Fluorescent-labeled oligonucleotide probes: detection of hybrid formation in solution by fluorescence polarization spectroscopy.

Fluorescein-labeled oligonucleotides as DNA-probes were synthesized and used to monitor hybrid formation, namely to detect DNA or oligonucleotide sequence in solution. The introduction of fluorescein to oligonucleotides was carried out by oxidation of a hydrogen phosphonate linkage with ethylenediamine or hexamethylenediamine as a tether and by a subsequent labeling of the primary amine moiety by FITC. Fluorescence anisotropy, r, was adopted as an index to monitor the behavior of F-probe in solution. An increase in the anisotropy was observed upon an increase in the chain-length of F-probe. When F-Probe formed a hybrid with its complementary oligonucleotide in solution, the r value increased compared to that of F-Probe itself. These observations clearly indicate that measurements of r in solution will readily lead to the monitoring of the presence of a hybrid in solution. Consequently, it is promising to detect a certain nucleic acid sequence in solution using fluorescent-labeled oligonucleotides.

Inhibitory effects of new types of biflavonoid-related polyphenols; lophirone A and lophiraic acid, on some tumor promoter-induced biological responses in vitro and in vivo.

Lophirone A, isolated as a new type of biflavonoid-related inhibitor of Epstein-Barr virus (EBV) activation, was tested for further inhibitory properties against tumor promotion by short-term system. Lophirone A (200 micrograms) significantly inhibited inflammation of mouse ear (inhibitory effect (IE) = 70%) by 12-O-hexadecanoyl-16-hydroxyphorbol-13-acetate (HHPA, 2 micrograms). It also inhibited both Ca(2+)- and phospholipid-dependent protein kinase C (PKC) activation by 12-O-tetradecanoylphorbol-13-acetate (TPA, IC50 = 50 microM). Application of lophirone A (160 nmol) reduced the number of tumors per mouse (IE = 85%) in an initiation-promotion experiment using dimethylbenz[a]anthracene (DMBA, 0.19 mumol) and TPA (1.6 nmol) on ICR mouse skin. Lophiraic acid, a related polyphenol, was negative in all of the short-term tests. An important chemical factor which may reduce the activities of flavonoid class of inhibitors for tumor promotion was indicated.

Characterization of benzodiazepinooxazoles: crystal and molecular structure of trans and cis isomers of oxazolam.

The stereochemistry of the trans and cis isomers of 2(S)- and 2(R)-oxazolam is described. A total of four molecular structures of the 11b(R) and 11b(S) diastereomers of 2(S)- and 2(R)-oxazolam have been revealed by X-ray diffraction techniques. 2(S)-Oxazolam crystallizes in space group P2(1)/c, with cell dimensions a = 16.180(2) A, b = 8.791(2) A, c = 13.574(2) A, and beta = 102.07(1) degrees. The molecule in an asymmetric unit exhibited disorder at the 2-carbon (C2) atom, and the structures for trans and cis isomers have been resolved crystallographically. 2(S)-Oxazolam forms polymeric chains with intermediation of ethanol through hydrogen bonding. 2(R)-Oxazolam crystallizes in space group P1, with cell dimensions a = 7.754(1) A, b = 8.654(1) A, c = 15.795(2) A, alpha = 104.09(1) degrees, beta = 91.19(1) degrees, and gamma = 114.00(1) degrees. A similar disorder at the C2 position has been elucidated by a mixture of trans- and cis-2(R)-oxazolams. The two 2(R)-oxazolam molecules form a dimer with a hydrogen bond net. The structural details, focusing on the conformation and molecular complex formation of both oxazolams, have been discussed in connection with the oxazolidine ring stability and the crystal polymorph.

A phase II pilot study of the combined application of hyperthermia and intra-hepato-arterial chemotherapy using cisplatinum and 5-fluorouracil.

