Formation of ATP-insensitive weakly-binding crossbridges in single rabbit psoas fibers by treatment with phenylmaleimide or para-phenylenedimaleimide.

Chaen et al. (1986. J. Biol. Chem. 261:13632-13636) showed that treatment of relaxed single muscle fibers with para-phenylenedimaleimide (pPDM) results in inhibition of a fiber's ability to generate active force and a diminished ATPase activity. They postulated that the inhibition of force production was due to pPDM's ability to prevent crossbridges from participating in the normal ATP hydrolysis cycle. We find that the crossbridges produced by pPDM treatment of relaxed muscle cannot bind strongly to the actin filaments in rigor, but do bind weakly to the actin filaments in the presence and also absence of ATP. After pPDM treatment, fiber stiffness, as measured using ramp stretches of varying duration, is ATP-insensitive and identical to that of untreated relaxed fibers (both at high [165 mM] and low [40 mM] ionic strength). These results suggest that the pPDM-treated crossbridges, in both the presence and absence of ATP, are locked in a state that resembles the weakly-binding myosin ATP state of normal crossbridges. Their resemblance to the ATP-crossbridges of relaxed untreated fibers is quite strong; both bind to actin about equally tightly and have similar attachment and detachment rate constants. We also found that crossbridges are locked in a weakly-binding state after treatment with N-phenylmaleimide (NPM). In muscle fibers, this method of producing weakly-binding crossbridges appears preferable to pPDM treatment because, unlike treatment with pPDM, it does not increase the fiber's resting tension and stiffness and it does not disrupt the titin band seen on SDS-PAGE.

[Pyogenic cervical osteomyelitis following pneumonia].

A 65-year-old man was admitted because of severe cervical pain radiating to both shoulders and bouts of fever, which followed pneumonia. Bone scan showed increased focal uptake in C6-7, while computerized tomography showed destruction of the 7th intervertebral disk, with bone sequesters and soft tissue swelling. Fine needle aspiration drew purulent material with gram-positive bacteria on direct staining. Culture grew Staphylococcus aureus, coagulase positive. Treatment with IV cloxacillin for 3 weeks, followed by oral treatment for another 3, resulted in complete remission.

Carcinoma of the colon in asbestos-exposed workers: analysis of asbestos content in colon tissue.

Epidemiological studies have indicated an increased incidence of carcinoma of the colon in asbestos workers. The present study evaluated the colon tissue asbestos burden, by light and electron microscopic analytic techniques, in patients with a history of occupational asbestos exposure and colon cancer. Asbestos fibers and/or asbestos bodies were present in colon tissue from 14 of 44 (31.8%) asbestos workers with colon carcinoma (range 142,199 to 15,231, 543 fibers/g/wet weight, mean 2,517,823). Chrysotile was identified in 9 patients and amosite in 3 patients. Both amosite and chrysotile were found in the colonic wall in one individual. Other forms of asbestos (e.g., crocidolite, tremolite, or anthophyllite) were not found. Asbestos fibers and asbestos bodies were not found in colon tissue from 20 control patients (colon carcinoma and no asbestos exposure). Asbestos fibers frequently enter and reside in the wall of the colon and are often intimately associated with tumor tissue at the site of colon carcinoma in workers with asbestos exposure and colon carcinoma.

A comparison of personality characteristics, family relationships, and drug-taking behavior in low and high socioeconomic status adolescents who are drug abusers.

Low socioeconomic status (SES) and high SES adolescents who met the DSM-III criteria for drug abuse were compared with respect to personality characteristics, family relationships, and drug-taking behavior. SES was determined by parents' income, education, and occupation. The two groups did not differ significantly with respect to self-esteem, quality of family relationships, impulse control, moral development, and drug-taking behavior. However, high SES adolescents were more depressed (p less than .009) and more anxious (p less than .02) than low SES adolescents, and these findings lent support to the argument that low and high SES drug abusers differ in personality and therefore require different kinds of treatment interventions.

