Pyrogen acted on mitochondrial outer membrane or structure to increase the penetration of substrate to mitochondrial MAO. Mitochondrial respiratory activity did not change significantly after acute administration of pyrogen. With chronic administration of pyrogen, the ADP/O ratio of liver mitochondria was increased. These results suggest that the acute administration of pyrogen affects the mitochondrial outer membrane, while the chronic administration affects both the outer and inner membranes.

It has been well documented that fever could be mediated with endogenous pyrogen released from reticuloendothelial system(RES) by administration of bacterial endotoxin(LPS) intravenously to rabbits. On the contrary, reserpine which has various pharmacological activities by depleting catecholamines decreases the normal body temperature as well as endotoxin induced fever. In this paper, we focussed our attention on the effect of reserpine on the production of endogenous pyrogen with relation to the antipyretic effect in endotoxin fever and obtained the following results: Endogenous pyrogen could be detected by intravenous administration of LPS(0.5 micrograms/kg) during the fever. However, endogenous pyrogen was undetectable with intracisternal administration of LPS(0.01 microgram/body) which provoked long-lasting fever. Reserpine (1 mg/kg, i.v.) decreased both body temperature induced by intracisternal administration of LPS(0.01 microgram/body) or intravenous administration of LPS(0.5 microgram/kg), however the degree was more extensive in cases of LPS-induced fever. Pretreatment of rabbits with reserpine (1 mg/kg, i.v.) suppressed the fever induced by an intravenous administration of LPS(0.5 microgram/kg), but did not suppress the release of endogenous pyrogen. These data suggest that endogenous pyrogen may not be an important factor in the pathogenesis of endotoxin fever.

In patients with sarcoma of the extremity, the tuberculin and dinitrochlorobenzene (DNCB) skin test were performed to correlate the results with the extent and prognosis of disease. Of the tumor-bearing patients, 60.0 per cent gave a positive tuberculin test and 48.9 per cent a positive DNCB test, with a significant difference from the control group with regard to the incidence of positive results in the later test (P less than 0.03). There was no difference between the different stages in the incidence of positive reactions to tuberculin but a marked difference in that to DNCB (78.6%, 40.7% and 0%). Between survivors and those who died, there was no difference in the incidence of a positive tuberculin test but a significant difference was present in that of a positive DNCB test (P less than 0.04), a difference which may be considered to reflect that in stage of disease process at the time of first examination.
A clinical investigation was carried out on a new penicillin derivative, bacampicillin with the following results. (1) Clinical effect--Bacampicillin was administered orally to 18 patients with acute simple cystitis, acute simple pyelonephritis, chronic prostatitis and acute gonorrhoic urethritis. Daily doses ranged from 750 mg to 1,000 mg. The administration continued for 4 to 14 days. Clinical effect was excellent in 10 patients and good in 5 patients. (2) Side effect--Side effects were noticed in 4 patients, one was allergic eruption and other 3 were gastralgia. In a patient the drug was discontinued to administer.

The development of pyrogenic tolerance was studied in rabbits treated with varying doses of E. coli lipopolysaccharide (LPS). The following results were obtained. 1) Development of pyrogenic tolerance seemed to proceed in two steps: that is, the first in which tolerance appeared rapidly and the second in which tolerance proceeded more gradually or steadily in response to repeated injections of a constant dose. 2) Tolerance induced by the latter method was not absolute; the rabbits were still sensitive to increased doses of LPS. 3) Rabbits immunized with E. coli vaccine lost their pyrogenic sensitivity of parent LPS to some extent. 4) Following intracisternal injection of LPS into tolerant rabbit, pyrogenic response was not decreased but rather enhanced in comparison with control. 5) The contents of nor-epinephrine and serotonin in the brain did not differ between normal and tolerant rabbits. 6) The mechanisms of endotoxin tolerance remain to be further studied.
