Inversion of chromosome 3 in a case of chronic myelogenous leukemia with abnormal thrombopoiesis.

A patient with chronic myelogenous leukemia (CML) associated with a remarkable increase of micromegakaryocytes in bone marrow was revealed to have an abnormality of a long arm of chromosome #3, i.e., inv(3)(q21q26), in addition to a complex Ph translocation: t(9;22;15)(q34;q11;q22). Although several cases of acute leukemia with inv(3)(q21q26) and abnormal megakaryocytes have been reported, this is the first case in which the association of inv(3)(q21q26) with a megakaryocytic abnormality was observed in a patient with CML. Our findings suggest that this structural rearrangement may be more specifically associated with abnormal thrombopoiesis than are other structural anomalies of 3q.

Sex difference of pinnal hyperemia in magnesium-deficient rats: effects of castration and administration of sex hormone.

In the course of studies on the effects of magnesium (Mg) depletion in the diet (0.001% Mg) on mature rats (10 week-old), it became apparent that females developed less hyperemia than males. In the females, the time of onset and severity of hyperemia were enhanced by castration, but not in the males. These data suggest that the development of hyperemia in Mg-deficient adult rats depends on the female sex hormone. The effects of administration of estradiol were examined, using immature rats (3 week-old), in relation to hyperemia and histamine metabolism. Two to 3 days after feeding young male and female rats a Mg-deficient diet, the pinnal hyperemia appeared in the same degree. The hyperemia was reduced by the administration of estradiol benzoate (0.04-0.08 mg/100 g body wt. per day, s.c.) every day. The increments in urinary histamine excretion, splenic histamine content and histidine decarboxylase activity during Mg-deficiency were markedly reduced by the administration of estradiol. The sex difference in the hyperemia which appeared upon Mg-depletion depends on the effects of the female hormone, estradiol, on histamine metabolism.

Inhibition of DNA synthesis by chromium compounds.

Cr(VI) irreversibly inhibited DNA synthesis in cultured mouse L cells to 50% of controls at 10 microM; 3.3 mM Cr(III) did not. At 0.3 mM, Cr(III) and Cr(VI) inhibited DNA synthesis in permeabilized L cells to 50% of control values. Cr(III) was a stronger inhibitor of DNA synthesis in the DNA-Escherichia coli DNA polymerase I system than was Cr(VI). The inhibitory effect of Cr(VI) depended on the ratio of Cr/DNA and Cr/enzyme; on the other hand, the increase in the concentration of DNA polymerase did not affect the inhibition of Cr(III), Cr(III), below the inhibitory concentration, produced an increase in the incorporation of [3H]dTMP into DNA; this was not observed with Cr(VI).

[The expression of oncogene fms in human chorionic tissue].

The roles of oncogenes have been a subject of great interest not only concerning oncogenesis but also recently concerning physiological regulation in cell proliferation and cell differentiation. We here report the expression of oncogene fms in human chorionic tissue using a molecular hybridization method. RNAs extracted from human chorionic tissue of the first and third trimester were electrophoresed in horizontal agarose gel, blotted according to Northern's method and were hybridized to 32P labeled fms-cDNA (complementary DNA). Four kb RNA molecules homologous to fms-cDNA were detected in chorionic RNAs of the first and third trimester placental tissue with an increase toward term. We then observed the distribution of fms-mRNA in chorionic tissue with in situ hybridization. The signals for fms were observed over trophoblasts without specific localization to the proliferative cells. These results revealed that oncogene fms is expressed in human chorionic tissue of both the first and third trimester. Furthermore the results suggest that the role of fms c-onc may be related to cell differentiation and not to cell proliferation.

[Complete remission obtained in refractory acute lymphocytic leukemia using high-dose cytosine arabinoside combined with low-dose L-asparaginase].

