Long-term results with autogenous microvascular grafts in various experimental models in rats.

Autogenous arterial and venous grafts were used to bridge defects in the femoral arteries, aortas, and renal arteries of 40 Wistar rats in an effort to determine the feasibility of using such grafts in the treatment of renovascular hypertension. The long-term results indicated that arterial grafts would be the most suitable in such a situation, but further study is needed before the technique can be applied clinically.

[Further investigations on the protective effect of cadmium acetate in bromotrichloromethane poisoning].

The rats were pretreated with cadmium acetate (20 mg/Kg.ip) and subsequently challenged with a dose of bromotrichloromethane (2,6 mmoles/Kg.po), the haloalkane produced no change on liver polyribosomes, as evidenced by disaggregation of sedimentation profile. Cadmium acetate failed to protect against CBrCl3-induced lipid peroxidation, as measured by malonaldehyde production of liver homogenate.

Early investigations on the effect of methyl mercuric chloride upon DMN-acute hepatotoxicity.

Acute toxicity induced by DMN was partially prevented by previously administering methyl mercuric chloride (MMC), a chemical inhibitor of the drug metabolizing enzyme system (DMES). We have studied the early changes occurring during the course of DMN-intoxication, namely disaggregation of polysomal profiles and necrosis, evaluated morphologically and by the release of S-GPT.

Protection by cerium chloride on CCl4-induced hepatotoxicity.

Pretreatment of rats with cerium chloride, an inhibitor of the DMES, protect against CCl4 intoxication. The protection was obtained against the fatty infiltration in the liver, the decrease of triglyceride secretion from the liver into the plasma compartment, the polysomal disaggregation and the rise of serum transaminases.

Simultaneous detection of reverse transcriptase and high molecular weight RNA in tissue of patients with Hodgkin's disease and patients with leukemia.

Complexes of high-molecular-weight RNA and reverse transcriptase (RNA-dependent DNA nucleotidyltransferase) have been detected in 14(77.8%) of 18 spleen from patients with Hodgkin's disease and in all samples tested of peripheral leukocytes and spleens from leukemic patients. The enzyme and its template are localized in a particle having a density between 1.16 and 1.19 g/ml. These observations describe characteristic features of RNA tumor viruses.

Cryopreserved human hepatocytes from cell bank: in vitro function and clinical application.

UNLABELLED: We Aimed to analyze the in vitro function of isolated and cryopreserved human hepatocytes (CHH) from a cell bank and to define their potential clinical application in a bioartificial liver (BAL) device. METHODS: Over 24 months, 103 not transplantable livers were utilized for human hepatocytes isolation and cryopreservation. Hepatocytes isolated by collagenase were analyzed for yield, viability, diazepam metabolism, and production of human albumin after isolation and cryopreservation in LN(2). RESULTS: The causes for refusal for transplantation were macrosteatosis >60%, ischemic damage due to donor hypotension, and nonviral cirrhosis in 60%, 11%, and 8%, respectively. Cell yields averaged 7 million hepatocytes per gram of liver of mean viability of 80% +/- 13%. The viability of CHH after thawing averaged 50%. Thawed hepatocytes showed diazepam metabolism, and human albumin synthesis comparable to fresh cells. CHH were utilized as the biological component of a BAL for temporary support as three applications of two patients affected by fulminant hepatic failure awaiting urgent transplant. Ten to 13 billion viable CHH were loaded into each BAL. Liver function showed bilirubin and ammonia reduction at the end of each treatment. One patient was successfully bridged to emergency OLTx after one BAL; in the second case there was spontaneous recovery of liver function after two BAL. CONCLUSIONS: Recovery of donor human livers unwanted for transplantation allowed isolation and cryopreservation of viable and functionally active human hepatocytes, which have been banked and successfully used for clinical applications of a BAL device.

Inhibition of late influenza virus genome expression by diamidinophenylindole.

The growth cycle of influenza virus strain FPV, Ulster 73, was altered by treatment of LLC-MK2 cells with diamidinophenylindole. Viral protein synthesis was restricted to the early pattern of virus multiplication, and post-treatment experiments showed the ability of the drug to block virus replication until the 4th hour p.i. Drug addition (followed by removal) revealed the inhibition of synthesis of late viral products, and especially of membrane protein. Kinetic studies on the production of viral RNA indicated a decrease in the synthesis of late virus-induced RNA species, suggesting that the target of DAPI is probably the late transcription of the virus genome. The nonpermissive condition mediated by the drug could represent a suitable model to study cellular intervention during viral growth.
