A theoretical note on models of interlimb coordination.

This article clarifies some points about models of interlimb coordination. The authors discuss in detail the predictions and extensions of the H. Haken, J.A.S. Kelso, and H. Bunz (1985) model and compare these to a truncated version used by R.C. Schmidt and colleagues. It is demonstrated that procedures commonly used to fit experimental data to model parameters require the fulfillment of certain assumptions. Finally, the authors show how recent experimental findings can be modeled straightforwardly using a system of coupled nonlinear oscillators.

[Postoperative management after cesarean section--infusion therapy and role of intestinal stimulation with parasympathomimetic drugs and dexpanthenon].

In the last decade the incidence of cesarean section has raised worldwide to 15-30% of all deliveries. A standardized postoperative protocol can be applied to most cases and it should be easy to handle and at low costs. In the present randomized, prospective study 76 consecutive patients who hat a cesarean section were investigated. Two different infusion therapy protocols were compared and the role of prophylactic stimulation of peristalsis by parasympathometic drugs and by the pantothenic acid derivative dexpanthenon was evaluated. In group A patient (n = 36) were treated postoperatively with intravenous glucose 5% plus electrolytes and received routinely neostigmine and dexpanthenon, while patients in group B (n = 40) were treated with standardized commercially available solutions without intravenous stimulation of the bowel. Both groups were similar in terms of age, parity, gestational age, past medical history and previous abdominal surgery. The volume of urine and the serum electrolyte levels were similar in both groups postoperatively. The first evacuation of the bowel occurred in most cases on day 3 (A = 69.5%, B = 82.5% of the patients). 50% of the patients in group A complained of abdominal discomfort and pain postoperatively and needed some therapy, while in group B only 22.5% of the cases hat similar complaints. This difference was statistically significant (chi 2 Test: p < 0.02). The present data suggest that prophylactic postoperative stimulation of the peristalsis after cesarean section increases the incidence of abdominal discomfort significantly without influencing the bowel function. Standardized solutions can be used mostly for immediate intravenous fluid replacement and oral intake can be started after 2-6 hours.

[Epidemiology of FeLV and FIV infection in the Federal Republic of Germany].

In a nationwide study 6101 cats were tested for presence of feline leukemia virus (FeLV) antigen and for feline immunodeficiency virus (FIV) antibodies utilizing enzyme linked immunosorbent assay (ELISA). 21.8% of the German cat population may be FeLV- and/or FIV-positive. Of the animals tested, 13.4% were found to be FeLV carriers, while 8.4% showed evidence of FIV infection. Infection with both viruses was identified in 2.1% of the cats tested. Of the animals showing clinical symptoms, nearly one cat in three was found to be carrying either one or both of the viruses. Male cats were more likely to be infected then were females, similarly as were free-roaming cats, compared with confined cats; domestic cats, compared with purebred cats; and cats > 6 years old, compared with younger cats.

Immunoaffinity purification of an oxidase-activating cytosolic complex from bovine neutrophils.

An oxidase activating complex from the cytosol of bovine neutrophils was purified by immunoaffinity using a monoclonal antibody specific for the 67 kDa cytosolic factor of oxidase activation (p67) and assayed for production of superoxide O2- in a cell-free system. The complex comprised not only p67, but also the second cytosolic factor of 47 kDa (p47) in equivalent amounts. The p47-p67 complex showed a good oxidase activating potency when added to neutrophil membranes in the presence of GTP-gamma-S and arachidonic acid. A ras-related small G protein could not be immunodetected in the p47-p67 activating complex, indicating that the GTP required for oxidase activation in the cell free system bound to a protein that was either present in catalytic amounts in the cytosolic complex or present in sufficient amount in the membrane fraction.

Folding and assembly of phage P22 tailspike endorhamnosidase lacking the N-terminal, head-binding domain.

