A human monoclonal IgG1 with anti-idiotypic activity against anti-human thyroglobulin autoantibody.

Sixty-one human myeloma proteins (HMP) from patients with multiple myeloma and Waldenström macroglobulinemia were tested for anti-idiotypic (Id) activity against autoantibodies to double-stranded DNA, small nuclear ribonucleoproteins, and human thyroglobulin (HTg), by competitive radioimmunoassays and enzyme immunoassays. An IgG1, lambda HMP from patient BEN with anti-Id activity against antibodies to HTg is reported. IgG1 BEN was not directed toward human Fc fragments and its activity was not related to allotypic determinants. IgG1 BEN molecules recognized Id determinants (idiotopes) on F(ab')2 anti-HTg fragments, but not idiotopes of F(ab')2 fragments of antibodies of other specificities. This observation supports the general significance of Id network interactions in regulation and diversification of immune responses in man.

Regulation by muramyl dipeptide (MDP) of the lymphoproliferative responses and polyclonal activation of human peripheral blood mononuclear cells.

The ability of muramyl dipeptide (MDP), its adjuvant inactive stereoisomer, MDP(D-D), and the non-pyrogenic, adjuvant active analogue, MDP-butyl ester (MDP-BE), to induce in vitro proliferation and/or polyclonal activation (PA) of peripheral blood mononuclear cells (PBMNC) from normal volunteers, was studied. MDP, as well as its two analogues, were incapable of inducing 3H-thymidine uptake or immunoglobulin synthesis in PBMNC cultures from the majority of the individuals tested. However, these muramyl peptides were capable of regulating the in vitro proliferative responses of some individuals to concanavalin A and to soluble antigens of Candida albicans. At the same time, enhancement of the pokeweed mitogen-induced IgA and IgM but not IgG PA was observed with MDP, its adjuvant active analogue MDP-BE, but not with the adjuvant inactive stereoisomer MDP(D-D). Results are discussed with relation to a possible genetic restriction of the responsiveness to MDP.

Expression of anti-idiotypic clones against auto-anti-DNA antibodies in normal individuals.

A serum pool from 280 blood donors and individual samples from blood donors were assayed for anti-idiotypic activity to auto-anti-DNA antibodies by competitive radioimmunoassays. We found that serum from several normal blood donors inhibited the binding activity of anti-DNA antibodies affinity purified from the sera of patients with systemic lupus erythematosus. This inhibition was due to immunoglobulin molecules but was not due to rheumatoid factor activity. Antiallotypic antibodies were not responsible for the anti-anti-DNA activity detected. Because this inhibition was blocked by DNA molecules, the observed reactivity was probably caused by idiotype-anti-idiotype interactions. These results provide evidence that anti-idiotype antibodies against anti-DNA autoantibodies are present in certain normal human sera. Anti-anti-DNA antibodies could play a role in the regulation of autoimmunity to DNA.

Idiotypic and anti-idiotypic determinants on lymphocytes during anti-Rh immunization.

Evolution of idiotypic determinants on lymphocytes membrane and presence of other lymphocytes carrying anti-idiotypic determinants, were studied in Rh negative human volunteer blood donors during immunization towards Rh factor. For this purpose E-Rh Rosettes, direct immunofluorescence, inhibition of E-Rh Rosettes by anti-idiotypic sera as well as EA-Rh Rosettes--induced with Fab'2 fragments from anti-Rh antibodies and lymphocytes from the same subject--were examined and compared to the evolution of circulating antibodies. E-Rh Rosettes preceded or accompanied production of anti-Rh antibodies; their frequency decreased after the fifth month following immunization, meanwhile EA-Rh Rosettes increased in parallel with antibody decrease. The direct immunofluorescence and the inhibition of E-Rh Rosettes, by anti-idiotypic sera, show the presence of idiotypic determinants on lymphocyte membranes; the presence of EA-Rh Rosettes, coinciding with the decrease in antibodies demonstrate the existence of lymphocytes bearing auto-anti-idiotypic determinants.

[Demonstration of an abnormal increase of anti-T hemagglutinin titers in malaria infected patients].

