Cellular immunity in asbestosis.

A highly statistically significant correlation was found between asbestosis and impaired responsiveness in skin reaction to intermediate and second strength tuberculin and SK-SD. Considering ANA incidence these antibodies were found with higher frequency in asbestos workers who lacked a cutaneous response to the recall antigens. In asbestosis cases peripheral blood lymphocyte profiles are also abnormal demonstrating low proportions of E-RFC. Moreover, MIF test results showed the impairment of this cytokine generation when lymphocytes were stimulated with SK-SD, PPD and PHA in asbestosis cases. The lymphocyte transformation study documents an impaired response mainly to a lower dose of PHA and ConA in asbestosis cases and in asbestos workers with ANA.

[Behaviour of drugs under conditions of ultrasonic thermo aerosols (author's transl)].

The behaviour os histamine, acetylcholine, mucosolvine, panthenol, prednisolute and dexamethasone is investigated under conditions of thermoaerosol in the USI 50. The concentration of the drug is estimated in the original solution, in the aerosol and in the solution remaining in the nebulizer by means of specific chemical methods. The concentration in the aerosol is nearly unchanged in the case of acetylcholine, panthenol and dexamethasone. In the case of histamine there is only an insignificant decrease. Using prednisolute and dexamethasone the concentration amounts to 86%. Generally the concentration of the solution in the nebulizer increases. Decompositions or alterations of the drugs are not to be seen spectrophotometrically and by means of thin layer chromatography, nor is the pH-value changed. Signs for reactions or interferences with the nebulizer are to be observed in some cases. Therefore regular controls are necessary. In all cases of changing parts of the apparatus the needed material is to be tested concerning its indifference against the drugs used under conditions of thermoaerosol.

Diagnostic specificity of autoantibodies. III. Age related changes in appearing autoantibodies serum IgD levels and their association with peripheral blood lymphocyte profiles.

Comparisons made between old and young people depicted the increase with aging in ANA and thyroid antibody incidence. In contrast to these antibodies SMA were frequently seen in young women. Also in women serum IgM levels were higher as compared to men. Conversely serum IgD levels were higher in men; both these differences were seen in the young group only. Proportions and numbers of E-RFC were significantly reduced in the older group of people as compared to the values of the younger group. Interestingly, the autoantibodies appearing were correlated in women with a lower percentage of E-RFC. Also in women low IgD levels were associated significantly with higher percentage of E-RFC.

Hepatitis associated antigen, autoantibodies and serum immunoglobulin levels in patients with liver injury following administration of antituberculosis drugs.

32 patients with liver injury following the administration of tuberculosis drugs were examined for HAA, ANA, SMA and serum immunoglobulin levels. HAA was found in 38-7% of patients. SMA were detected in some HAA positive as well as in HAA negative cases. ANA except in one case were found in the HAA negative group. Serum IgM levels are significantly higher in patients with liver injury as compared to the normal values. The highest levels of IgG and IgM were found in the ANA or/and SMA-positive cases.

HLA antigens in asbestosis.

One hundred thirty-four workers in an asbestos factory who had been in contact with asbestos for about 20 years, were typed for 22 HLA antigens of the A and B loci. In this group, 22 workers were diagnosed as asbestosis patients on the basis of chest radiography and clinical findings; the remaining 112 were without signs of asbestosis. In the group of asbestosis patients an increase of B27 antigen was observed in comparison with the group without asbestosis (27-27% and 9-82% respectively). The relative risk = 3-44 is significant at the level of p less than 0-05. These results may indicate that a genetic factor, connected with HLA locus has influence on the development of asbestosis under conditions of contact with asbestos.

A double antibody solid phase assay for DNA autoantibodies for clinical use.