Fourteen patients (group A) with unresectable metastasis to the liver from colorectal cancers (11 patients) and gastric cancers (3 patients) were treated with the combined application of hyperthermia and intra-hepato-arterial (IHA) chemotherapy with cisplatinum and 5-fluorouracil. Ten patients were treated with regional hyperthermia applied with a radiofrequency capacitive heating system and four patients were treated with total-body hyperthermia concurrently with IHA chemotherapy. Thirty-one patients (group B) with liver metastases from colorectal and gastric cancers received only IHA chemotherapy with the same chemotherapeutic regimen. In group B, partial responses (PRs) were obtained in 8 of 17 evaluable patients with colorectal cancer and in 1 of 10 evaluable patients with gastric cancer. In group A, PRs were found in 6 of 11 patients with colorectal cancer and in 2 of 3 patients with gastric cancer. Furthermore, improvements due to the combination of chemotherapy with hyperthermia were noted in 3 of 6 patients with colorectal cancer who had received unsuccessful prior IHA chemotherapy. The 50% survival period was prolonged to 23 months in group A from 11 months in group B, for patients with colorectal cancer. Toxicity of IHA chemotherapy was not potentiated by combination with hyperthermia. These results indicate that the combination of hyperthermia with IHA chemotherapy has a therapeutic benefit in the treatment of unresectable metastatic liver tumors derived from colorectal and gastric cancers.

Heating efficiency of radiofrequency capacitive hyperthermia for treatment of deep-seated tumors in the peritoneal cavity.

We analyzed heating profiles from 318 hyperthermic treatments of 39 patients with recurrent or inoperable cancers of the digestive organs whose deep-seated tumors were treated by radiofrequency (RF) capacitive heating of the abdominal region, and we investigated the heating efficiency and antitumor effect of such treatment. It was apparent that heating with a mean maximum RF output of 1,000 watts (700 watts at least), repeated four times or more, was necessary for a high rate of response by the tumor. Although it was difficult to heat tumors of the bile duct/pancreas to 42 degrees C or more, there was a strong positive correlation between maximum output of RF energy and maximum temperature of tumors (r = 0.839, P less than 0.001). The antitumor effect of RF hyperthermia was augmented with increasing output of RF energy. Therefore, the maximum level of RF output may be a useful index for expressing the heating efficiency with respect to intra-abdominal deep-seated tumors.

Identification of RB and p53 mutations in mouse lymphoma cell lines.

We analyzed the genomic structure and mRNA of the RB and p53 genes in four mouse lymphoid leukemia cell lines (DL-1, DL-5, DL-8, and DL-12). Although no gross structural alteration of the RB gene was observed in any cell line, abnormalities of RB mRNA were detected in at least two cell lines. RB mRNA expression was greatly reduced in DL-12. In addition, cloning and sequencing analysis of the RB cDNA revealed that the RB mRNA in DL-8 had a 276-nucleotide deletion presumably consisting of exons 10, 11, and 12, suggesting that altered splicing resulted in the loss of these exons. Analysis of the p53 gene indicated that DL-5 had a deletion in both alleles and expressed a smaller mRNA. These results suggest that mutations of the RB or p53 genes, or both, are associated with lymphoid leukemogenesis in mice.

Diagnostic accuracy of combination of assays for immunosuppressive acidic protein and carcinoembryonic antigen in detection of recurrence of gastric cancer.

Two tumour markers, immunosuppressive acidic protein (IAP) and carcinoembryonic antigen (CEA), were assayed in gastric cancer patients. Levels of IAP and CEA were measured simultaneously in the preoperative and postoperative periods. The usefulness of the combined assay of these markers for detection of recurrence of cancer was investigated in terms of sensitivity, specificity and diagnostic accuracy. Sensitivity was not high (69.2%), but specificity and diagnostic accuracy were 96.7% and 86.9%, respectively. In cases with metastases in the liver, sensitivity (100.0%), specificity (100.0%) and diagnostic accuracy were high. In cases of peritoneal dissemination, these indices were low. The combination assay of IAP and CEA appears to be useful for detection of recurrence of gastric cancer, especially in patients with liver metastases.

Effect of imposed head vibration on the stability and waveform of flagellar beating in sea urchin spermatozoa.

