Specificity of Shore's method for the histamine determination was investigated by examining the fluorescence spectrum of the extract from rat gastric mucosa, biopsy specimen of human gastric mucosa, human gastric juice and human whole blood. An almost identical spectrum to that of pure histamine was obtained with the extract from rat gastric mucosa and human whole blood, whereas the spectrum with the extract of human gastric biopsy specimen and human gastric juice was different from that of histamine. By means of high-performance liquid chromatography, an approximately three times more concentrated extract from human gastric mucosa gave a distinct peak of histamine while a hundred times concentrated gastric juice gave no measurable peak of histamine. Fluorescence spectrum of the substances which might be extracted with histamine was examined. The ratio of the fluorescence of histidine to that of equimolar concentration of histamine at the wave length for histamine determination was 4.62%. From the above results, it has been concluded that Shore's method is useful for the determination of histamine contents in the gastric mucosa and whole blood if approximately 5% contamination with histidine is taken into consideration.
ACTH stimulation tests were performed in 15 premature infants in a serial fashion at the ages of 5--10 days and 27--31 days. Six of the 15 had subsequent ACTH stimulation tests at 6--8 weeks and/or 12--13 weeks. The pre- and post-ACTH serum cortisol and dehydroepiandrosterone sulfate (DHAS) levels were determined by radioimmunoassay. The mean basal levels of cortisol, 55 ng/ml, and DHAS, 4108 ng/ml, were significantly higher (P less than 0.05 and P less than 0.025, respectively) at 5--10 days than those of 24 ng/ml and 1858 ng/ml, respectively, at 27--31 days. The mean net change (delta) of cortisol after ACTH at 5--10 days, 95 ng/ml, increased significantly to 148 ng/ml at 27--31 days. However, deltaDHAS did not differ significantly between the two periods (1514 ng/ml vs. 972 ng/ml). Therefore, deltaDHAS/deltacortisol was lower (P less than 0.05) at 27--31 days than at 5--10 days. No further significant changes were observed after 4 weeks of age in the levels of the two steroids. There was little correlation of basal levels between cortisol and DHAS, nor between deltacortisol and deltaDHAS at any age period we studied. There were four infants whose mothers had prenatal steroid treatment for the prevention of hyaline membrane disease and their values were not different from the other infants.

Thiobacillus thiooxidans was acidostable even in the absence of its respiratory substrate, elementary sulfur. This suggests that the acidostability of the bacterium was enery-independent. The organism was subjected to osmotic shock with 0.75 M sucrose at 0 degrees C and then treated with snail intestinal juice in the presence of 0.3 M sucrose. The decrease in the optical density of the sample thus prepared on dilution with deionized water and electron microscopic observation of the sample showed that spheroplasts were formed from the bacterium by this procedure. Spheroplasts were able to respire sulfur and their respiratory activity was acidostable. Spheroplasts, when treated with Nagase, proteolytic enzyme, lost their acidostability, and some protein components disappeared from the membrane fraction. This suggests that the acidostability of the bacterium may be related to protein conponents of the membrane.
A sulfate-reducing bacterium, a strain of Desulfovibrio vulgaris, when lactate or pyruvate was supplied as an electron and energy source, reduced acetylene only in the presence of sulfate. Acetylene reduction started after a lag of 1 h. H2 which acts as an electron and energy source for sulfate reduction was unable to reduce acetylene even in the presence of sulfate. It was suggested, from the measurements of the amounts of adenine nucleotides of the bacterium incubated under various conditions, that the bacterium is able to reduce acetylene at a high ATP level or a high ATP/ADP ratio.
Myocardial contractility was estimated in 27 patients with hyperthyroidism (10 men and 17 women, aged from 16 to 38) by echocardiography. Echocardiographic examination was performed before and on the 7th day after the subtotal thyroidectomy. Thyroid functions (PBI, T3, and T4) decreased significantly after the operation (p less than 0.01). Heart rate and systolic blood pressure showed a slight decrease (N.S.), while diastolic blood pressure showed a slight increase (N.S.), while diastolic blood pressure showed a slight increase (N.S.). Mean AWVs decreased from 35.8 +/- 7.0 mm/sec to 27.6 +/- 5.7 mm/sec and correlated wtih T3 (r=0.4008, p less than 0.05). Mean PWV's also decreased from 53.2 +/- 12.2 mm/sec to 45.2 +/- 11.4 mm/aec and correlated with T3 (r=0.5237, p less than 0.02). DDR decreased from 108.0 +/- 36.8 mm/sec to 85.7 +/- 31.7 mm/sec and correlated wtih T3 (r=0.4008, p less than 0.05). Above results suggest that enhanced myocardial contractility and elevated rate of ventricular filling in hyperthyroidism are suppressed after the operation, and that echocardiography is useful for the evaluation of the clinical course after the operation.
