Adrenergic stimulation of adrenocortical secretion in immature fowl.

In 5-6-week-old cockerels the circulating corticosterone concentration was significantly increased in birds i.m. injected 30 and 60 min previously with adrenaline (0.33 mg/kg), noradrenaline (0.33 mg/kg) or a beta-adrenergic agonist (isoprotorenol, 1 mg/kg), but was reduced in birds pretreated with an alpha-adrenergic agonist (phenylephrine, 1 mg/kg). The stimulation of corticosterone secretion induced by a 30 min period of forced exercise (0.04 km/hr; 0 degree incline) was potentiated by noradrenaline, isoprotorenol and phentolamine pretreatment. In response to exogenous adrenocorticotrophin (ACTH, 8 i.u./kg), administered i.v., the increase in the plasma corticosterone concentration was elevated above that in the controls in birds pretreated with adrenaline, noradrenaline, isoprotorenol or phentolamine (an alpha antagonist administered at 1 mg/kg). The corticosterone response to ACTH was suppressed by phenylephrine pretreatment. These results demonstrate that both basal and stimulated levels of adrenocortical activity may be subtly regulated by adrenergic mechanisms acting at a site(s) within the hypothalamo-pituitary-adrenal axis.

Deoxyribonucleic acid polymorphism in the apolipoprotein A-1-C-III gene cluster. Association with hypertriglyceridemia.

A DNA sequence polymorphism, revealed by digestion of human DNA with the restriction endonuclease Sst-1 and hybridization with an apolipoprotein A-I complementary DNA clone, has been shown to be located in or close to the 3' noncoding region of the apolipoprotein C-III gene. This polymorphism is found in significantly increased prevalence (P less than 0.001) in Caucasian hypertriglyceridemic subjects compared with race-matched controls, and its distribution in normal individuals of differing racial origins is reported. Furthermore, no alteration of high density lipoprotein or apolipoprotein A-I and apolipoprotein C-III phenotypes was observed in individuals with or without the polymorphism.

Stress and adrenal function.

The natural environment is composed of various potentially hostile stressors. It is a basic requirement of life that the cells of an organism must be maintained within closely defined physiological limits. The maintenance of a constant interior mileu results from physiological and behavioural homeostatic adaptations. The physiological regulation of homeostatis is achieved by complex endocrine interactions, principally by the hormones secreted from the adrenal glands. In this brief review the responses of the avian adrenal glands to stressful stimuli, the mechanism of adrenal activation, and the function of the adrenal responses will be considered.

Insulin and apolipoprotein A-1/C-III gene polymorphisms relating to hypertriglyceridaemia and diabetes mellitus.

Two gene specific probes have been used to identify polymorphic DNA loci on chromosome 11 close to the insulin and apoprotein A-1 genes in a genetic analysis of hypertriglyceridaemic patients with and without co-existing diabetes. Of the 45 patients studied with both probes, 15 were diabetic of whom nine possessed class 3/3 insulin polymorphism genotypes, compared with none in the non-diabetic group (p less than 0.001; chi 2 test). In contrast, an uncommon apolipoprotein A-1 polymorphism was found to be equally distributed in the diabetic and the non-diabetic patients. No co-segregation of these two particular genetic polymorphisms was found in either patient group. The differing associations of the two disease-related polymorphism genotypes in patients with hypertriglyceridaemia with or without co-existing diabetes may possibly reflect differing aetiologies of the hyperlipidaemia.

Adrenocortical and adrenomedullary responses of fowl to treadmill exercise.

Adrenomedullary and adrenocortical responses of 40-day-old cockerels to treadmill exercise (0.4 km/hr, 0 degrees incline) were determined. Plasma concentrations of adrenaline were increased above both resting and control levels (P less than 0.001) after 30 min exercise and continued to increase (P less than 0.01) until the cessation of exercise. Plasma noradrenaline and dopamine levels were increased after 60 min of exercise (P less than 0.01 and 0.05, respectively). The adrenaline component of the plasma catecholamine response increased significantly above that of noradrenaline (P less than 0.001). Plasma corticosterone levels were also increased (P less than 0.001) during exercise and were closely correlated with plasma adrenaline concentrations. Exercise depleted (P less than 0.01) adrenal stores of adrenaline, which were inversely proportional to plasma adrenaline concentrations (P less than 0.001). Neither adrenal noradrenaline nor dopamine were significantly correlated with their plasma levels. These results suggest that adrenocortical (corticosterone) and adrenomedullary (adrenaline) responses during exercise may occur in response to similar stimuli or may be interrelated.

The prognostic value of intraoperative pressure gradients with congenital aortic stenosis.