The heads of live spermatozoa of the sea urchin Hemicentrotus pulcherrimus were held by suction in the tip of a micropipette mounted on a piezoelectric device and vibrated either laterally or axially with respect to the head axis. Within certain ranges of frequency and amplitude, lateral vibration of the pipette brought about a stable rhythmic beating of the flagella in the plane of vibration, with the beat frequency synchronized to the frequency of vibration [Gibbons et al. (1987), Nature 325, 351-352]. The sperm flagella, with an average natural beat frequency of 48 Hz, showed stable beating synchronized to the pipette vibration over a range of 35-90 Hz when the amplitude of vibration was about 20 microns or greater. Vibration frequencies below this range caused instability of the beat plane, often associated with irregularities in beat frequency. Frequencies above about 90 Hz caused irregular asymmetrical flagellar beating with a marked decrease in amplitude of the propagated bends and a skewing of the flagellar axis towards one side; the flagella often stopped in a cane shape. In flagella that were beating stably under imposed vibration, the wavelength was reduced at higher frequencies and increased at lower frequencies. When the beat frequency was equal to or lower than the natural beat frequency, the apparent time-averaged sliding velocity of axonemal microtubules, obtained as twice the product of frequency and bend angle, decreased with beat frequency in both the proximal and distal regions of the flagella. However, at vibration frequencies above the natural beat frequency, the sliding velocity increased with frequency only in the proximal region of the flagellum and remained essentially unchanged in more distal regions. This apparent limit to the velocity of sliding in the distal region may represent an inherent limit in the intrinsic velocity of active sliding, while the faster sliding observed in the proximal region may be a result of passive sliding or elastic distortion of the microtubules induced by the additional energy supplied by the vibrating pipette. Axial vibration with frequencies either close to or twice the natural beat frequency induced cyclic changes in the waveform, compressing and expanding the bends in the proximal region, but did not affect bends in the distal region or alter the beat frequency.

Effect of aging, castration, and testosterone administration on ribonuclease and ribonuclease-inhibitor activities in the prostate of rats.

Free ribonuclease (RNase)-inhibitor activities in both ventral and dorsal prostates had their highest peaks in 4-week-old rats and smallest peaks in around 7-week-old animals. Total RNase activity in the ventral prostate decreased overall with age, while that in the dorsal prostate increased. No significant amount of free RNase activity was found in either prostate. Weight, protein content, and free RNase-inhibitor activity in both prostates decreased after castration and increased after administration of testosterone to castrated rats. Total RNase activity in the ventral prostate was increased by castration and decreased by testosterone administration. In the dorsal prostate, total RNase activity had two peaks, 7 d after castration and 2 d after testosterone administration. A large amount of free RNase activity was found in the ventral prostate 7 d after castration and this activity was decreased by testosterone administration. In the dorsal prostate, free RNase activity was not detected after castration and testosterone administration. These results suggest that changes in the level of RNase-inhibitor in both prostates are involved in the regulation of their RNA content through the control of free RNase activity.

[A case of ruptured aneurysm of the descending thoracic aorta associated with active Behçet's disease].

A 63-year-old male receiving steroid for active Behçet's disease was admitted of massive hemoptysis. A pseudoaneurysm of about 5 cm in diameter caused by rupture of the aortic wall penetrating into the lung was detected on the descending thoracic aorta. Operation was performed using left atrium-femoral artery bypass with centrifugal pump. The technique of graft interposition was selected rather than patch closure to prevent postoperative anastomotic aneurysm. We consider that in the treatment of inflammatory aneurysm, use healthy aortic wall for suture and end-to-end anastomosis are very important. In spite of steroid supplementation, the inflammatory reaction get worse postoperatively and the dosage of steroid had to be increased to control inflammation. The patient is well 20 months after surgery.

[A case of a calcifying odontogenic cyst associated with odontoma inducing impaction of a deciduous tooth].

A 4-year-old boy was examined with a chief complaint of "Delayed eruption of maxillary right deciduous central incisor". Oral examination revealed a firm swelling on the labial gingiva at the right deciduous incisor of maxilla. Radiographic examination revealed that odontoma-like substances disturbed the eruption of the central deciduous maxillary incisor. The odontoma-like substances were removed surgically and examined histopathologically. The thin sections showed findings typical of a Calcifying Odontogenic Cyst with Odontoma.

Characterization of antisense DNA derivatives having stereoisomeric linkages.

The isomer separation of oligonucleoside phosphoramidates (OPA) by RPLC was studied. All stereoisomers of OPA-tetramer (dATCG) modified with isopropylamine could be separated and they showed different CD spectra each other. OPA-decamer (dGGGCATCGTC) modified with 3-amino-1-propanol could be separated into five fractions. Each fraction was found to have different ability to form hybrids with its complementary oligonucleotide, indicating that it is possible to exclude stereoisomers which can hardly bind to target nucleic acids.

Effects of base damages on DNA replication.

