The effect of intracerebroventricular (i.c.v.) treatment of alpha-human atrial natriuretic polypeptide on the vasopressin secretion induced by i.c.v. injection of angiotensin II was studied in conscious, unrestrained rats. Pretreatment of alpha-human atrial natriuretic polypeptide (5 micrograms) significantly inhibited the angiotensin II (100 ng)-stimulated vasopressin secretion. This result suggests that atrial natriuretic polypeptide plays regulatory roles in vasopressin secretion in the central nervous system.

A radioimmunoassay for chicken calcitonin in chicken ultimobranchial glands was established utilizing a rabbit antiserum against eel calcitonin. This assay method, which is about 100 times as sensitive as the usual bioassay for hypocalcemic activity, was used for monitoring chicken calcitonin during its purification. The immunoreactivity in chicken ultimobranchial extract was separated by SP-Sephadex C-25 chromatography into two fractions. Chicken calcitonin I, which was occurred in the major immunoreactive fraction, was further purified to homogeneity as shown by reverse phase HPLC. In the end, 39 nmol of chicken calcitonin I was obtained from 3,384 chickens following a 12,000-fold purification. The complete amino acid sequence of purified chicken calcitonin I was determined to be H-Cys-Ala-Ser-Leu-Ser-Thr-Cys-Val-Leu-Gly-Lys-Leu-Ser-Gln-Glu-Leu-His-Ly s-Leu-Gln-Thr-Tyr-Pro-Arg-Thr-Asp-Val-Gly-Ala-Gly-Thr-Pro-NH2 and confirmed by synthesis. The specific biological activity of chicken calcitonin I (4,500 MRCU/mg) was identical to that of eel calcitonin, which has the highest specific biological activity among the calcitonins so far isolated. Chicken calcitonin I resembled the calcitonins from the ultimobranchial glands both of salmon and eel in sequence, biological activity, and immunological property.

A highly sensitive and simple radioimmunoassay for the measurement of 8-arginine vasopressin (AVP) in human plasma has been developed. The dose response curve ranges from 0.025 to 8 pg/tube. This simple extraction technique employing an ODS C18 column recovered 87.1 +/- 10.4 (mean +/- SD)% of AVP in the range of 1-10 pg/ml added to 0.5 ml plasma. Determination of AVP in each fraction of plasma, which was gel-filtrated through a Sephadex G25 (1 X 25 cm), revealed that the fraction of plasma AVP was superimposed on that of authentic AVP, and interference of non-specific substances was completely eliminated by an ODS C18 column. Using the assay, 13 of 16 patients with diabetes insipidus (DI) showed plasma AVP concentrations ranging from 0.03 to 0.21 pg/ml, and the other 3 patients had less than 0.03 pg/ml. The AVP concentrations of DI were clearly distinguished from those obtained in normal subjects (0.30-4.20 pg/ml, n = 65). The within and between assay variability was about 10% each. Plasma AVP concentrations (mean +/- SD) of normal subjects (n = 6) standing, sitting, and supine after an overnight of fluid deprivation were 2.41 +/- 1.15, 1.95 +/- 0.85 and 0.97 +/- 0.48 pg/ml (30 min after) respectively. Plasma AVP concentrations of normal subjects (n = 6) after water load (20 ml/kg wt) were clearly reduced from 1.89 +/- 1.00 (before) to 0.42 +/- 0.21 (standing for 60 min) and also from 0.89 +/- 0.41 to 0.40 +/- 0.22 pg/ml (supine for 60 min).
Mizoribine, an immunosuppressive agent developed and marketed in Japan, was tested in experimental keratoplasty and in Behçet's disease. The drug, 2 to 4 mg/kg of body weight, administered systemically or as 10% eyedrops five times daily, suppressed immune reaction after keratoplasty in rabbits. It seemed effective in 18 cases including 12 regrafted eyes. In 29 cases of Behçet's disease, the average recurrence rate dropped from once per month to once every three months in one year. Mizoribine is safe, easy to use, and may be of some value by itself or in combination with other drugs in the treatment of uveitis and in the prevention of corneal graft reaction.

Liquid protein diets have been widely used in the United States to control weight gain. Pregnant rats were fed a standard casein diet in which liquid protein was substituted for the protein portion; all other necessary nutrients were supplied. The average incidence of cataracts in the offspring from mothers fed liquid protein was 25%. Offspring born with cataracts were successfully raised. The results suggest that nutrition may play an important role in cataract formation in newborns, and that the use of liquid protein during pregnancy may be related to ocular abnormalities in the offspring.
A specific and sensitive radioimmunoassay for human pancreatic growth hormone-releasing factor (hpGRF) was developed. Using this radioimmunoassay, it was found that immunoreactive hpGRF (which is present in hypothalamic tissues) of at least two different molecular sizes is often produced by small cell carcinoma of the lung.
A case of corneal tattoo was studied histologically, and by electron microscopy. Most of the electron dense granules were seen in keratocytes, either singly or in clumps; some were found between collagen fibrils. Platinum was localized in the granules by means of x-ray microanalysis.
