Distribution of movement-related cortical potential upon jaw-biting in humans.

We examined the cerebral location of the readiness potential (hereinafter referred to as "RP") upon activation of the masseter muscle by voluntary and unilateral jaw-biting movement. Four normal adults served as subjects. Five scalp electrodes were placed according to the international 10-20 method at CZ, C3, C4, T3 and T4. In addition, in one of the subjects, RPs were recorded by 12 scalp electrodes in order to study changes occurring in the distribution of RPs with time. The maximum amplitude of the RP was located at T4 in the temporal area, which was involved in the biting movement as a negative slow potential occurring 1.4-0.8 s before the beginning of the discharge to the masseter muscle. The maximum amplitude of the RP on the contralateral side was located at T3. This meant that the amplitude of the RP tended to be higher on the same side as the biting movement than on the contralateral side in all subjects. From scalp topography, the maximum amplitude of the RP was shown to be confined to T4, and was especially marked immediately before the beginning of discharge to the masseter muscle. From these results it is thought that in the case of voluntary biting movement, the RP reflects activities of the pyramidal cells in the masticatory area of the cortex, and that strong descending impulses from both sides then reach the masseter muscles.

Effects of X-ray irradiation on natural killer (NK) cell system. I. Elevation of sensitivity of tumor cells and lytic function of NK cells.

The in vitro effect of X-ray irradiation on the human natural killer (NK) system was studied. When K562 cells were irradiated with X-rays and cultured for 18 hours, they showed increased sensitivity to lysis by blood lymphocytes and purified large granular lymphocytes (LGL). The X-ray-induced augmentation was observed as little as 2 Gy irradiation, reaching maximum at 5 to 20 Gy. The doses of X-rays did not influence the viability and spontaneous release of the target cells. On the other hand, irradiation with X-rays of NK cells at 5 to 15 Gy resulted in a transient increase in NK activity at 1 hour, and then the activity declined and was completely lost after 24 hours. However, when LGL were cultured with interferon immediately after irradiation, they maintained elevated NK activity. These results suggest the possible use of low doses of X-ray irradiation in combination with biological response modifiers for treatment of cancer.

Effects of X-ray irradiation on natural killer (NK) cell system. II. Increased sensitivity to natural killer cytotoxic factor (NKCF).

Irradiation with low-doses of X-rays of tumor cells elevated their susceptibility to lysis by natural killer (NK) cells in an accompanying paper. Cytotoxicity assays conducted at the single cell level revealed that X-ray irradiation of K562 cells did not affect the number of effector-target conjugates but increased the frequency of dead conjugated target cells. During interaction with K562 cells large granular lymphocytes released a soluble cytotoxic factor (NKCF) that killed the target cells. X-ray irradiation did not affect the NKCF stimulatory ability of K562 cells, while it elevated their sensitivity to the lytic effect of NKCF. In contrast to X-rays, exposure to ultraviolet (UV) radiation of K562 cells did not elevate their NK sensitivity but rather reduced it. Treatment with mitomycin C produced no effect on NK sensitivity. These results indicate that X-ray irradiation elevates the target sensitivity to NKCF, which may be involved in the increased NK sensitivity, and that the X-ray effect may be different from that of UV radiation or DNA synthesis inhibition.

[Clinical evaluation of limb-salvage procedures in malignant bone tumors--a study of 54 cases].

Recent advances in therapy such as local and systemic chemotherapy, wide resection, and aggressive thoracotomy improved the survival of the patients with malignant bone tumors. Since 1981, we have performed limb-salvage operations for 54 cases of malignant bone tumors of the extremities. In this study, we report the rate of local recurrence and the prognosis of the patients. We treated 37 osteosarcomas, 8 malignant fibrous histiocytomas of bone, 3 Ewing's sarcomas, 3 parosteal osteosarcomas, 2 chondrosarcomas, and one fibrosarcoma. The age of the patients ranged from 11 to 65 years (mean, 18.5). The male/female ratio was 2:1. The surgical stage was 47 at II B and 7 at III. The follow-up period was 14 to 84 months (mean 36). They received preoperative chemotherapy including intraarterial infusion of Adriamycin, and postoperative chemotherapy for ten months. Local recurrence was detected in 2/54 (3.7%). Lung metastases (Stage II B) developed in 9/47 (19.1%). Five of them (10.7%) died of metastasis, one is alive with metastases, and 41 cases are now disease-free. Six of seven cases (Stage III) died of metastasis, despite aggressive chemotherapy and thoracotomies.