A 41-year-old male was diagnosed as acute lymphocytic leukemia (ALL) in November, 1982 and partial remission was obtained by a combination chemotherapy of LVP, DVP ABOP and VAMP. In January, 1983, peripheral blood showed an increasing number of leukemic cells and he was readmitted to our hospital. WBC count in the peripheral blood was 13,200/mm3 and an 82% ratio of leukemic cells was observed. Bone marrow aspiration showed a hypercellularity of 89.4% leukemic cells. High-dose Ara-C therapy was started at a dose of 3 g/m2 i.v. every 12 hours for 6 days. Leukemic cells in peripheral blood were rapidly decreased in number, and the nucleated cell count of bone marrow was also reduced after 3 weeks of treatment, however 95% of leukemic cells remained. Low-dose L-asparaginase was then supplemented at a dose of 2000 U for 3 days, and 2 months later complete remission was achieved. The side effects associated with this high-dose Ara-C therapy were nausea, vomiting, diarrhea, fever and conjunctivitis, although these were tolerable. These observations suggest that high-dose Ara-C combined with L-asparaginase should be added to the treatment of leukemia which is refractory to conventional chemotherapy.

[Sequential combination of high-dose cytosine arabinoside and L-asparaginase in the treatment of refractory acute leukemia and malignant lymphoma].

Clinical effects of sequential administration of high-dose cytosine arabinoside with L-asparaginase were studied in 5 cases of refractory acute leukemia and 2 cases of non-Hodgkin's lymphoma. A total 12 courses were carried out on these 7 patients and complete remission was obtained in 2 courses and partial remission in 3 courses. Two cases of lymphoma with pleural effusion or CNS invasion achieved partial and complete remission, respectively. The side effects associated with this sequential therapy were nausea, vomiting, diarrhea, fever and conjunctivitis, although these were tolerable. These observations suggest that high-dose cytosine arabinoside combined with L-asparaginase is a useful regimen for refractory leukemia and lymphoma.

Effect of benzamide on cell growth, NAD and ATP levels in cultured chick limb bud cells.

Benzamide, an inhibitor of poly (ADP-ribose) synthetase, which augments chondrocytic differentiation, significantly decreased poly (ADP-ribose) synthetase activity in cultured chick limb bud cells. Benzamide significantly increased cell proliferation, but did not affect DNA synthesis per cultured cell. Furthermore, benzamide significantly increased NAD levels and synthesis, while it did not affect ATP levels and synthesis except for ATP synthesis on day 1. Thus, we suggest that benzamide augments both cell proliferation and NAD synthesis during chondrocytic differentiation.

Histamine content and histidine decarboxylase activity in the spleen of the magnesium-deficient rat: comparison with the skin and peritoneal mast cells.

In young rats, some effects of magnesium (Mg) depletion in the diet on histamine content and histidine decarboxylase (HDC) activity in spleen were studied in comparison with those in the skin and peritoneal mast cells. In the case of the young rats fed a Mg-deficient diet (0.001% Mg), the splenic histamine contents increased to levels about 1.3, 2.8 and 23 times as high as those in the rats fed a control diet (0.07% Mg) on the 4th, 6th and 8th day, respectively; histamine contents in the peritoneal mast cells also increased to about 1.2 and 1.8 times the control levels on the 6th and 8th day, respectively; no change was observed in histamine contents in the skin during 8 days of Mg depletion. HDC activities in the spleen of Mg-deficient rats on the 4th, 6th and 8th day increased to levels about 5.5, 15.5 and 35 times as high as the respective control values; the activities in the skin increased to about 37, 7 and 10 times the control values on the 4th, 6th and 8th day, respectively; while in the peritoneal mast cells, the activities increased to about 1.2 and 2.2 times the control values on the 6th and 8th day, respectively. On the 8th day of the Mg deficiency, some studies were made on the effects of histamine releasers on histamine contents in the spleen and peritoneal mast cells. Compound 48/80 (0.5 microgram/ml) or polymyxin B (5 micrograms/ml) induced a release of histamine from the peritoneal mast cells, but not from the spleen cells isolated from the Mg-deficient rat in vitro.(ABSTRACT TRUNCATED AT 250 WORDS)