Tryptic digestion of a thermal unfolding intermediate of the phage P22 tailspike endorhamnosidase produces an N-terminally shortened protein fragment comprising amino-acid residues 108-666 [Chen, B.-L. & King, J. (1991) Biochemistry 30, 6260-6269]. In the present work, the 60-kDa C-terminal fragment was purified to homogeneity from the tryptic digest by gel-fitration chromatography. As in the case for the whole tailspike protein (72 kDa), the purified fragment was found to remain stably folded as a highly soluble, SDS-resistant, enzymatically active trimer. However, its unfolding in the presence of guanidinium chloride was accelerated at least 10-fold compared to the complete, native tailspike protein. Shortened tailspike trimers reconstituted spontaneously and with high yield after diluting a solution containing acid-urea-unfolded fragment polypeptides with neutral buffer. Upon recombinant expression of the 60-kDa polypeptide in Escherichia coli, it also assembled efficiently and formed SDS-resistant trimers. The refolding and assembly pathway of the N-terminally shortened tailspike paralleled that of the complete protein with slightly, but significantly, accelerated folding reactions, at both the subunit and the trimer levels. As found for the complete tailspike protein, yields of refolding and assembly of the 60-kDa fragments into SDS-resistant trimers decreased with increasing temperature. The refolding yield of fragments derived from a temperature-sensitive mutant (Gly244-->Arg) tailspike protein was affected in similar fashion as shown for the whole protein. We conclude that the N-terminal domain (residues 1-107) is dispensable for folding and assembly of the P22 tailspike endorhamnosidase both in vitro and in vivo.

A generally applicable ELISA for the detection and quantitation of the cytosolic factors of NADPH-oxidase activation in neutrophils.

An enzyme-linked immunosorbent assay (ELISA) has been developed using polyclonal antibodies raised against two cytosolic proteins of 47 kDa (p47) and 67 kDa (p67) which behave as activation factors for the superoxide-generating NADPH oxidase of neutrophils at the onset of phagocytosis. These two proteins become associated with the NADPH oxidase complex during activation. This immunological technique has been used to follow the purification steps of p47 and p67. It allows the detection of very small amounts of cytosolic factors in a crude neutrophil extract. It is straightforward and much more sensitive (about 1000 times more) than the classical assay based on the use of a cell-free system of oxidase activation and production of superoxide anion. The percentages of p47 and p67 assessed by ELISA with respect to total cytosolic protein were estimated to amount to 0.13 and 0.20%, respectively. The described method has potential applications for the titration of the cytosolic factors of oxidase activation in autosomal recessive forms of chronic granulomatous disease.

Video rating analysis of effect of maprotiline in patients with dementia and depression.

In patients with dementia and mild depression (DSM-III-R 290.21), the effect of low doses of the antidepressant maprotiline (up to 75 mg/d) was examined. The main parameter was a video rating of global impression. The Mini-Mental State Examination (MMS) and the Geriatric Depression Scale (GDS) were applied to evaluate the effect of maprotiline on cognitive and depressive symptoms. The double-blind, placebo-controlled trial was of eight weeks' duration and included 127 patients, randomized in two groups. The antidepressant effect of maprotiline was reflected in the GDS. There was, however, no indication of an effect of maprotiline on cognitive performance. The global impression, evaluated by video rating, gave no indication as to a beneficial effect of the treatment. - The video analysis showed a significant interrater reliability. The discrepancy between the results of the video rating and the GDS is discussed. - The results confirm similar findings of other authors; i.e., that a sedating antidepressant with some anticholinergic effects cannot be expected to improve cognitive functions despite its antidepressant effect. The main interest of this study, however, lies in its methodology (video analysis).

[Results of zidovudine treatment of 9 vertically HIV infected children].

BACKGROUND AND METHODS: Zidovudine has been shown to be an effective antiretroviral drug in the treatment of AIDS, AIDS-related complex and also symptomatic or asymptomatic HIV infection in adults. First studies concerning zidovudine therapy in children are now available. Since 1987 we take care of HIV-infected and -exposed children at our clinic in a multicenter study. In this report we present data of a 12 months treatment period with zidovudine of 9 perinatally infected children. At enrollment the children's age ranged from 5 months to 6.8 years. The drug was given orally as capsules or syrup at a dose of 10-17 mg/kg/d every 6-8 hours. RESULTS: Zidovudine was well tolerated. Only in one case we had to reduce the dosage for a longer period of time because of hematologic abnormalities. There were no other side effects. During the study 2 patients died before the scheduled treatment regimen could be completed. In 5 children we saw a clinical improvement like weight gain, decreased hepato- and/or splenomegaly, decreased lymphadenopathy and improvement in neurologic abnormalities. The CD4+ lymphocyte counts showed a transient increase. The in vitro mitogen induced proliferation also showed a transient improvement during the first months of treatment. The p24-antigen was detected in 7/9 children. No significant changes were observed over time. CONCLUSIONS: Zidovudine can be administered safety to children with HIV infection. It is well tolerated at a dose of 10-17 mg/kg/d given every 6-8 hours. In some children clinical improvement can be observed. The immunologic and virologic improvements are only transient.