[Development of a slow release system of anti-cancer drugs retained in calcium-hydroxyapatite ceramic].

A new type of anticancer material with sustained release by enclosure of cisplatin (CDDP) into porous calcium hydroxyapatite ceramic (CDDP-CHA) was developed. A slow release of anticancer drug from CDDP-CHA was confirmed in in vitro experiments. When this material was implanted into normal thigh muscle of mouse, a sustained release of CDDP was observed during over 8 weeks after implantation. The diffusion of drug into blood and other organs from the implanted site was very small. CDDP-CHA placed into the implanted tumor of mouse allowed a slow release and high drug level from the material in the tumor. The concentration of the drug in other organs such as liver and kidney was very low compared with that of the tumor. These results suggest that this material has an important role for cancer chemotherapy, particularly in bone tumors for the reasons of calcium hydroxyapatite ceramic carrier having a mechanical strength.

[Ileovesical fistula complicating Crohn's disease: a case report].

A 43-year-old woman was admitted on October 6, 1987 with the chief complaint of fecaluria, pneumaturia and miction pain. She had been diagnosed as Crohn's disease in March, 1987. Urinalysis revealed numerous leucocytes, and streptococcus faecium was identified by urine culture. Contrast film of small intestines showed ileovesical fistula arising from terminal ileum. Cystoscopy revealed a papillary tumor-like appearance at the dome of the bladder. An operation was performed on November 9 under the diagnosis of ileovesical fistula complicating Crohn's disease. It was found that ileal region formed a hard adhesion to the bladder wall. Partial resection of the ileum and bladder was performed. Ileovesical fistula was found in the adhesion. Histological diagnosis of the affected ileum was Crohn's disease, showing noncaseating granuloma with the multinucleated giant cells. This case is the first report of female urological complication of the Crohn's disease in the Japanese literature.

[Mixed gonadal dysgenesis and gonadal tumor--growth of seminoma after 27 years of an exploratory laparotomy].

A case of mixed gonadal dysgenesis with anaplastic seminoma is herein reported. A 38-years-old man was admitted to our hospital with the chief complaint of a painful mass, 19.0 cm in long diameter in his lower abdomen. At 11, 13 and 14 years old he had the history of treatment for an ambiguous external genitalia and defect of both scrotal contents. In the concrete, laparotomy, chordectomy and urethroplasty were performed at those times and the laparotomy revealed a right immature abdominal testis and Müllerian structures. Laboratory examination revealed elevated levels of serum lactic dehydrogenase and HCG. The karyotype from the cultured lymphocyte was that of a normal male. Because the histological finding of biopsied abdominal tumor was shown to be an anaplastic seminoma, firstly two courses of chemotherapy using cisplatin vinblastine and bleomycin were induced, and that after an extensive resection for the tumor and Müllerian structure including some portions of surrounding tissue was performed. We reviewed 40 cases of mixed gonadal dysgenesis reported previously in the Japanese literature and 8 cases of that with a gonadal tumor.

[Study on hypoosmotic swelling test for infertile male patients].

The hypoosmotic swelling test was done on 74 infertile men, 7 patients who achieved pregnancy by AIH (AIH group) and 10 fertile men and the usefulness of its test was assessed. 1) The rate of total swollen sperm and that of g type swollen sperm from infertile men were significantly lower than rates in the patients in the AIH group and fertile men. 2) There were significant but weak positive correlations between semen analysis (total sperm concentration, motility, motile sperm concentration) and hypoosmotic swelling test results (total swollen sperm, g type swollen sperm). 3) When the cut off value of the rate of total swollen sperm and g type swollen sperm was set at 42% and 14%, respectively, rates were above the cut off value except for g type swollen sperm in patients in the AIH group.

[Clinical study of terodiline hydrochloride on nervous pollakisuria].

TD-758 (12 mg tablets) was given in two tablets daily to 19 patients with nervous pollakisuria. The changes in frequency of daily micturition and in subjective symptoms were studied referring to the diary kept by each patient, along with the occurrence of side effects. Before the medication, urodynamic studies were made on each patient and all the cases were thus confirmed being free from detrusor instability or uninhibited contraction. The wash-out period of other drugs and control observation period, each one week, were set before the medication period which lasted for four weeks. The frequency of micturition per day showed a significant decrease from the first week of the medication period (p less than 0.05), and at the fourth week there was noted a significant reduction by twice per day on the average (p less than 0.001). Subjective efficacy was noted as follows: excellent in 3 cases (15.8%), good in 9 cases (47.7%), unchanged in 7 cases (36.8%), and thus 12 cases (63.2%) showed an efficacy of the rating of good or higher. In four cases dry mouth and in two cases sense of abdominal distention were noted out of the total of 19 cases, but no side effects of severe degree to stop the medication were noted during the medication period. In conclusion TD-758 is a clinically useful remedy for treatment of nervous pollakisuria.

