patients with malignant tumours. The stromalreaction is determined semi-quantitatively as a cell-mediated infiltration of the tumour by neutrophiles, lymphocytes, plasma cells and macrophages. In regard to immunological parameters circulating immuncomplexes, K-cell-activity, auto-antibodies, B- and T-cells and immunglobulins are determined among others. It is shown that the stromalreaction is to be regarded as a part of an immunological answer of the organism against the tumour.

[Indirect immunofluorescence with autoantibodies on cryostat sections of human tumors].

31 carcinomata, 14 benign neoplasms, 3 mastopathies and human placenta tissue were investigated with autoantibodies of defined specificity using an indirect immunofluorescence method. In the cells of carcinomata and the trophoblast layer of the placenta studied, immunofluorescent staining with antimitochondrial antibodies were strongly positive and membranously when compared with cells of benign tumors. The negative results obtained with other autoantibodies in the same material indicate the specificity of the reaction.

[Relevant prognostic results in stomach cancer (analysis of variance and discriminance with clinical, morphologic and immunologic features)].

In 47 patients with an endoscopic-bioptic reliable gastric cancer two prognostic groups (good/poor) were set up by the help of clinical TNM-determination (gastroscopy, laparoscopy, operation), histology (bioptate and resectate) and long-term observation. In order to determine their prognostic significance 37 clinical, morphological and immunological parameters were recorded and analysed by the help of a multivariate analysis of variance and discriminance. The TNM-stage, erythrocyte sedimentation rate, the acid mucopolysaccharides in the tumour, circulating, complement binding immuncomplexes, B-cells, IgM, C3 and C4, IgE, gastritic degree and the infiltration of the tumour by neutrophile granulocytes and lymphocytes prove to be prognostic-discriminatoric significant. The parameters allowed a retrogradually-mathematically reliable classification of the patients into one of the both prognostic groups. The remaining features (age, sex, duration of the case history, Rohrer-index, tumour superficial spread, histological differentiation, neutral mucopolysaccharides, gastric activity, plasma cells and macrophages as stroma cells, IgG, IgA, auto-antibodies, dysproteinaemies, PEG-precipitate, T-cells, LAI-test, haemoglobin, total leucocyte count and total protein) prove to be redundant, i.e. they are unnecessary for calculation of the prognosis.

Effects of drugs influencing the cAMP level on hippocampal seizure activity.

Changes of hippocampal EEF seizure activity elicited by electrical stimulation of the hippocampus or penicillin injection into the hippocampus were investigated under the effect of locally applied drugs influencing the cAMP level in the brain. It was observed that some drugs which elevate the cAMP level such as papaverine, histamine +K+ and dibutyryl cAMP elevated the electric seizure threshold while in penicillin-induced epilepsy they reduced the occurrence of ictal activity and the interictal spike frequency. These drugs when applied before penicillin prolonged the time necessary for development of the epileptic focus. The effect of imidazole was the opposite in every respect. On the basis of these data the possible role of cAMP in the pathomechanism of epilepsy is discussed.
