Habitual dislocation of the hip.

We report the case of a girl, aged 2 years, who was able to dislocate her right hip voluntarily and in whom there was no other abnormality. She was given psychological treatment and the condition resolved after 2 years. Eight similar cases of habitual dislocation of the hip are reported in the world literature.

A PvuII polymorphism in the 5' flanking region of the apolipoprotein AIV gene: its use to study genetic variation determining serum lipid and apolipoprotein concentration.

We have used a 1.05-kb unique genomic fragment from the 5' end of the apolipoprotein (apo) CIII gene to identify a restriction fragment length polymorphism (RFLP) detected with the restriction enzyme PvuII, in the apoCIII-apoAIV intergenic region. In a sample of 220 normolipidaemic individuals from the UK population, the frequency of the rare allele, VB2 is 0.054. The PvuII polymorphism is in apparent linkage equilibrium with three other RFLPs of this gene cluster, detected with the restriction enzymes XmnI, PstI and SstI, but in linkage disequilibrium with an RFLP in the apo-CIII gene also detected with PvuII. Taken together, these five RFLPs have a PIC (polymorphism information content) value of 0.8, and therefore are informative for genetic studies. Individuals with the genotype VB1VB2 had lower mean concentrations of apoAI, and HDL-cholesterol than individuals with the genotype VB1VB1. However these differences were not statistically significant.

Effects of verapamil and nifedipine on human uterine artery "in vitro".

We have studied, in isolated human uterine arteries, the effects of verapamil and nifedipine on the contractions induced by noradrenaline (NA) and KCl in standard Krebs and those produced by CaCl2 and NA added to a free-calcium medium (128.5 mM K+). Both calcium antagonists reduce in a dose-dependent way the contractions induced by NA, KCl and CaCl2. Smaller doses of nifedipine were needed to produce the same inhibitory effects. The maximal doses of calcium antagonists utilized had no effect on the contractions induced by NA when the calcium was removed from the medium. NA, in equal doses, had similar contractile effect whether after incubation with the maximal doses of verapamil and nifedipine or in a calcium-free medium. The inhibitory effects produced by verapamil and nifedipine are only adequately explained by a blockage of the entry of calcium. Other intracellular changes do not seem to play a role in this case.

[An awareness campaign on the diagnosis and prevention of colon and rectal cancer. Planning and evaluation of its results].

Colorectal cancer is very frequent in Argentina. It is the third most frequent cancer, after skin and breast tumor in the city of Rosario. Our population usually ignores the symptoms of colonic cancer and early diagnosis are infrequent. The Societies of Gastroenterology, Oncology and Colonoproctology organizes a public awareness campaign during july, august and september 1985 in Rosario and its surrounding (population 1,300,000) by means of newspaper, radio and specially by television, describing the symptoms were present, as well as insisting of the possibility of prevention by endoscopy diagnosis and removal of polyps. Literature and pamphlets were distributed, and information letters were send to all the physicians in Rosario. An International update meeting on colon and rectal took place. The results of this campaign were evaluated through the Tumor Registry of Rosario. Since september 1984 still february 1986, the monthly incidence of polyps and cancer of the colon and rectum was recorded. The monthly average incidence of polyps was 18.9. Previous to the campaign, the monthly average of diagnosed polyps was 17.5; during the campaign, 27.5; and after it, 13.8. The monthly average of colorectal cancer was 39, before the campaign, 41; during the campaign, 49, and 26 afterwards. These results were statistically studies by the parametric test of Mann and Whitney. During the campaign a significant (p = 0.05) increase of diagnosed polyps and cancers, was found. Our conclusion is that the awareness was successful, but we think that it is necessary to insist with periodic and longer campaigns.

Influence of some inhibitors of arachidonic acid metabolism on oxytocin contractions in the isolated testicular capsule of the rat.

Oxytocin (50-750 nM) contracted the isolated testicular capsule of the rat. Mepacrine, a phospholipase A2 inhibitor (3 X 10(-5) M) and the lipoxygenase inhibitor nordihydroguaiaretic acid (10(-5) M) inhibited this response whereas the cyclooxygenase inhibitor, diclofenac sodium (10(-5) M), and the thromboxane synthetase inhibitor imidazole (10(-5) M) did not. It appears that metabolites of arachidonic acid dependent on lipoxygenase are involved in the contractile response of the rat testicular capsule to oxytocin.

[Effect of tamoxifen on blood calcium and bone structure of normal and ovariectomized rats].