Understanding the response of DNA polymerase to the encountered damage in a template is a key to assessing lethal and mutagenic events of cells exposed to genotoxic agents. In the present study M13 (or f1) DNA templates containing 4 types of thymine damages were prepared, and DNA synthesis was carried out in vitro with the templates. The extent of inhibition of DNA synthesis by the damages was evaluated by measuring [3H]dTMP incorporation. Furthermore, newly synthesized DNA was analyzed on a sequencing gel to determine termination sites of DNA synthesis. The results showed that DNA synthesis was differentially inhibited by the damages, and the termination sites of DNA synthesis were dependent on the structures of the damages and the 3'-5' exonuclease activity of DNA polymerase used.

Unusual signal broadening in NMR of oligonucleotides.

Solution structure of single-stranded small oligonucleotides (up to pentamer) has been studied by 1H-NMR (600 MHz) and it has been found that when those having some specific base sequences produce unusual broadening of the lines attributed to the protons of guanine residues. These broadened signals did not become sharp even at elevated temperature. This extremely unusual behavior has been attributed to the interaction between the guanine residues.

[A study of glomerular damage due to extracorporeal circulation--meaning and usefulness of measuring of urinary microalbumin].

An increase of excretion of urinary microalbumin (U-Alb) is thought to demonstrate earlier glomerular damage than deterioration of Glomerular Filtration Rate (GFR). In this study, the meaning and usefulness of measuring U-Alb before, during and after Extracorporeal Circulation (ECC) was estimated. In 25 cases of adult cardiac surgery, we measured U-Alb, and simultaneously as current indices of glomerular function, serum and urinary creatinine, blood urea nitrogen, serum beta 2-microglobulin, urine volume, and cardiac output were measured. Albumin Excretion Rate (AER) [U-Alb X urine volume] and Albumin Index (AlbI) [U-Alb/U-Cr] were calculated as indices of U-Alb, and Creatinine clearance (Ccr) was calculated as GFR. The changes of each parameter, the correlation AER and AlbI during and after ECC with parameters that relate to ECC, and the assessment of the prolongation of glomerular damage after operation that possibly progress to acute renal failure were investigated. Our results showed AER and AlbI to increase following ECC with high values maintained during and after ECC, in spite of recovery of Ccr, from 24 hours to 4 days postoperatively. AER and AlbI recovered at 7 days and 14 days after surgery, respectively. The existence of glomerular damage, not indicated by a change in Ccr, was indicated by increases of AER and AlbI. The damage was thought to be temporary glomerular dysfunction. In correlation of AER and AlbI with ECC parameters, ECC of long duration influenced the glomerular damage as indicated by an increase of AER and AlbI.(ABSTRACT TRUNCATED AT 250 WORDS)

[A successful case report of mediastinitis after coronary bypass surgery treated with total excision of the sternum and omental transposition].

A 61-year-old male case, who was complicated with mediastinitis after coronary bypass surgery with left internal mammary artery, was reported. He was treated with open packing of gauze saturated with povidone iodine solution for 17 days, and thereafter pedicle transposition of great omentum with total excision of the sternum as a second procedure, was performed. His postoperative course was uneventful without any respiratory complication. From this experience, we concluded that omentum was very effective strategy in management of mediastinal infection following cardiac surgery.

[A case of esophageal duplication cyst associated with a total left pericardial defect].

Esophageal duplication cyst associated with pericardial defect in a 57-year-old man is reported. Differential diagnosis was very difficult in the preoperative examination. The tumor was a cyst and its wall was a perfect replica of normal esophagus e.g. esophageal duplication, and total left pericardial defect also was found at the operation. There was no malignant or inflammation findings in the excised specimens. The association of esophageal duplication cyst associated with pericardial defect is very rare.

[Primary cardiac malignant mesenchymal tumor containing chondrosarcomatous component--a case report].

A case of 70 years old female bearing a primary cardiac tumor was presented. Mitral valve replacement and reconstruction of the interatrial septum were necessary because the tumor originated from the left atrial side of the interatrial septum and invaded the anterior mitral valve leaflet and around the annular area. Pathological examination of the resected specimens revealed mesenchymal tumor containing chondrosarcomatous component. The patient received radiation therapy (50 Gy) postoperatively and she resumed normal daily activity. Preoperative echocardiographic manifestations were of proved benefit in the diagnosis of malignant cardiac tumor and a proper therapeutic strategy could be chosen.
