Synthesis of 4-(3-benzazolymethyl)aminodiphenyl ethers and other related products as potential antimicrobial agents.

4-Aminodiphenyl ethers (2) obtained by reducing the corresponding nitro analogue (1), have been incorporated into benzazoles, phthalimides and 4-quinazolone in the presence of 40% formalin to furnish a series of 4-(3-benzazolymethyl)aminodiphenyl ethers (3) under the conditions of the Mannich reaction. In an alternate procedure, 3 were obtained on refluxing the N-hydroxymethyl derivatives of benzazoles etc. with 2 in methanol. A few amine exchange reactions were also performed and the stability of a few representative compounds was studied in different acidic media. The antibacterial spectrum of all the synthesized compounds was determined against Bacillus subtilis as well as Bacillus pumilus by the agar diffusion technique.

Serum high density lipoprotein cholesterol in patients with abnormal coronary arteries.

Previous studies showing the inverse relationship between high density lipoprotein cholesterol (HDL-C) and coronary artery disease were based on myocardial infarction survivors and presumably normal subjects. To determine whether a similar relationship exists between patients with abnormal coronary arteries (ACA) and those with normal coronary arteries (NCA), the serum HDL-C and other lipoproteins of these patients and those of a group of presumably healthy control subjects (CTL) were determined. The ACA males had lower HDL-C and % HDL-C but higher TG, VLDL-TG, LDL-C/HDL-C and VLDL-C/HDL-C than the NCA and CTL males. They also had higher VLDL-C and % VLDL-C than the CTL males. Adjustment of HDL-C for serum TG eliminated the difference in HDL-C between the ACA and NCA groups but that between ACA and CTL groups remained. The ACA females had lower % HDL-C than the NCA and CTL females. They also had lower HDL-C but higher LDL-C/HDL-C and VLDL-C/HDL-C than the CTL females. The NCA and CTL groups did not differ in any of the lipid variables, although the NCA group values were intermediate to those of the ACA and CTL groups. Using various lipoprotein profiles, it was possible to classify the patients into the 3 groups.

Defective yeast opsonisation of serum in sarcoidosis.

We have studied the opsonising ability of the sera of 49 patients with sarcoidosis. The serum of 11 (22%) patients was defective in this ability, whereas only two (4%) of 46 clinic control subjects and three (7%) of 43 laboratory control subjects showed this defect. The difference between the prevalence of the defect in sarcoidosis and the control groups was statistically significant (p less than 0.01). Although the patients with sarcoidosis who had this defect tended to have pulmonary infiltration, this relationship was not statistically significant. Similarly, there was no correlation between the activity of sarcoidosis and this defect, although there was a significant (p less than 0.05) relationship between the opsonising defect and the persistence of circulating immune complexes. Serum opsonisation is a genetically linked host defense mechanism. Our findings suggest that the presence of this serum defect may render a person more susceptible to sarcoidosis.

Defective yeast opsonisation of serum in tuberculosis.

We have studied the opsonising ability of the sera of 92 patients with tuberculosis. Fourteen per cent of these patients showed defective opsonising ability compared with 4% in a clinic control group. This increased frequency in TB was accounted for by the greatly increased proportion with defects found in patients with intrathoracic TB (21%). We may have identified a section of the population with a specific genetically linked abnormality of a host defence mechanism which renders them more susceptible to intrathoracic TB. Further population studies are required.

Effects of phenylbutazone, tolbutamide, and clofibric acid on binding of racemic warfarin and its enantiomers to human serum albumin.

The effect of phenylbutazone, tolbutamide, and clofibric acid on the binding of racemic warfarin and its enantiomers to human serum albumin was studied by equilibrium dialysis. Warfarin had one primary and two secondary binding sites on the albumin molecule. No difference in binding was detected at the primary binding site; the extent of R(+)-isomer binding at the secondary binding sites was 2.5 times greater than the corresponding S(-)-isomer binding. Phenylbutazone and warfarin appear to compete for the same primary binding site on the albumin molecule. Tolbutamide interferes with the binding of warfarin enantiomers at their secondary sites. Clofibric acid has a less pronounced effect on warfarin binding than does phenylbutazone or tolbutamide.

80% of patients with intrinsic asthma are homozygous for HLA W6. Is intrinsic asthma a recessive disease?

Of 26 patients with intrinsic asthma, 21 (81%) were homozygous for the histocompatibility antigen HLA-W6. Also, half the patients had complement defects, in particular low levels of C2. This is the first indication of a strong genetic component in intrinsic asthma, apart from that already known from family studies. These findings suggest that intrinsic asthma may be a recessive disease.

Characterization of an alkaline subtilopeptidase type Pfizer.

The physiochemical properties, amino acid composition and profile of the the tryptic peptides for an alkaline subtilopeptidase type Pfizer have been determined. The enzyme is stable in the pH range from 5 to 10, has a pH optimum of 9.5 to 10, and is relatively stable for a period of 2 h up to a temperature of 50C. Homogeneity was demonstrated by electrophoretic techniques and the mobilities indicated on isoelectric point of 8.7. The molecular weight was found to be 25,000 by gel filtration. The amino acid composition was found to be Ala32, Arg4, Aspgamma8, Glu15, Gly29, His4, Ile9, Leu13, Lys11, Met5, Phe4, Pro14, Ser31, Thr17, Tyr9, Val22, a total of 247 amino acid residues. The enzyme does not contain either disulfide bonds or cysteine, and lacks tryptophan as well. The N-terminal end-group residue is alanine: the C-terminal amino acid is arginine. Tryptic hydrolysis of the enzyme produced 15 peptides which were separated by gradient elution on Dowex 50-X2. The amino acid composition of each appropriately purified tryptic peptide was established.