A rapid and simple method of extracting asbestos bodies from lung tissue by cytocentrifugation.

During analyses of alkali digested lung tissue for asbestos bodies, we observed that the number of asbestos bodies in the discarded waste frequently exceeded the number in the filtered residue, the number reported in the standard diagnostic method. This observation led to the exploration of alternative techniques that would optimize the recovery of asbestos bodies. We describe a new, simple, and rapid method for extracting asbestos bodies from digested lung tissue using a cytocentrifuge, in which the waste extraction and filtration steps are eliminated. Samples of digested lung tissue are ready for light microscopy after 10 minutes of cytocentrifugation directly onto a glass slide. The standard method was compared with the cytocentrifuge technique using lung tissue from four asbestos-exposed workers and four controls with no known history of exposure. The number of asbestos bodies extracted by the centrifuge method was, on average, seven times higher than the number found with the standard method. A detailed quantitative study was made of the case that had the most asbestos bodies (comparison of the number of asbestos bodies counted in both "residue" and "waste", applying the filtration and cytocentrifuge methods). The number of asbestos bodies found in the discarded waste significantly exceeded the number in the "reportable" filtered residue.

Purification and characterization of chymodenin. A hormone-like peptide from porcine duodenum.

Chymodenin, a hormone-like duodenal peptide which rapidly alters the proportions of secreted pancreatic digestive enzymes to a mixture relatively richer in chymotrypsinogen than that found in basal secretion, has been purified to homogeneity. The starting material was an acidic methanol-soluble, neutral pH-insoluble fraction of an acetic acid extract of porcine duodenum; the purification consisted of cation-exchange chromatography on SP-Sephadex and CM-Sephadex in ammonium bicarbonate gradients, and gel filtration on Sephadex G-75 in dilute acetic acid. The yield of material was followed by radioimmunoassay. Homogeneity was determined from chymodenin's behavior in disc gel electrophoresis in an acidic counter-migration-of-dye system, sodium dodecyl sulfate-urea gels, gel filtration, dansyl-Edman reaction, reversed-phase high pressure liquid chromatography, isotachophoresis, and sedimentation equilibrium ultracentrifugation. The electrophoretic mobility, the molecular weight of 9,000-10,000, and the biological activity differed from those of other gastrointestinal peptide hormones. The amino acid composition was unique. Chymodenin is the first purified hormone-like substance reported capable of altering the composition of the mixture of secreted digestive enzymes, independent of the stimulation of massive pancreatic protein output.

A serum inhibitor of neutrophil chemotaxis associated with hyperglobulinaemia E in a patient with lymphoma, and recurrent skin infection.

A patient with a strong serum inhibitor of neutrophil chemotaxis, a possible chemotactic factor inactivator and hyperimmunoglobulinaemia E is described. The patient had a persistent chronic dermatitis which appeared to develop following radiation of the central nervous system for an unclassified lymphoma. Other abnormalities included a general increase of serum immunoglobulins, impaired lymphocyte response to mitogens, and a high Epstein-Barr antibody titre. This case illustrates that an increased IgE level, previously reported in association with a cellular chemotactic defect, may coexist with serum inhibitors of neutrophil chemotaxis.

Influence of thymic hormones on mitogen-induced interferon production in lymphocytes. II. Enhancement of Con-A-induced immune interferon production by a synthetic thymopoietin pentapeptide.

A synthetic thymic pentapeptide was used for prestimulation of mouse lymphocytes in vivo. This pentapeptide (DAc- Glu3 -TP5) affected the cAMP and cGMP levels positively and decreased the 3H-thymidine incorporation in lymphocyte cultures significantly. In combination with Con A DAc- Glu3 -TP5 influenced the production of interferon. After in vivo pretreatment of adult hybrid mice with DAc- Glu3 -TP5 the mitogen-induced interferon production was 3 to 7 times higher than in controls. The interferon produced was immune interferon (IFN gamma) as determined by pH 2-sensitivity and lack of neutralization by mouse IFN beta-antibodies.