An increased frequency of high titers of antibodies against A, B, ORh+ trypsinized, and ORh+ neuraminidase-treated human red blood cells was observed in the sera of Africans living in a malaria endemic area, and Europeans with primary malaria attacks. In the latter group, titers of agglutinins against neuraminidase treated RBC were increased in 84% of individuals and were of the IgM class. Inhibition experiments with beta-D-galactose and lactose showed that this agglutinin was directed to the T antigen of human RBC. Its possible implication in malaria related anemia is discussed.

Cytotoxicity reactions against target cells infected with rabies virus.

Some aspects of the cytotoxicity reactions were studied in the rabies system. The antibody-dependent complement cytotoxicity (ADC), the cellular cytotoxicity (CC), and the antibody-dependent cellular cytotoxicity (ADCC) are shown, being the cytotoxic effect as evidenced by the 51Cr released from the cells infected with the Pasteur strain of rabies virus. Some parameters such as time of cellular infection, the amount of infected cells, the concentration of complement, and the incubation time of the ADC reaction, which help to increase the performance of this reaction, are discussed. The detection and the level of the cellular response against the Pasteur strain of rabies virus in immunized mice is shown. Evidence is presented that in the ADCC test, specific human antibodies and non-immune human lymphoid cells are able to mediate in vitro lysis of cells infected with rabies virus. A comparison of the ADCC test with serum neutralization and immunoenzymatic tests is shown.

High titres of anti-T antibodies and other haemagglutinins in human malaria.

The prevalence of antibodies against (i) human red blood cells (RBC) of A and B groups, (ii) trypsinized O Rh+ RBC and (iii) neuraminidase treated O Rh+ RBC were investigated both in sera of Africans from a malaria endemic area of Upper Volta and in sera of Europeans with acute malaria from a Paris hospital. An increased frequency of high titres of haemagglutinins was observed against A and B as well as O Rh+ trypsinized human RBC, thus confirming previously published results. In addition, agglutination of neuraminidase treated RBC showed that the titres were increased in about 40% of Africans studied and in about 80% of patients with acute malaria. Using agglutination with a specific anti-T lectin and inhibition with two ligands, it was found that sera of malarious patients contain high titres of antibodies directed against the T antigen of neuraminidase treated RBC. The mechanisms of appearance of high titres of autohaemagglutinins in malaria and their possible interference in the anaemia associated with this disease are discussed.

[Immunocytochemical detection in Stannius corpuscles of the eel (Anguilla anguilla L.) of a hormone similar to the mammalian parathyroid hormone].

In eels the parathyrin of the corpuscles of Stannius (PCS), mammalian parathyroid-like hormone, has been localized in the cytoplasm of all the cells in the corpuscles. This detection was done by indirect immunofluorescence with an antiserum anti 1-84 bovine parathormone. The specificity of the reaction was demonstrated by inhibition of the coloration obtained with the 1-84 bovine parathormone and the active fragment 1-34 of human parathormone. Variations of the cellular localization of the PCS or a complete depletion of the hormonal content were observed in eels made hypercalcaemic by calcium overloading.

[Systemic lupus erythematosus and anti-native desoxyribonucleic acid antibodies: clinical and laboratory comparison of the results given by two methods of detection (author's transl)].

Simultaneous application of two methods for the detection of anti-native DNA antibodies (indirect immunofluorescence on DNA fibres and Farr's radioimmunological method with native DNA labelled C14) to 53 serum samples from 32 patients suffering from systemic lupus erythematosus (SLE) indicated a certain number of discordances between the results obtained. Correlations between clinical and laboratory findings were found, such that certains types of anti-native DNA antibodies (n) may be specific to certain clinical manifestations of SLE: a good correlation was seen between She fixation of anti-C14 nDNA and the titre of IF anti-nDNA in the group of SLE with renal involvement, but was not found in the group of SLE with central nervous system involvement. There was no parallel between the course of renal involvement and anti-nDNA antibody titres, without treatment being directly responsible. The significance of the presence or absence of anti-DNA antibodies with the two methods of detection used is not of any single significance. Several types of anti-nDNA antibodies are present in a given specimen of lupus serum. Certain anti-nDNA antibodies are not detected or are at the limit of a positive result in the presence of neurological manifestations. From a practical standpoint, the use of several laboratory tests is desirable for the possible investigation of SLE.

