Pharmacokinetics of theophylline and its metabolites in rabbits.

The pharmacokinetics of theophylline (1,3-DMX) and its metabolites were investigated in detail in four male rabbits after bolus intravenous injection (12 mg/kg) of the compound. Theophylline was measured in blood and urine, and its metabolites were determined only in urine. Apparent first-order rate constants for the metabolic processes involved in the formation of 1,3-DMX metabolites and their excretion in urine were calculated. An appropriate 13-compartment model was formulated to describe the disposition of 1,3-DMX and its metabolites.

[Current diagnosis and therapy of undescended testis].

Although the etiology of maldescended testes is multifactorial, dysfunction of hypothalamic-pituitary-gonadal axis should be considered as the most important factor. The hormonal treatment with Gn-RH on its own or followed by hCG can provoke testicular descent and should be applied at 10th month of life. Failure, partial or transient success needs orchidopexy within the first two years of life to prevent infertility. If a concomitant hernia or a true ectopia is encountered, if patients are seen at an adolescent age and if the testes are truly cryptorchid, surgery is the treatment of choice.

[Verification of therapeutic levels of procainamide and N-acetyl- procainamide in ventricular arrhythmia].

Therapeutical efficacy was clinically evaluated in 21 patients with ventricular cardiac arrhythmias. The drug was given orally with preceded intramuscular dose. Therapeutic effect was verified by the measurements of procainamide and N-acetylprocainamide concentrations in blood serum to determine the minimal effective concentration of the drug required to obtain satisfactory antiarrhythmic effect. Procainamide proved effective in cardiac arrhythmias in 14 patients (66.7%) with statistical significance in the acute myocardial infarctions; blood serum procainamide plus N-acetylprocainamide levels being were below the therapeutical range. The poor correlation of the dose of the drug and respective procainamide, N-acetylprocainamide concentrations in blood was observed. Relationship of the therapeutical effects blood serum level of the drug should be estimated basing of the assays of both procainamide and N-acetylprocainamide .

Correlation between serum concentration and hypoglycemic effect of chlorpropamide in man.

The relationship between the serum concentration and some of pharmacokinetic parameters of chlorpropamide and the hypoglycemic effect of this drug was studied. Studies were carried out in a group of 18 patients in whom the concentrations of drug and sugar in blood were determined simultaneously. Close correlation between the concentration and some pharmacokinetic parameters of chlorpropamide and the intensity of decrease of the blood glucose level as well as the time of its occurrence has been found.

[Small intestine transplantation--current status of animal experiments].

The most frequently used methods are discussed of small intestine transplantation (SIT) in experimental models. The advantages and drawbacks of various operational techniques are characterized. The problems are presented associated with the intensity of graft rejection reaction. The presently used methods of preservation of small intestine taken for transplantation are discussed.

[Small intestine transplantation--immunologic problems in experimental studies].

The basic immunological problems are discussed associated with small transplantation (SIT) in experimental models. The types of immunological reactions are characterized. The status is presented of studies on the used methods of immunosuppression and the decrease of immunogenicity of graft. The role is stressed of histological monitoring of graft rejection reaction.

[Immunologic state and effect of long-term levamisole therapy on selected immunologic parameters in children with steroid sensitive nephrotic syndrome during remission].

Immunological system was tested in 14 children aged between 1.5 and 14 years (mean 5.4 years) with steroid-sensitive nephrotic syndrome during remission. Levamisole (Decaris Richter) was administered in the dose of 2-3.5 mg/kg twice a week for 2.5-6 months. Immunological tests carried out before treatment showed a decrease in total T-cells and T2 subpopulation in 78.6% of patients in comparison with normal values for this age group; a decrease in total B-cells was noted in 87.6% of patients, and percentage of these cells was decreased on 57.1% of cases; gamma-globulins in blood serum were decreased below lower normal values in all children whereas serum IgM levels were above the upper limit of the normal values. Therapy with levamisole normalized total T-cells in 91.7% of patients (p < .05) mainly due to an increase in the number of T1 cells (by 127% in 7 patients in comparison with baseline values considered as 100%); total number of B-cells reached the values normal for this age group in 83.1% of patients (< .05) while percentage of these cells normalized in 66.7% of patients (p < .05), increasing by mean 66.7% in 6 children. It seems that immunologic status of patients with steroid-sensitive nephrotic syndrome during remission depends, at least partially, on the recurrent respiratory tract infections and/or urinary tract infections accompanying the underlying disease. No significant levamisole effect on blood SGOT, SGPT, platelet count and leucocytosis was noted.