Chemical risk assessment and management in Czech and Slovak Republics.

For more than 40 years, former Czechoslovakia has made minimal efforts to protect the environment. As a result, newly formed republics suffer from some of the worst environmental pollution in Europe. In all stages of a complex process of improving chemical safety both countries are facing tremendous problems: lack of efficient and powerful legislative measures, inadequate and sometimes non-reliable data, shortage of sophisticated equipment, virtual absence of basic knowledge and, generally, persisting distrust between public and governmental institutions in health and environmental issues. These factors have made the beginning of new approaches extremely difficult. However, a great effort of many people in both republics together with a substantial assistance of both international organizations and national institutions from developed countries seem to be promising. It could help to move former position of largely ineffective measures and investments to one where limited funds are spent to achieve public benefits.

Comparative histopathological study of rabbit haemorrhagic disease (RHD) and European brown hare syndrome (EBHS).

Histopathological lesions due to rabbit haemorrhagic disease (RHD) and European brown hare syndrome (EBHS) were studied in 35 rabbits and seven hares. Both rabbits (Oryctolagus cuniculus) and hares (Lepus europeus) regularly showed severe necrotizing hepatitis. In RHD, coagulation necrosis, mainly confined to the periphery of the lobules, was consistently found. In EBHS, lytic necrosis affecting the whole lobule was conspicuous, at least in severe cases. Particularly in EBHS, necrotic hepatocytes exhibited a special kind of karyorrhexis. In rabbits with a subacute or subclinical form of disease, early liver cirrhosis was observed. Depletion of lymphocytes was a regular feature in spleens of rabbits and was frequently found in hares. In RHD, disseminated intravascular coagulation (DIC) and haemorrhages in different organs, especially in kidneys and lungs were constant findings. DIC was never seen in EBHS and haemorrhages were an infrequent finding.

Combination of very-low-calorie diet and behavior modification in the treatment of obesity.

Very-low-calorie diets (VLCDs) cause rapid weight loss. However, weight regain is rapid upon discontinuing the VLCD unless lifestyle is altered. The addition of a behavioral-modification (BMOD) program improves the long-term outcome. The major components of a BMOD program to alter lifestyle are education about nutrition and eating habits with alteration of amount and patterns of eating, institution of an aerobic activity program designed to increase energy expenditure, training in self-awareness and assertiveness, and training in coping techniques for long-term alteration of lifestyle. By retrospective chart review, we evaluated two VLCD programs at the same institution. One program gave 12 wk of BMOD before a 12-wk course of VLCD (Program 1), then scheduled individual visits regularly thereafter. The other program used simultaneous VLCD and BMOD in a 26-wk treatment course (Program 2), consisting of 2 wk of stabilization on 5020 kJ/d (1200 kcal/d), 12 wk of VLCD, and 12 wk of transition to a low-calorie solid food diet. Follow-up was obtained at 1 y in both groups. Maximum weight loss was similar, but subjects in Program 2 regained about one-third of the lost weight versus approximately 10% in Program 2. A third program of preceding BMOD followed by a low-calorie diet (Program 3) gave similar weight maintenance at 1 y as Program 1. Prospective studies are needed to determine if BMOD simultaneously with or preceding VLCD promotes better long-term weight maintenance. These data confirm the benefits of long-term follow-up for maintaining weight loss.

Dose-dependent effects of recombinant human erythropoietin on the reticulocyte population of rabbits.

