Biochemical and morphological studies on the effects of anthocyans and vitamin E on carbon tetrachloride induced liver injury.

The effects of the natural antioxidants-anthocyans and vitamin E (in a solubilized pharmaceutical form) on carbon tetrachloride-induced liver injury in rats are studied. The changes in the activity of serum transaminases (ALAT and ASAT), the content of the reduced glutathione and cytochrome P-450 as well as the intensity of the processes of lipid peroxidation are assessed. The anthocyans exert a protective effect comparable to that of vitamin E on liver cells. The favorable effects of the combination of the antioxidants on the content of the reduced glutathione and on the processes of lipid peroxidation are more intensely expressed. The morphological changes occurring in hepatocytes correlate with the results of the biochemical studies. It is evident that both substances have a marked hepatoprotective activity.

[A case of carcinoid tumor of the breast].

A rare tumor of the mammary gland, built up of argyrophilic cells, with numerous metastases in the axillary lymph nodes is reported. Histochemical and electron-microscopic studied unequivocally demonstrated that this tumor belonged to the APUD system. The possible histogenesis of carcinoid tumor of the mammary gland is briefly discussed.

Expression of ABO blood-group antigenic determinants identified by monoclonal antibodies B7H4 and D1E8 in normal human tissues and organs.

Human blood group antigens are genetically determined structures of undefined nature found not only in red blood cells but also in various tissues. Monoclonal antibodies against A and B human blood group antigens (B7H4 and D1E8) were obtained. Their tissue compatibility was tested in 11 different normal human organs taken from people with A1 and B blood group phenotypes. The antigen-antibody reaction was visualized using the indirect immunoperoxidase technique. The results of the study showed that the antibodies B7H4 and D1E8 react specifically with the tissue blood group antigens in healthy subjects. The findings obtained in most of the tested organs were in accord with those described by other researchers. Differences were found only in the stomach, urothelium, the lungs and the thyroid gland. A possible explanation of the results is the difference in the epitopes recognized by the monoclonal antibodies used by us and the blood group antibodies applied by other researchers. It is suggested that the monoclonal antibodies against the human blood group antigens A and B obtained by us could be used to establish suspected modification of ABH-antigens in the process of oncogenesis.

[Immunohistochemical evidence of vimentin in fibrocystic breast disease and mammary carcinoma].

Immunohistochemical investigations were conducted of cryostat sections taken from 25 cases of fibrocystic breast disease and 100 cases of mammary carcinoma (among them 15 cases with lymph node metastases) to obtain information on expression of vimentin. Immunoreactive epithelial cells were recordable from 12 cases of fibrocystic disease (48%) and 32 tumors (32%). In the majority of all cases with vimentin-expressing carcinoma, positive staining was exhibited by less than 10% of the neoplastic cells. No correlation was found to exist between histological tumor grade, lymph node status and vimentin expression.

Carcinosarcoma of the breast. Immunohistochemical and ultrastructural studies.

A case of a carcinosarcoma in the breast of a 51-year-old woman is described. The tumor consisted of two diversely differentiated components with cells possessing epithelial and mesenchymal characteristics. Plenty of cytokeratin and CEA were immunohistochemically recordable from the carcinomatous component. Some neoplastic cells also stained positively for actin, S-100 protein and smooth muscle antigen. The sarcomatous component was positive only for vimentin. Electron microscopic analysis revealed presence of the following cell types: epithelial cells, myoepithelial cells and polymorphous mesenchymal cells.

Morphological characteristics of the endocrine pancreas in alloxan diabetes after cyclosporin A administration.

Histological, immunocytochemical, morphometric and electron microscope studies were carried out on the pancreas of alloxan diabetic rats pretreated with cyclosporin A. High mortality, severe destruction of pancreatic B-cells and presence of sporadic mononuclear infiltrations in islets and around excretory ducts were observed. The results obtained show that cyclosporin A potentiates the toxic effect of alloxan on the pancreatic B-cells.

[Lymphoid cell infiltration into the pancreas of C57BL/6J mice].

Peri-insular lymphoid infiltrations were observed in nondiabetic C57BL/6J mice. The type of infiltrating cells was determined by direct immunoperoxidase technique. A larger part of lymphoid cells did not show reaction with a goat antimouse IgG Rh conjugate. Intensive hyperplastic reaction and the presence of many immunoglobulin producing cells were observed in the lymph nodes adjoining the pancreas. Probably the observed infiltrations in the pancreas of nondiabetic C57BL + 6J mice were composed mainly of T lymphocytes, belonging to T helper subpopulation.