The effect of tamoxifen on calcaemia and framework was studied, both in normal and ovariectomized rats. Tamoxifen caused hypercalcaemia. Only in one of seven animals hypercalcaemia due tamoxifen was accompanied by radiological changes although no structural modification was histologically evident. In ovariectomized rats, tamoxifen dit not produce either hypercalcaemia or radiological changes. These data suggest that the antioestrogenic activity of tamoxifen is involved in the hypercalcaemia it produces.

[Characterization of beta-2 adrenergic receptors in the human uterine artery].

The actions of the hexoprenaline, salbutamol and terbutaline on adrenalin-induced effects in the human artery were studied in vitro. The human uterine artery has a two-part response to adrenalin: The first, rapid (phasic component), and the second, slow and sustained (tonic component). Both beta-2 stimulators relax the tonic component and reduced the amplitude of the phasic component. The depressor effect of the responses to adrenalin is reduced in the presence of butoxamine and intensified in the presence of caffeine. These results suggest the existence on the human uterine artery of beta-2 adrenergic receptors acting as mediators of smooth muscle relaxation, and suggest also that the mechanism of this relaxation is most likely cAMP dependent.

[Different behavior of the rat vas deferens of castrated animals and those treated with cyproterone acetate].

The effects of castration and treatment with the antiandrogen cyproterone acetate (CPA) on the responses of the vas deferens of the rat induced by phenylephrine, KCl and BaCl2 has been studied. Both castration and CPA induced a spontaneous motility in the rat vas deferens. Castration produces a decrease of the response amplitude induced by phenylephrine and KCl and an increase of those induced by BaCl2 in animals killed 30 days after castration. CPA increases the response amplitude induced by phenylephrine and KCl without modifying those induced by BaCl2. These results suggest that the antiandrogen CPA produces modifications qualitatively different from castration.

[Effect of calcium antagonists on the response of the rat vas deferens to noradrenaline and field stimulation].

The effects of two calcium antagonists, Verapamil (VRP) and Methoxyverapamil (D-600) on the contractile responses of the isolated rat vas deferens to norepinephrine (NE) and to field stimulation (FS) were studied. VRP (1 X 10(-6)-2 X 10(-6) M) decreased the responses to NE. It also decreased the amplitude of the FS elicited responses but only when 100 time higher doses were used. Similarly, D-600 only reduced FS responses with equally high doses. In addition, when trains of pulses were used for FS, VRP (1 X 10(-5)-1 X 1(-4) M) caused an initial increase in the responses followed by a decrease. The fact that much higher doses of VRP are necessary to reduce the FS elicited responses than the contraction due to NE, suggests that the neurotransmitter released by FS might not be NE but a different one which seems to be much less dependent on external calcium than NE for producing its action.

[Hormonal influence on rat vas deferens responses induced by field stimulation].

The effect of castration and treatment with testosterone propionate (PT), estradiol benzoate (E2) and ciproterone acetate (CPA) on the responses of the vas deferens in rat has been studied. Castration produces a time-dependent decrease of the response amplitude. PT augments the response amplitude in normal rats and reverses the effect of castration. E2 augments the response amplitude in normal rats without modifying the castration effect. CPA does not change the response of the vas deferens. The results suggest that PT and E2 through possible different mechanisms, facilitate the transmission in rate vas deferens, whereas castration obstructs it.

[Effects of subacute treatment with clomiphene, ethamoxytriphetol (Mer-25), and nafoxidine on blood calcium and bone density in rats].

The authors study the effect of clomiphene, ethamoxytriphetol and nafoxidine on calcemia and bone density of the rat. These antiestrogens induce hypercalcemia, reproducing the bone radiologic changes produced after castration. The different effects obtained in normal and ovariectomized rats are probably due to the small estrogenic activity of antiestrogens.

Effects of oxytocin on the isolated vas deferens of the rat.

1 The effect of oxytocin was studied on the isolated vas deferens of the rat. 2 Oxytocin (25 to 200 mu/ml) reversibly reduced the contractile response induced by noradrenaline, dopamine and acetylcholine. 3 Oxytocin (400 mu/ml) abolished the response to potassium. 4 The contractions evoked in the vas deferens by field stimulation were also reduced by oxytocin (50 to 200 mg/ml). The inhibitory effect of the drug was related to the external calcium concentration. 5 It is suggested that oxytocin depresses the contractile responses in the vas deferens at least partly by reducing the availability of calcium from an extracellular source.