Patients with congenital aortic stenosis are subject to sudden death without prior symptoms. Indications for operation are based on pressure gradients, but intraoperative evaluation of results after relief of obstruction can be subjective and arbitrary. Between September, 1981, and October, 1983, 21 patients underwent operation for relief of congenital aortic stenosis. Fifteen had valvular, 4 had supravalvular, and 2 had subvalvular stenosis. Seven patients were 1 year of age or younger, 2 being 3 days old. Preoperative inotropic support and emergency operation were necessary in 2 patients. Intraoperative pressure gradients were measured in all but the 2 patients who were 3 days old. All patients survived without complications. Intraoperative and postoperative pressure gradients were available in 16 patients. The average preoperative pressure gradient was 76 mm Hg. It was significantly lowered to 20 mm Hg (p less than 0.001) intraoperatively, which compared with 15 mm Hg postoperatively. Three patients had an unsatisfactorily high intraoperative pressure gradient after initial intervention. Immediate reexploration and incision of the rudimentary commissure in 2 and excision of redundant valvular tissue in the other resulted in a more satisfactory pressure gradient without substantial aortic insufficiency. We suggest that measurement of intraoperative pressure gradients can accurately assess the results of therapy and allow the surgeon the option of reexploration for further intervention. Moreover, the high correlation between intraoperative and postoperative pressure gradients can help in planning postoperative management and future cardiac catheterizations.

DNA polymorphism adjacent to human apoprotein A-1 gene: relation to hypertriglyceridaemia.

Polymorphism in a DNA sequence has been observed on the 3'-flanking region of the human apoprotein-A-1 gene. The frequency of the heterozygous state in a healthy control population (n = 73) is around 0.05. However, 12 (frequency 0.34) out of 35 subjects with hypertriglyceridaemia were found to have the polymorphic site, and 2 were homozygous for this variant. The mutant allele may constitute a linkage marker for some abnormality within the apoprotein-A-1 gene, affecting either expression of or some minor structural modification of the A-1 apoprotein, that may predispose to hypertriglyceridaemia.

Habituation of the corticosterone response of ducks (Anas platyrhynchos) to daily treadmill exercise.

The habituation of the corticosterone response of ducks to treadmill exercise (1.1 km/hr 0 degrees incline for 60 min) has been determined over a 28-day period. In untrained birds 30 min of treadmill exercise resulted in a marked (P less than 0.001) increase in the basal plasma corticosterone level. After 60 min of exercise the corticosterone concentration remained elevated (P less than 0.001) in comparison with basal and control (nonexercised) levels but was less (P less than 0.01) than that at 30 min. Following exercise the concentration declined to basal levels within 60 min. After 7 days of daily training the peak (30 min) corticosterone response to exercise diminished by greater than 50% and was absent in birds trained for 21 days. This habituation of the corticosterone response may be due to adrenocortical stimulation by pituitary adrenocorticotrophin (ACTH) being reduced. As the stimulatory effect of ACTH administration (8 IU/kg) on corticosterone secretion does not habituate in birds treated daily (for 28 days), the reduced corticosterone response to exercise may be due to decreased ACTH release rather than to a reduction in adrenal ACTH sensitivity.

Responses of neurons in the inferior colliculus of the rat to AM and FM tones.

The responses of units in the inferior colliculus of the urethane-anaesthetized rat were recorded extracellularly. They responded to sinusoidal AM and FM tones with a modulation of their spike discharge usually at the same, or occasionally at twice, the modulation rate of the stimulus. The modulation depth of the response initially increased with the modulation depth of the stimulus, but usually saturated or decreased at higher stimulus depths. The units showed a bandpass tuning to stimulus modulation rate which was independent of modulation depth and, in all cases, the most effective modulation rate was below 120 Hz. The modulated response to temporally varying stimuli could not be predicted from the pure tone discharge patterns or, in some cases, the unit's mean firing rate to modulated tones; temporally varying stimuli gave temporally varying responses. When compared with the data available from units at other levels in the auditory system, the results indicate a trend in which units at successively higher levels in the pathway respond most effectively to progressively lower rates of modulation.

A DNA polymorphism adjacent to the human apolipoprotein CII gene.

We have used a cDNA clone for human apolipoprotein CII (apo CII) to detect a common DNA polymorphism with the enzyme TaqI. This polymorphism is probably caused by a single base change approximately 2000 base-pairs from the 3' end of the structural gene. In the normal population (n = 90) the frequency of the less common allele is approximately 0.44. No significant differences were observed in the allele frequency in individuals with type IIa, IIb, III, IV and V lipoprotein patterns. There does not seem to be any population association between the TaqI polymorphism and factors that predispose an individual to hyperlipidaemia.
