Effect of denervation and/or reserpine-induced changes on adrenomedullary catecholamines in pigeon: a fluorescence histochemical study.

Splanchnic denervation of the left adrenal gland was made in adult pigeons of both sexes. Denervation of the adrenal medulla alone did not produce any appreciable change in adrenomedullary catecholamine fluorescence in pigeon. Reserpine, at the doses of 0.05 mg, 0.2 mg and 0.8 mg/100 gm body weight was injected in both innervated and denervated pigeons. Reserpine, at all doses, induced catecholamine fluorescence depletion from the innervated pigeon adrenal medulla. Denervation failed to affect reserpine-induced epinephrine depletion while it prevented reserpine-induced norepinephrine depletion only at a low dose of reserpine.

BamH I polymorphism in the Chinese: its potential usefulness in prenatal diagnosis of beta thalassaemia.

The prevalence of the BamH I site 3' to the beta globin gene in Chinese people was determined in 123 normal subjects, 40 patients with heterozygous beta thalassaemia, and 25 patients with homozygous beta thalassaemia. The site was present in 71.1% and absent in 28.9% of the chromosomes carrying normal beta genes. All 25 patients with beta thalassaemia major had the site. This BamH I polymorphism may be used for prenatal diagnosis in about 29% of the pregnancies at risk.

Primary and secondary structure of black beetle virus RNA2, the genomic messenger for BBV coat protein precursor.

The nucleotide sequence of black beetle virion (BBV) RNA2 has been determined. RNA2 is 1399 b long. Its 5' terminus is capped. Its 3' terminus has an unidentified moiety that renders the RNA resistent to polyadenylation and ligation. The first AUG codon at base 23 is followed by an open reading frame for a protein 407 amino acids long, the predicted size of coat protein precursor. A second open reading frame for a putative protein 72 amino acid residues long begins at base 1110. No other large open reading frames exist. The 5' half of the RNA can be folded into a long, imperfect hairpin of high predicted stability. The 3' half of the RNA can fold into a complex set of multiply bifurcated stem and loop regions.

Assessment of the carcinogenic potential of a proposed food coloring additive, laccaic acid, using short-term assays.

Laccaic acid is a red colored natural dye produced by the insect Laccifer lacca or Coccus lacca. It is obtained in large amounts as a by-product of the shellac industry and has been considered for general use as a food coloring agent. Laccaic acid is found to have no mutagenic activity as assessed by two short-term assays: the Salmonella/microsome mutagenicity test, and the phi X fidelity assay. However, laccaic acid did inhibit metabolic cooperation in Chinese hamster V79 cells. These results suggest that laccaic acid should be tested in animals with particular emphasis on in vivo models for tumor promotion.

Effect of erythropoietin on the lipid composition of red blood cell membrane.

Effect of Ep on [14C]acetate incorporation into different lipid fractions of RBC membranes in starved and phenylhydrazine-treated rats was studied. The incorporation was increased into both neutral and phospholipid fractions on Ep treatment to starved or phenylhydrazine-treated rats. A slight decrease in the ratio of neutral lipid to phospholipid was observed under the influence of Ep in starved rats (23%) or in phenylhydrazine-treated rats (36%). Incorporation of radioactivities into different phospholipid fractions of RBC membrane increased on Ep treatment to starved rats, whereas, the relative percentages of these phospholipids (except LPC) remained more or less unchanged under similar conditions. Phenylhydrazine treatment increased the relative percentage of PC and concomitantly decreased the percentage of Sph. Percentage composition of both these two phospholipids showed a tendency to return to their normal levels on administration of Ep to phenylhydrazine-treated rats. Ep decreased the sigma saturated/sigma unsaturated ratio of fatty acids in PE, PS, and PC of RBC membrane in starved rats. On the other hand, no significant change was observed in this ratio of fatty acids in the phospholipids except Sph of RBC membrane in the presence of phenylhydrazine and Ep. In Sph, the ratio went down under similar conditions.

Serum copper and caeruloplasmin activity in experimental malignancies.

Serum copper content and caeruloplasmin activity were measured in 595 Strain A male mice. The mice carried either benzo(a)pyrene induced fibrosarcoma, transplanted sarcoma-180, Dalton's lymphoma, or Ehrlich carcinoma. The serum copper concentration was raised in all the tumours when compared with normal controls. The observed fall in activity of caeruloplasmin during malignancy may be related to defective iron mobilization.

Sub-cellular distribution of copper and caeruloplasmin in chemically induced tumour tissue.

Copper and caeruloplasmin concentrations were determined in the subcellular fraction of tumour tissue from 15 Strain A mice bearing an induced fibrosarcoma. It was observed that copper concentration was higher in the mitochondrial fraction of tumour tissue when compared to that of tissues of similar site from normal controls. However, the caeruloplasmin concentration was lower in the various fractions of tumour tissue when compared to that of normal control tissue.

Fetal loss from umbilical cord abnormalities--a difficult case for prevention.

The summaries of 12 pregnancies in which fetal distress and death were thought to be the result of umbilical cord abnormalities are presented. These included six cases of stricture and torsion of the cord, one case of umbilical vein aneurysm, one case of perivascular haemorrhage near the fetal end of the cord, one case of umbilical vein thrombosis, two cases of true knot of the cord and one case of very short cord. The perinatal mortality was 75% in the whole group (9 out of 12 fetuses), there being eight stillbirths and one neonatal death. Of the 12 fetuses, only three were small for dates at delivery. Nine patients volunteered the history of a decrease in fetal movements, and non-stress cardiotocography (NST) was abnormal in eight patients. It was noted, however, that fetal death tended to occur rapidly after an abnormal test. Observations in our series indicated that antepartum fetal deaths due to umbilical cord abnormalities are still difficult to prevent, as it is often impossible to detect fetal distress in time for appropriate intervention. Prompt action to deliver the baby after an abnormal NST appears to be a necessary step to prevent antepartum deaths due to such causes.