Nephrotoxic effect of the specific brain serotonin depletor para-chlorophenylalanine.

Brain serotonin depletion induced by peripheral parachlorophenylalanine (pCPA) is frequently used to evaluate the role of the central serotoninergic system in the regulation of a number of physiological functions, including the secretion of renin by the kidney. We found that due to the treatments applied in the protocol used for the investigation of pCPA effect on renin and vasopressin secretion in rats (300 mg/kg i.p. 64 and 40 h before sacrifice) renal injury was induced as well. Typical changes indicating acute renal failure were observed--an initial polyuria, natriuresis and body mass loss, succeeded by oliguria, decreased glomerular filtration rate, and salt and creatinine retention. Morphological changes in the glomeruli included a thickening of the basal membranes, a confluence and a reduced number of podocyte pedicles. A slight to moderate granular degeneration was observed in epithelial cells of the proximal convoluted tubule, combined with mitochondrial changes--an increase in number, matrix disorganization, and myelin degeneration. In conclusion, the renal function changes after i.p. pCPA may be due not to brain serotonin depletion-alone, but also to nephrotoxic effect.

[Immunomorphologic study of various endocrine cells in chronic gastritis and in intestinal metaplasia of the stomach].

The amount of some endocrine cells in various forms of chronic gastritis with and without the presence of intestinal metaplasia of the stomach mucosa was studied immunohistochemically (PAP-reaction). The study was carried out on 18 samples of biopsied material for D-cells and on 31--for G cells. It was established that the amount of these cells as well as their ratio was changed in accordance with the form of chronic gastritis. Thus, for example++ , the number of D-cells was increased in mild forms of gastritis, but they were reduced in its grave form, while the number of G-cells gradually diminished from mild to grave forms. These changes give foundation to assume that hr hormonal disturbance of the stomach mucosa plays important role in pathogenesis of this disease. This suggestion is supported by the observed in dynamics quantitative changes of these cells, beginning even at the initial stages of this disease.

[Localization of receptors for phytospermoagglutinin from Arum maculatum on human spermatogenic cells and spermatozoa].

Our precious studies showed that delipidized water-saline extract from roots of Arum maculatum (AM) contained lectin-phytospermoagglutinins (PSA), which caused agglutination, manifested by the type of a tail with a tail and a head with a tail. This extract we conjugated with fluorescein-isothiocyanate and by means of the conjugate we studied the localization of receptor molecules for PSA on the plasmalemma of human spermatogenic cells and human spermatozoa, obtained from the testis, epididymis and ejaculate. It was established that receptor molecules for PSA were distributed irregularly on the plasmalemma of spermatozoa, that the basic part of these molecules were synthesized on spermatogenic cells and that ejaculate spermatozoa adsorbed such molecules from the sperm plasma.

[Lipopolysaccharide-containing cytoplasmic membranes as immunostimulants of peritoneal macrophages].

It was established that cytoplasmic membranes from the stable L-forms of E. coli WF+ induced a 4-5 fold increase of the number of peritoneal exudative cells in mice after single i.p. inoculation. The animals treated with membranes reacted with 4-5 fold higher number of these cells after an i.p. infection by parent form cells or L-form cells, as compared with the reaction of infected, nontreated by membranes mice. The macrophage bactericidal activity was 6-10 fold increased. Using electron microscopy, it was established that the phagosomes containing membranes and their remnants were localized in the peripheral part of the cytoplasm or near the nucleus, without neighbouring lysosomes during the 24-hour interaction. Single cases of phagolysosome fusion were observed.

[Experimental liver damage by furosemide for studying drugs with hepato-protective activity].

Some biochemical, pathohistological and ultrastructural changes in liver after the action of single intraperitoneal administration of furosemide (Furantryl-Pharmachim) in a dose of 300 mg/kg of body mass were studied in male white rats of Wistar strain. The results from the performed experiments established increased activity of serum enzymes GOT, GPT, GGT, alkaline phosphatase as well as low values of serum cholinesterase. Pathohistologic and electron microscopy examination discovered liver damage with typical congestive changes mainly--manifested local erythremia and a reduced fluid content of the blood in the liver with blockage in the sinusoidal pole of hepatocytes; there were also focal micronecrosis, considerable reduction of glycogen and slight centrolobular steatosis. The possibility is discussed for usage of hepatotoxicity, induced by furosemide, in examining the effects of some drugs with potential hepatoprotective activity.