DNA antoantibodies in serum will bind to antigen-coated polystyrene tubes and can be detected by radiolabelled anti-immunoglobulin. The method is quantitative, gives information on the antibody class and is not readily subject to interference by factors such as the size of the DNA, minor contamination of double-stranded with single-stranded DNA and the presence of materials which can combine "non-specifically" with the antigen. Double-stranded DNA gave good discrimination between SLE and rheumatoid arthritis but with single-stranded preparations approximately 40% of the RA patients showed elevated antibody values. Individual results obtained with the two antigens were compared and correlated with the Farr test and ANA titres. Surprisingly, a proportion of the SLE sera gave high background binding to tubes coated with gelatine.

[Differentiation and identification of mycobacteria by means of thin-layer chromatography of their lipids. II. Application for the classification of slowly growing mycobacteria: M. avium and M. intracellulare (author's transl)].

Experiences with the thin-layer chromatography of mycobacterial lipids for the differentiation of the species M. avium and M. intracellulare are described. Details of the procedure used have been published in a recent paper. The 61 strains investigated belong to 16 different serotypes. Twenty of the 21 strains of M. avium present three distinct lipid patterns corresponding to the serotypes 1, 2 and 3. One strain had an aberrant lipid pattern suggesting that serotyping was at fault. The results obtained with 40 M. intracellulare strains of the serotypes III, IV, VI, VII, Davis, Watson, Howell, Chance, Boone, Yandle, Wilson, Altman and Arnold show that a differentiation by thin-layer chromatography of their lipids--using two different developing solvents--is possible. In allbut one case there was a complete correspondence between classification by lipid analysis and the results of serological typing. The distinction between M. avium and M intracellulare on the basis of species specific lipid patterns is not possible. This supports the conclusion that M. avium and M. intracellulare appear to be the same species having a number of distinctive serotypes.

[Differentiation and identification of mycobacteria by means of thin-layer chromatography of their lipids. III. Application for the classification of scotochromogenic mycobacteria (author's transl)].

Experiences with the lipid analysis of scotochromogenic mycobacteria by means of thin-layer chromatography are described. Out of 18 strains of M. scrofulaceum 11 belong to the serotypes Lunning, Gause, and Scrofulaceum. The similarity of the lipid patterns of the M. scrofulaceum strains to the ones of the M. avium-M. intracellulare group supports the contention of some authors that these species are closely related. Lipid analysis of further species of RUNYON's group II (M. xenopi, M. gordonae, M. kansasii var. aurantiacum, M. flavescens, M. acapulcensis, M. paraffinicum, M. thermoresistibile) gives the evidence that a differentiation by means of thin-layer separation is possible in these cases, too. The answer to the question whether these lipid patterns are species specific or not can only be given when a large number of strains of every species will be investigated.

[Electromyographic diagnosis of tetany].

Whilst the classical picture of tetany is easily to be diagnosed, diagnostic problems may arouse if vegetative, visceral or vascular symptoms predominate. The well-known clinical signs of provocation (Chvostek, Trousseau, Bonsdorff) frequently do not allow any diagnostic ascertainment. With the help of the classical electrodiagnostics after Erb (in the positive case the provocation of the cathodal opening contraction with an intensity of current below 5 mA is possible) or the electromyography we, as a rule, succeed in proving the diagnosis. In the EMG characteristic repetetive potentials in derivation from a small hand muscle are found, either spontaneous or after provocation by means of ischaemia and hyperventilation. The electromyographic examination is regarded as the most sensitive method in the diagnostic spectre at disposal and should be used for proving or excluding a tetany in unclear stenocardias, vasomotor circulatory disturbances, in spastic disturbances in the gastrointestinal tract, in unclear headache as well as in tetanic cataract and after operations of the thyroid gland.

[Differentiation and identification of mycobacteria by means of thin-layer chromatography of their lipids. I. Application for the classification of rapidly growing mycobacteria (author's transl)].

Experiences with the analysis of mycobacterial lipids by means of thin-layer chromatography are described. The influence of different media used for the cultivation of the strains, the age of cultures and heating of the bacteria on the lipid patterns was investigated. 25 strains of rapid growers belonging to various species were examined by a definite procedure. As the results indicate the lipid analysis allows a differentiation of more species than with the usual culture and enzymatic methods. The conclusions especially for taxonomic problems are discussed.