Generation of cytotoxic factor by human large granular lymphocytes during interaction with autologous tumor cells: lysis of fresh human tumor cells.

Large granular lymphocytes (LGL) obtained by centrifugation (Percoll gradient) of blood from patients with carcinomatous pleural effusions and solid tumors lysed autologous, freshly isolated tumor cells in a 4-hour 51Cr-release assay. When cocultured with autologous tumor cells, LGL released soluble cytotoxic factor(s), large granular lymphocyte-derived cytotoxic factor (LGL-CF). In contrast, small T lymphocytes were unable to kill autologous tumor cells or to produce a cytotoxic factor. The LGL-CF demonstrated cytotoxicity against autologous fresh tumor cells but also against allogeneic fresh tumor cells in a 48-hour microcytotoxicity assay and in an 18-hour 51Cr-release assay in the presence of dactinomycin. Binding of LGL-CF to autologous tumor cells occurred within 2 hours and reached the maximum by 6 hours; this binding was sufficient to cause subsequent lysis of the target cells without the continued presence of LGL-CF. Neither the supernatants produced by culture of LGL alone nor the lysates of LGL had detectable cytolytic activity. In addition, treatment of LGL with cytochalasin A inhibited both direct cell-mediated autologous tumor lysis and generation of LGL-CF. Production of LGL-CF required active cell metabolism and protein and RNA syntheses in LGL, but not DNA synthesis. Addition of dactinomycin to LGL-CF in assays augmented the lysis. LGL-CF was stable at 56 degrees C, but it was reduced at 70 degrees C and destroyed at 100 degrees C. Treatment of LGL-CF with trypsin or proteinase K reduced or abrogated the lytic effect, respectively, while the lytic effect was not affected by papain, catalase, or superoxide dismutase. These results indicate that during interaction with autologous tumor cells, LGL release a soluble cytotoxic factor that mediates lysis of fresh human tumor cells.

Autologous mixed lymphocyte-tumor reaction and autologous mixed lymphocyte reaction. II. Generation of specific and non-specific killer T cells capable of lysing autologous tumor.

The specific and non-specific nature of autotumor cytotoxicity induced in autologous mixed lymphocyte-tumor culture (AMLTC) and autologous mixed lymphocyte culture (AMLC) was studied in patients with carcinomatous pleural effusions. Small- and medium-sized blood lymphocytes that were isolated by centrifugation on discontinuous Percoll gradients did not lyse autologous, freshly isolated effusion tumor cells. In vitro activation of the small lymphocytes, but not of the medium lymphocytes, with autologous tumor cells generated cytotoxic potential restricted to autologous tumor. When stimulated with autologous non-malignant non-T cells, the medium lymphocytes, but not small lymphocytes, were triggered to cytotoxicity that acted not only on autologous tumor cells but also on allogeneic tumor cells, T blasts, and tumor cell lines. Experiments using monoclonal antibodies (MAb) and complement (C') showed that both types of killer cells were CD2+ CD3+ CD16- T cells. Autotumor cytotoxicity developed in AMLTC was mediated by the CD4- CD8+ T cell subset in 6 of 9 cases and the CD4+ CD8- subset in the other 3 cases. In contrast, cytotoxicity induced in AMLC was exerted exclusively by the CD8+ subset. The enrichment of blasts from cultured T cells on discontinuous density gradients enhanced autotumor killing activity, with no reactivity recorded for blast-depleted, resting T cells. Addition of mitomycin-C-treated large granular lymphocytes (LGL) to AMLTC abolished the induction of autotumor killer cells, whereas non-specific killer cells were generated in AMLC irrespective of the presence of LGL. These results indicate that stimulation of autoreactive T cells in AMLTC and in AMLC could induce 2 distinct types of autotumor killer cells.

Hyporeactivity to mitogens of retroplacental blood lymphocytes in human pregnancy.