Sequence of the black beetle virus subgenomic RNA and its location in the viral genome.

BBV (black beetle virus) RNA3, the subgenomic messenger RNA for BBV protein B and its double-stranded form (dsRNA3) were purified from cells infected with BBV and were sequenced. RNA3 is 389 bases long. The sequence is homologous to that of the 3'-terminal region of virion RNA1. RNA3 has a very limited homology to virion RNA2. RNA3 is capped at its 5' terminus and has a structural feature at its 3' terminus that renders it inert to the action of the enzymes RNA ligase and poly(A) polymerase. RNA3 has two overlapping reading frames for putative proteins of size 10,768 and 11,633 Da. The positive and negative strands of dsRNA3 are not capped and correspond in length and sequence to RNA3 itself.

Subcellular localization of alkaline phosphatase in Bacillus licheniformis 749/C by immunoelectron microscopy with colloidal gold.

Subcellular distribution of the alkaline phosphatase of Bacillus licheniformis 749/C was determined by an immunoelectron microscopy method. Anti-alkaline phosphatase antibody labeled with 15- to 18-nm colloidal gold particles (gold-immunoglobulin G [IgG] complex) were used for the study. Both the plasma membrane and cytoplasmic material were labeled with the gold-IgG particles. These particles formed clusters in association with the plasma membrane; in contrast, in the cytoplasm the particles were largely dispersed, and only a few clusters were found. The gold-IgG binding was quantitatively estimated by stereological analysis of labeled, frozen thin sections. This estimation of a variety of control samples showed that the labeling was specific for the alkaline phosphatase. Cluster formation of the gold-IgG particles in association with the plasma membrane suggests that existence of specific alkaline phosphatase binding sites (receptors) in the plasma membrane of B. licheniformis 749/C.

Specificity of subcellular distribution of alkaline phosphatase in Bacillus licheniformis 749/C.

The objective of this investigation was to examine the in vivo characteristics of binding sites for alkaline phosphatase in Bacillus licheniformis cell surface. An attempt was made to correlate the results from several experimental approaches, namely (i) cell fractionation; (ii) ultrastructural cytochemistry; (iii) MgCl2 extraction and sodium dodecyl sulphate--polyacrylamide electrophoresis of the extracted material; (iv) labelling with 125I-labelled diazonium salt to determine the subcellular origin of MgCl2-extracted material. Results show that 40% of the alkaline phosphatase was bound to the plasma membrane, 35% to the cell wall, and 15% was free in the cytosol. The enzyme was present as aggregates in a few discrete sites in the membrane, wall, and cytoplasm. The membrane enzyme was associated with the inside surface. A few aggregates were enclosed in single-layered vesicles which appeared to protrude through the cell wall. The material extracted with magnesium salt consisted of 8-10 proteins including alkaline phosphatase. The majority of the proteins extracted by MgCl2 originated from the outside half of the plasma membrane, whereas, only a few, including alkaline phosphatase, came from the inside half of the plasma membrane. All of these proteins may have formed a complex which was removed by MgCl2 extraction. Patch formation in the membrane indicated specific aggregation of intramembrane proteins after MgCl2 treatment.

Controlled study of self-exposure treatment for phobics: preliminary communication.

Patients with phobic disorder (mainly agoraphobics ) of minimum one year duration were treated by self-administered exposure in vivo treatment. Seventy-one patients were randomly assigned to one of three groups: (A) book-instructed, (B) computer-instructed, or (C) therapist-instructed. All three groups improved significantly to a similar extent on various phobic measures at the end of the treatment and maintained their treatment gains at 6-month follow up. Mean clinicians' time spent with each patient was 40 minutes, 4.2 hours and 3.2 hours in group A, B and C respectively. Similar small numbers of patients defaulted from each group.

Ceruloplasmin in human malignancies.

The ceruloplasmin concentration was determined in 145 cancer patients prior to and after treatment with different radiotherapeutic and chemotherapeutic regimes. The ceruloplasmin concentration was observed to be higher in patients with malignancies than in healthy controls. There was a positive correlation of the values with the clinical condition of the patients. The ceruloplasmin concentration was noted to stop increasing and subsequently fall in patients who responded to therapy, and, in contrast, to remain high or become higher in those who did not respond to therapy. The diagnostic and prognostic value of ceruloplasmin determination is discussed.

Abdominal circumference vs abdominal area--which is better?

The efficacies of ultrasonic abdominal circumference (AC) and abdominal area (AA) measurements in the evaluation of intrauterine fetal growth were compared. The intraobserver random and technical errors were found to be significantly higher with AA measurements. The correlation with intrauterine fetal weight was observed to be better with AC as compared with AA measurements, the fetal weights being expressed as either linear regressions or multiple regressions involving the two parameters. Between 32 and 38 weeks, a single AC measurement detects 74 per cent of small-for-date fetuses, while a single AA measurement detects only 58 per cent of such fetuses. The false-positive rates were similar for the two parameters. It is concluded that AA measurements are no more useful than AC measurements in the routine assessment of intrauterine fetal growth.