The effects of repeated applications of recombinant human erythropoietin (rhEPO, epoetin beta, Recormon, CAS 122312-54-3) on the reticulocyte count and the reticulocyte maturation distribution of rabbits were investigated using the automated reticulocyte analysis system Sysmex R-1000. Therefore, 150, 300, and 600 IU rhEPO per animal were injected 3 times in intervals of 5-6 weeks. The strongest reaction was found on the second day after application of rhEPO with respect to maturation distribution characterized by the distribution quotient (DQ) and on the fourth day with respect to reticulocyte count. At this time these parameters showed a distinct dose dependence (p < 0.001). The effect of rhEPO on the erythropoietic system in the same rabbit population could be repeated. Thus, immunological neutralization of rhEPO did not take place in rabbits in response to a 3-fold application. The results indicate a potential basis for a simple bioassay in order to determine rhEPO activity in normocythemic rabbits.

In vitro folding pathway of phage P22 tailspike protein.

The intracellular chain folding and association pathway of the thermostable, trimeric phage P22 tailspike endorhamnosidase has been the subject of a previous detailed study employing temperature-sensitive folding mutants. Recently, reconstitution of native tailspikes from completely unfolded polypeptides has been accomplished, providing a model system to compare protein folding pathways in vivo and in vitro. The in vitro reconstitution pathway of the protein after dilution from guanidine hydrochloride or acid-urea solutions at 10 degrees C was characterized by spectroscopic and hydrodynamic techniques, and may be summarized as an ordered sequence of folding, association, and folding reactions. Multiphasic folding of monomers was indicated by changes in circular dichroism and fluorescence, with a rate constant of k = 1.6 X 10(-3) s-1 for the slowest phase observed spectroscopically. Trimerization of structured monomers was followed by size-exclusion HPLC and was completed within 1.5 h at a protein concentration of 20 micrograms/mL. Although at this time trimers did not exchange subunits, they were readily dissociable by dodecyl sulfate in the cold. Formation of native, detergent-resistant trimers was only completed after 3 days of reconstitution at 10 degrees C. The reconstitution pathway of the tailspike protein closely resembles its intracellular maturation path. Thus, the in vitro reconstitution system, as a valid model of chain folding and association in vivo, should provide the tools to localize the steps or intermediates on the pathway that are the targets of temperature-sensitive folding mutations.

Influence of magnetic resonance imaging on evoked potentials and nerve conduction velocities in humans.

The authors investigated the influence of a 1.0-T magnetic field of a magnetic resonance (MR) imaging device, as well as the possible effects of the changing magnetic field and the RF field, on somatosensory, visual, and auditory evoked potentials and on peripheral nerve conduction velocities. The neural potentials outside and inside the static magnetic field were recorded before and after MR imaging. The measured latencies before and after MR imaging were within the normal range for healthy volunteers. The magnetic field did not significantly alter central or peripheral conduction velocities.

[Number and maturation distribution of the reticulocytes from six animal species].

With the fully automated reticulocyte analyzer Sysmex R-1000 (Fa. Toa Medical Electronics Co., LTD) the reticulocyte count and the reticulocyte ratio of rabbit, rat, mouse, mastomys, dog and swine was determined. Simultaneously, the reticulocytes were subdivided into 10 maturation classes according to their fluorescence intensity. The maturation distribution can be characterized by a distribution quotient. Both the maturation distribution as well as the distribution quotient are considered to be useful as good indicators of erythropoietic activity. The investigation establishes, that the reticulocyte count and the maturation distribution of these six species can be determined quickly and easily. The distinct differences between the species reflect the maturation locus of the reticulocytes, which varies among the species (medullary or humoral type of maturation).

Immunoglobulin-related amyloidosis presenting as recurrent isolated lymph node involvement.

Up to 37% of cases of generalized primary and secondary amyloidosis demonstrate lymph node involvement. Lymph node involvement as the presenting feature of generalized amyloidosis is uncommon. Isolated lymph node amyloidosis (that is, with no extranodal amyloidosis) is exceedingly rare; review of the literature reveals only two reported cases. A case of recurrent isolated lymph node amyloidosis is presented, with review of the literature.