Retroplacental blood lymphocytes (RPL) obtained from women without complications at the term delivery were studied for proliferative response to mitogens and surface phenotype and were compared with autologous peripheral blood lymphocytes (PBL). Proliferation of RPL induced by phytohemagglutinin (PHA) was significantly lower than that of PBL in 11 of 13 cases. Also RPL proliferated poorly in response to concanavalin A (Con A) in 8 of 10 cases. On the other hand, flow cytometry analysis of T cell subsets revealed that RPL contain comparable or higher number of CD3+ cells and CD4+ cells and similar numbers of CD8+ cells as compared with PBL in most cases. These results indicate that T cell proliferative response in retroplacental space is impaired, which could be due to a functional defect of RPL.

Depression of proliferative response to fetal lymphocytes of retroplacental blood lymphocytes in human pregnancy.

Retroplacental blood lymphocytes (RPL) in human pregnancy were studied for proliferative response to related fetal lymphocytes and it was compared with that of peripheral blood lymphocytes (PBL) of the same donor. RPL proliferated poorly to related cord blood lymphocytes in 9 of 10 cases when compared with autologous PBL. RPL also showed lower proliferative response to unrelated adult lymphocytes in 7 of 12 cases. Proliferation of RPL was weaker to related cord blood lymphocytes than to allogeneic adult lymphocytes. These results indicate that the response of T cells at feto-maternal interface is impaired, especially to fetal antigens.

Complete amino acid sequence of ferredoxin from Peridinium bipes (Dinophyceae).

The amino acid sequence of the major ferredoxin component isolated from a dinoflagellate, Peridinium bipes, was completely determined. Staphylococcus aureus V8 proteolytic, tryptic and chymotryptic peptides of Cm-ferredoxin were prepared and sequenced. The sequence was Phe-Lys-Val-Thr-Leu-Asp-Thr-Pro-Asp-Gly-Lys-Lys-Ser-Phe-Glu-Cys- Pro-Gly-Asp-Ser-Tyr-Ile-Leu-Asp-Lys-Ala-Glu-Glu-Glu-Gly-Leu-Glu-Leu-Pro- Tyr-Ser - Cys-Arg-Ala-Gly-Ser-Cys-Ser-Ser-Cys-Ala-Gly-Lys-Val-Leu-Thr-Gly-Ser-Ile- Asp-Gln - Ser-Asp-Gln-Ala-Phe-Leu-Asp-Asp-Asp-Gln-Gly-Gly-Asp-Gly-Tyr-Cys-Leu-Thr- Cys-Val - Thr-Tyr-Pro-Thr-Ser-Asp-Val-Thr-Ile-Lys-Thr-His-Cys-Glu-Ser-Glu-Leu. It was composed of 93 amino acid residues with 7 cysteine residues, the highest number found among the chloroplast-type ferredoxins so far sequenced. A cysteine residue was found for the first time at the 89th position in a chloroplast-type ferredoxin. Calculation of the numbers of amino acid differences among chloroplast-type ferredoxins indicates that the Peridinium ferredoxin is far divergent not only from higher plant ferredoxins but also from blue-green algal ferredoxins.

The effect of mechanical stress on cultured growth cartilage cells.

Mechanical stress seems to influence the growth of bone, but little is known about the transduction of mechanical forces into biochemical signals. The present study was done to resolve the important question of how a mechanical stimulus is transduced into a metabolic response by chondrocytes. Cultured growth cartilage cells isolated from rat ribs and subjected to mechanical stress (tensile force) showed a significant increase in intracellular cyclic AMP levels, but no increase in prostaglandin E2 levels. In glycosaminoglycan synthesis measured by radioactive sulfate incorporation, long-term exposure of chondrocytes to mechanical loading caused increased synthesis. Moreover, long-term exposure of chondrocytes exerted an alteration of responsiveness to parathyroid hormone and calcitonin measured as intracellular cyclic AMP content. These results suggest that mechanical stress can alter bone growth by modulating the metabolism of growth cartilage cells.

Osteogenic capacity of cultured human periosteal cells.

We developed a culture system of cells isolated from juvenile human periosteum. The culture consisted of epithelial-like and fibroblast-like cells. Both types of cells had intense alkaline phosphatase activity maintained in subculture. When these cells, loaded into diffusion chambers, were implanted subcutaneously in rats, cartilage tissue was mainly formed and bone was seen scantily. 1 alpha-OH-D3 and 1,25-(OH)2-D3 increased the alkaline phosphatase activity and proteoglycan synthesis in the periosteal cells. Calcitonin also stimulated the proteoglycan synthesis, but parathyroid hormone had no effect.