Study of immunoglobulins in pleura and pleural effusions.

The protein concentration of 35 pleural effusions was compared with that in the serum. The ratio of the pleural and serum concentration of albumin, IgG, IgA, and IgM is always below unity and appears to have no diagnostic value. However, the ratio of the concentration of these proteins was inversely related to their molecular weight. The underlying mechanism in malignant and inflammatory effusions appear similar and is in keeping with a diffusion process. Immunofluorescent staining of the pleura suggests the intercellular passage of the proteins through the mesothelial barrier.

[Evolution of anti-Rh antibodies idiotypy in a blood donor over 9 years].

Anti-idiotypic sera were obtained in Rabbits immunized with Rh antibodies (isolated from the serum sample collected in 1974 from a Blood Donor). The sera agglutinate, at high titers, red cells coated with the immunizing antibodies and, at different titers, cells coated with antibodies from serum samples taken at other periods. Inhibition of hemagglutination of anti-idiotypic sera by different samples from the same Donor was complete with the immunizing serum and partial with other samples. These results show that idiotypes or idiotypic specificities appeared or disappeared during the period studied and represent the first observation on evolution of antibody idiotypy in Man.

Thyroid autoantibody levels during lithium therapy.

The study of the levels of thyroid antibodies was conducted in 58 patients treated with lithium and in 40 subjects who received other psychotropic drugs and who were used as controls. The measure of the antithyroglobulin antibodies were made by passive hemagglutination and that of the antimicrosomal antibodies by indirect immunofluorescence. For the whole control group, the research of the antithyroglobulin antibodies was positive in 3 cases, i.e. 7.5%. For the 58 patients treated with lithium, this research was positive in 11 subjects, i.e. 19%: thus, lithium seems to cause immunological reactions in the thyroid. 5 lithium-treated patients developed a goiter. The antibody determination was positive in only 2 subjects and was negative in the others. The measure of antithyroid antibodies before lithium did not enable us to predict the emergence of thyroid complications. On the contrary, subjects treated with lithium presented signficantly high antibody levels without developing clinical thyroid manifestations.

Implications of heterophile antigens in immunological infertility in males.

Immunological study was conducted on serum and hydrocel fluids from males belonging to an equatorial African population heavily contaiminated by Filaria and Schistosoma. Anti-spermatozoa antibodies were detected by agglutination in 31 of 64 sera and 24 of 60 fluids, and by cytotoxicity in 19 of 64 sera and 8 of 60 fluids. Antinuclear antibodies were detected in 21 of 64 sera and 8 of 60 fluids. However, the titers of these antibodies were low, as were the frequency and titers of other tissues antibodies. The appearance of antispermatozoa antibodies is also correlated with extensive skin burns and infection by Pseudomonas aeruginosa or severe Candidiasis.

[Evaluation of cellular immunity after rabies vaccination in man].

The peripheral lymphocytes of vaccinated men were stimulated by homologous antigen or its subunits (nucleocapsid and glycoprotein) or by some parent viruses (Mokola and Lagos). All subjects produced a high level of antibody. A difference was noted between the two types of immunity. A lack of cell-mediated immunity could explain the very few failures in Pasteur antirabies treatment.

[Calcitonin in the ultimobrancial body of Anguilla (Anguilla anguilla L.): cytologic localization by indirect immunofluorescence using human anti-salmon-calcitonin antibodies].

The localization of intracellular calcitonin has been achieved by immunofluorescence in the cytoplasm of all cells forming the epithelium of the ultimobranchial body of eels, using a human antiserum against synthetic Salmon calcitonin I. The specificity of the reaction is demonstrated by inhibition with synthetic salmon calcitonin (S.C.T.); the fluorescence is not inhibited by synthetic human calcitonin (H.C.T.).
