Epoxide-diol metabolic pathway of cytenamide in the rat.

Cytenamide administered intraperitoneally to rats is biotransformed to cytenamide-10,11-epoxide and 10,11-dihydro-10,11-dihydroxycytenamide. These metabolites were separated by chromatographic methods and their structures elucidated by mass spectrometry. The structure of the epoxide was confirmed by direct comparison with an authentic sample. The formation of a chemical artifact, cytenamide-syn-11-hydroxylactone, in the urine was observed. The formation of these metabolites in vitro was demonstrated by incubation of cytenamide with rat liver microsomes.

Epoxide-diol pathway in the metabolism of 5H-dibenzo[b,f]-azepine (iminostilbene).

5H-Dibenzol[b,f]azepine-10,11-epoxide and 10,11-dihydro-10,11-dihydroxy-5H-dibenzo[b,f]azepine were identified by gas chromatography-mass spectrometry in rat urine as the main biotransformation products in the metabolism of 5H-dibenzo[b,f]azepine (iminostilbene). The presence of these metabolites was confirmed in vitro by incubating iminostilbene with rat liver microsomal enzymes.

Biotransformation of trithiozine. III - Isolation and identification of further metabolites in rat urine.

The metabolism of a new antisecretory and antiulcer drug, trithiozine (I.S.F. 2001, T), was studied in 4 hr rat urine samples after i.p. administration. After extraction at pH 7 with chloroform, the urine was either incubated with beta-glucuronidase or acidified to pH 2 and subsequently extracted with chloroform. The organic layers were evaporated to dryness and the residues used for TLC analysis. The neutral extracts revealed five spots, not present in control rat urine, corresponding to the unchanged drug T and to four metabolites. Two of the metabolites had been previously identified as the 4-(3,4,5-trimethoxybenzoyl)tetrahydro-1,4-oxazine (TBO) and the 4-(3,4,5-trimethoxythiobenzoyl)tetrahydro-1,4-oxazine-S-oxide (TO). Three other metabolites were found in the extracts after beta-glucuronidase incubation. TLC, U.V. and M.S. data were consistent with the structure 4-(3,5-dimethoxy-4-hydroxythiobenzoyl)tetrahydro-1,4-oxazine (HT), the corresponding S-oxide (HO) and the 4-(3,5-dimethoxy-4-hydroxybenzoyl)tetrahydro-1,4-oxazine (HBO). The acidic extracts revealed two spots structurally identified as the 3,4,5-trimethoxybenzoic acid (TBA) and the previous HBO. On the basis of present knowledge, a possible metabolic pathway of T is reported, consisting in a rapid metabolic oxidation on the sulfur atom and a slower demethylation on the para methoxy group. The presence of TBA is indicative of subsequent enzymatic hydrolysis of TBO. The intense and long-lasting inhibitory effect of T on gastric secretion is tentatively correlated with the pharmacological activities of some of its metabolites.

Gas chromatographic method for the determination of nomifensine in human plasma.

An analytical method based on solvent extraction, formation of a fluorinated derivative and quantitation by gas-liquid chromatography using an electron capture detector has been developed for the determination of nomifensine in biological fluids. The specificity (controlled by mass spectrometry) and the sensitivity appear to be satisfactory for drug level measurements in human body fluids. Its relative simplicity in fact permits its use in serial analysis.

Activity of liver microsomal mono-oxygenases on some epoxide-forming cyclic tricyclic drugs. I. Kinetics in vitro.

1. The mono-oxygenase activity that forms epoxides has been studied in rat liver microsomes using as substrates carbamazepine and cyclobenzaprine, tricyclic drugs which form stable epoxides in vivo and in vitro. 2. A simple gas chromatographic method has been used to determine the amount of epoxide formed and the linearity of the enzymic reaction with time and protein concentration has been demonstrated. 3. Pre-treatment with carbamazepine increases the rate of formation of carbamazepine epoxide in rat liver microsomal preparations. 4. The effect of SKF 525-A on the formation of these epoxides has been studied.

Gas chromatographic determination of dimethophrine in biological specimens of rabbit.

A method to measure the alpha-sympathomimetic amine, 1-(3,5-dimethoxy-4-hydroxyphenyl)-2-monomethyl-amino-ethanol (dimethophrine, DMP), was developed and applied for the determination of the drug in biological specimens. The gas chromatographic degradation of the drug was investigated, and in order to avoid this process, DMP is converted to the trifluoroacetyl derivative and assayed by gas liquid chromatography on an OV 1' column. The minimum detectable amount is 25 pg per injection. Linearity (using hexachlorobenzene as internal standard for quantitation) was found up to mug amounts of the substance, without any interference with endogenous substrates. Values from plasma, red blood cells and urine in rabbits treated with a single dose of DMP (10 mg/kg) are also reported.
