[Cytotoxic activity of large granular lymphocytes (LGL) infiltrated into metastatic tumors of lung cancer].

Large granular lymphocytes (LGL) showing Leu 7 and weak E rosettes were separated from two metastatic tumors of lung cancer. Weak cytotoxic activity of these cells was recognized against autochthonous tumors, but never against K-562 cells. After OK-432 therapy, this cytotoxicity was not enhanced, but antibody dependent cell-mediated cytotoxicity (ADCC) of peripheral blood mononuclear cells of the patient was induced against metastatic tumors. These signs may suggest that specific immunotherapy should be available for these cancers.

[Salivary peptide P-C (III). Its relation to the pathogenesis of diabetes mellitus].

Previous immunohistochemical study showed that salivary peptide P-C like immunoreactivity, originally isolated from whole human saliva, was present not only in human salivary glands but also in human pancreatic B-cells. To elucidate the pathophysiological role of this peptide-like immunoreactivity in human pancreatic B-cells, immunohistochemical study using antisera against both insulin and peptide P-C was carried out on the paraffin embedded pancreatic tissues of 27 diabetic patients and 30 control subjects. Positive immunofluorescence due to insulin was detected in 96% of the diabetic pancreases and 100% of the controls. A pancreas of only one IDDM did not have any immunofluorescence due to insulin. Thus, no significant differences were seen in connection with the presence of insulin between pancreases of NIDDM and the controls, though the present study did not examine whether the number of pancreatic B-cells in the pancreas of NIDDM was the same as that in the controls or not. In contrast, positive immunofluorescence due to peptide P-C like immunoreactivity was demonstrated in only 41% of the diabetic pancreases but in 87% of the controls. In view of the fact that the diabetic pancreases and the controls were fixed and embedded into paraffin in the same department of pathology and under the same conditions, the negative finding for salivary peptide P-C like immunoreactive in the diabetic pancreas seems to result neither from the effects of fixatives nor the destructive postmortem effects of enzymes on the antigenicity of peptide P-C like immunoreactivity but from the decreased content of peptide P-C like immunoreactivity in the diabetic pancreas.(ABSTRACT TRUNCATED AT 250 WORDS)

[The presence of proline-rich peptide P-C like immunoreactivity in the human pancreatic B-cells].

It is well known that there are functional and morphological similarities between the salivary glands and the pancreas. Amylase, kallikrein and glucagon are present in both tissues. Morphological similarities of the two tissues have been observed by using a light and electron microscope. In order to examine the pathophysiological relationship between the pancreas and the salivary glands, an immunohistochemical study using antisera against proline-rich peptide P-C, which was recently isolated from human whole saliva, was carried out on the human salivary glands and the pancreas. Peptide P-C like immunoreactivity was found not only in the salivary glands but also in the pancreatic islets. Furthermore, observation of serial thin sections immunostained with insulin, glucagon, somatostatin, PP antisera and antisera against peptide P-C revealed that peptide P-C like immunoreactivity-containing cells were identical to insulin containing B-cells. As the antisera against peptide P-C did not have any cross-reactivity to other kinds of peptide including insulin, glucagon, somatostatin, pp, VIP, human C-peptide and kallikrein, the present finding suggests that peptide P-C like immunoreactivity is present in the B-cells independently of insulin and proinsulin. The finding seems to be a new addition to the lists of proof which support the presence of a pathophysiological relation between the salivary glands and the pancreas. Although it seems likely that peptide P-C like immunoreactivity in the pancreatic B-cells may play some role in the function of the B-cells, since this material was present only in the B-cells among four kinds of cells in the pancreatic islets, its exact pathophysiological role remains to be elucidated.

Evidence for human T cell lymphoma-leukemia virus infection of family members of human T cell lymphoma-leukemia virus positive T cell leukemia-lymphoma patients.

Sera of family members of patients from the United States, the Caribbean, and Japan, with human T cell lymphoma-leukemia virus (HTLV) associated T cell malignancies, possess HTLV-specific antibodies directed against internal structural components of HTLV, p24 and p19. The prevalence of antibodies to HTLV is greater in family members than in random healthy donors, which supports the infectious nature of HTLV and its association with particular aggressive T cell malignancies. Expression of HTLV p24 and p19 has also been observed in cultured T cells of some healthy relatives, and intact virus particles have been released from cells of one possibly pre-leukemic family member.

A possible autonomic modulation of sinus node recovery time in overdrive suppression test.

The effect of blood pressure fall on sinus node recovery time (SNRT) was studied in 17 patients during the overdrive suppression test. Their maximal SNRT was less than 150% of the mean P-P interval of the control state (=SCL) and CSNRT (=SNRT-SCL) was less than 550msec. Blood pressure was found to fall in about a half of the patients at a high pacing rate. Concomitantly, SNRT was found to shorten in those who showed such a fall in blood pressure. The subjects were then divided into two groups; one with a shorter SNRT at 150bpm than at 90bpm of pacing rate and the other without such shortening. The changes in SNRT and CSNRT were both significant in the first group (p less than 0.02 and p less than 0.001, respectively). The fall in blood pressure at 150 bpm was significant in the group with shortened SNRT and CSNRT and it was larger than in the other group: 23 +/- 14mmHg vs. 1 +/- 5mmHg (p less than 0.001). This result suggests a possible modulation of SNRT by a fall of blood pressure via baroreflex.

High incidence of human type-C retrovirus (HTLV) in family members of a HTLV-positive Japanese T-cell leukemia patient.

Sera and peripheral blood cells of an adult T-cell leukemia patient and several clinically normal members of his family from the northwest coast of Japan were examined for evidence of infection with human T-cell leukemia (lymphoma) virus (HTLV). The sera of the patient and his parents had antibodies to HTLV, whereas these antibodies were absent in the sera of the patient's brother and sister. T-cell lines were established from the peripheral blood lymphocytes of all of the family members, and all except the patient's sister expressed HTLV antigens (p19, p24, and reverse transcriptase) and type-C virus particles. Not only the fresh peripheral blood lymphocytes from the patient but also those from his clinically normal mother showed abnormal morphology of the kind characteristic of some patients with T-cell leukemia. These studies are consistent with previous evidence indicative of a high rate of HTLV infection within families, and they show that people whose sera are negative for antibodies may still be infected by HTLV. In addition, the results indicate that infection of T cells by HTLV can be associated with morphological transformation of the cells without other signs of leukemia.

Zearalenone reductase from rat liver.

Zearalenone is known to be reduced by rat liver preparations to a more active estrogenic metabolite, alpha-zearalenol. To elucidate the enzymatic feature of zearalenone reductase, we worked out a simple assay procedure for this enzyme using [3H]-zearalenone and thin layer chromatography (TLC). The NAD(P)H-dependent zearalenone reductase localized in the microsomes was most active at pH 4-4.5, while the reductase localized in the cytosol was active at neutral pH. The former enzyme reduced zearalenone only to alpha-zearalenol and the latter reduced both alpha-zearalenol and beta-zearalenol. The microsomal enzyme was solubilized with Triton X-100 and purified by DEAE-Sephadex, hydroxyapatite and Sepharose 4B column chromatographies. The partially purified enzyme showed the following properties. a) The molecular weight of the enzyme was estimated to be about 230,000 by Sepharose 4B chromatography. b) The apparent Km value of the enzyme was 1.0 X 10(-5) M for zearalenone at an NADPH concentration of 1 mM. c) The enzyme activity was inhibited by a high concentration of KCl but not by 1 mM Co2+, Mn2+, Zn2+, EDTA, or 20 mM N-ethylmaleimide.
