Deletions of the iso-1-cytochrome c and adjacent genes of yeast: discovery of the OSM1 gene controlling osmotic sensitivity.

Some of the deletions in the yeast Saccharomyces cerevisiae that encompass the CYC1 gene, which determines iso-1-cytochrome c, extend into the OSM1 gene, causing inhibition of growth on hypertonic media, and into the RAD7 gene, causing sensitivity to UV light. Two deletions (cyc1--363 and cyc1--367) encompass only the CYC1 gene, two deletions (cyc1--366 and cyc1--368) encompass the CYC1 and OSM1 genes, three deletions (cyc1--1, cyc1--364 and cyc1--365) encompass the CYC1, OSM1 and RAD7 genes, while none of the deletions extend into the closely linked SUP4 gene.

Thyroxine, triiodothyronine and thyrotrophin levels in meningococcal meningitis, typhoid fever and other febrile conditions.

Thyroid status was estimated serially by measuring triiodothyronine (T3), thyroxine (T4) and thyrotrophin (hTSH) in 20 patients suffering from meningococcal meningitis, typhoid fever and other acute febrile illnesses. Significantly low T3 and only slightly increased T4 were observed in all the patients. hTSH was normal in all of these. A significant reciprocal relationship was found between the degree of fever and fall in T3 concentrations. T3 tended to rise in patients who recovered but in those who deteriorated or died, T3 remained persistently low.

Familial nonobstructive cardiomyopathy with endocardial fibroelastosis beyond infancy.

A 10-year-old boy with congestive heart failure died in five months in spite of comprehensive medical treatment. Autopsy showed patchy areas of endocardial fibroelastosis of the left ventricle. The sister of this patient had followed a similar course at 13 years of age with death within six months of the onset of congestive failure. Her postmortem examination also showed endocardial fibroelastosis. The clinical presentation of familial endocardial fibroelastosis in the preteen and teenage years is a rare event. Probably the endocardial fibroelastosis was secondary to a familial nonobstructive cardiomyopathy.

Authoritarianism and social distance attitudes towards offenders released from prison.

It was hypothesized that authoritarian individuals maintain attitudes of greater social distance towards (a) violent offenders, (b) violent young offenders, (c) non-violent offenders, and (d) non-violent young offenders released from prison. Correlations of scores on Boehm's Legal Attitudes Questionnaire for the offender groups supported the hypothesis. Subjects were 48 students in management science. Apparently high authoritarians would like to maintain attitudes of greater social distance from violent offenders than non-violent offenders released from prison.

Microbodies of the pig liver. A morphologic and morphometric study.

The ultrastructure of hepatic microbodies of pigs was studied in liver samples fixed in phosphate-buffered 2% glutaraldehyde (pH 7.4) and postfixed in 2% osmium tetroxide. The microbodies were rounded or ovoid in shape and contained a granular matrix enclosed with a single limiting membrane. The matrix, in many of the organelles, contained an amorphous or an organized (crystalloid) area. The crystalloids were organized as fine, alternating electron-opaque and electron-lucent lines, or as groups of tubules arranged in more than one plane. Profiles of smooth endoplasmic reticulum were usually found near the microbodies. 34 microbodies were approximately 0.67 +/- 0.02 micrometer (mean +/- standard error) in length and were approximately 0.56 micrometer (+/- 0.02 micrometer) wide. The marginal plates consisted of strata which exhibited a periodicity. The average length and width of 17 plates was approximately 0.34 +/- 0.03 micrometer and 0.002 micrometer, respectively. This study clearly establishes the occurrence of marginal plates in microbodies of porcine liver.

Thyroid hormone levels in Nigerians living in endemic goitrous and non-goitrous areas.

Serum thyroxine (T-4) levels, triiodothyronine (T-3) sponge uptake and free thyroxine index (FTI) were determined in Nigerians living in endemic goitrous and non-goitrous areas. T-4 and FTI values in inhabitants of the non-goitrous area were found to be significantly higher and T-3% sponge uptake values significantly lower, than those from the goitrous area. The range of T-4 levels for normal subjects falls between 4.0 and 13.5 Ug%, that of T-3% uptake between 21.0 and 35.0 and that of FTI values between 1.25 and 4.60.

Nonsense suppressors of yeast cause osmotic-sensitive growth.

Many nonsense suppressors of Saccharomyces cerevisiae cause growth inhibition on hypertonic media. Eight tyrosine-inserting UAA (ochre) suppressors, eight tyrosine-inserting UAG (amber) suppressors, a leucine-inserting UAG suppressor, and a serine-inserting recessive lethal UAG suppressor cause osmotic sensitivity, whereas a serine-inserting UAA suppressor does not cause sensitivity. Although the mechanism is not understood, the growth inhibition of specific suppressors on hypertonic media is correlated with their efficiencies of suppression. This heretofore unknown property of nonsense suppressors is useful for mitotic mapping, selecting tRNA mutants, selecting antisuppressors, and scoring nonsense suppressors.

Enzyme associations in T4 phage DNA precursor synthesis.

A DIRECT APPROACH IS DESCRIBED TO THE QUESTION: Are enzymes of DNA precursor synthesis organized into a supramolecular structure? This approach involved sedimentation analysis of several T4 phage-coded early enzyme activities in crude lysates of infected Escherichia coli. One-third to one-half of several activities tested-dCMP hydroxymethylase, dTMP synthetase, deoxynucleoside 5'-monophosphate kinase, deoxyuridine triphosphatase, and probably dCMP deaminase, but not dihydrofolate reductase or DNA polymerase-sedimented much more rapidly than expected from molecular weight. About 5% of the host cell nucleoside diphosphate kinase, known to participate in T4 DNA precursor synthesis, cosedimented with these activities. To show that this rapidly sedimenting material represents an organized enzyme complex rather than a nonspecific aggregate, we studied the kinetics of formation of dTTP with dUMP as the initial substrate. This three-step reaction sequence reached its maximal rate within a few seconds when catalyzed by enzymes in the aggregate, whereas an equivalent mixture of uncomplexed enzymes required nearly 20 min before dTTP synthesis reached its maximal rate. The effect of aggregation is evidently to decrease the volume into which intermediates are free to diffuse. Because there is reason to believe that intracellular concentration gradients of DNA precursors exist, the properties of this enzyme aggregate in vitro may help to explain how such gradients are maintained.

Short colon malformation with imperforate anus.

Ten cases of a rare high anorectal malformation associated with short colon are presented. The significance of a large gas fluid level (occupying more than half of the abdominal cavity on invertogram) is stressed for pre-operative diagnosis. Terminal colostomy or ileostomy with disconnection of colo-vesical/colo-vaginal fistula has been considered the only adequate therapy. Associated congenital malformations have been responsible for high mortality in these patients.

Comparison of Tc-99m methylene diphosphonate with Tc-99m pyrophosphate in the detection of acute myocardial infarction: concise communication.

Myocardial scans were obtained in ten patients with proven myocardial infarction. Scintigraphy on consecutive days was performed with technetium-99m methylene diphosphonate and technetium-99m pyrophosphate. Images obtained with the two tracers were generally equal in quality. Those with PPi exhibited denser uptake, while background activity was lower with MDP.
